[Detection of Clostridium perfringens toxins on cell cultures].

An important prerequisite for the possible application of cell culture systems as an in vitro method for the potency test of C. perfringens vaccines is the detection of cytotoxic effects of non-neutralized toxins on cell cultures. For this purpose eight cell lines were investigated in terms of the sensitivity to toxins using microscopy and the MTT assay. The results suggest that the MDCK cell line is a sensitive and specific detection system for epsilon-toxin. The VERO cell line is a suitable indicator for the detection of beta-toxin. Another task was the selection of a sensitive method for the analysis of cytotoxic effects of the toxins used. The neutral red and the tetrazolium-(MTT)-assays were compared for three cell lines (MDCK, FBTR, MA 104). The MTT assay was proved to be the optimal test system under our conditions.

Epidemiological data on Werdnig-Hoffmann disease in Germany (West-Thüringen).

This study contains the largest body of epidemiological data on Werdnig-Hoffmann disease (acute infantile spinal muscular atrophy; ASMA) in West-Thüringen in Germany. The incidence of ASMA was calculated to be 1 in 10,202 live births. The prevalence was 1 in 595,362 of the general population (as of 31 December 1987). The study gives an unexpectedly high incidence rate confirming the suggestion that ASMA in Central and Eastern Europe might be more frequent than in Western Europe. However, we consider that this high incidence rate in West-Thüringen is a result of the almost complete ascertainment made possible because of the well-organised and centralised health system existing in Thüringen over the last few decades.

[Campylobacter as the cause of diarrhea in calves].

The modified Preston medium allows the isolation of C. jejuni, C. coli and C. fetus subsp. fetus from intestinal samples of calves at an incubation temperature of 37 degrees C. In the first series of investigation, Campylobacter excretion in calves (n = 7) was followed up to the age of 4 months. In the first 4 days of life, these bacteria could not be detected in any of the animals. Thereafter first C. coli we found in all calves. In 4 animals, only strains of this species were isolated during the whole investigation period. In 3 animals C. fetus subsp. fetus could be detected repeatedly, however C. coli and sometimes C. jejuni were found, too. In the second series of investigation, isolation of Campylobacter from different parts of the gastrointestinal tract or organs was successful in 19 out of 25 diarrhoeal, moribund calves. 16 out of 19 positive animals harboured large amounts of these gramnegative bacteria in the distal jejunum and ileum. In 10 animals out of these 16, the germ colonized also the proximal jejunum and abomasum. From 6 calves, C. fetus subsp. fetus was isolated, and C. jejuni from 7 calves. C. coli was relatively rare. From the lymph nodes of the proximal and distal jejunum, Campylobacter (exclusively C. jejuni) were isolated from 5 animals. Due to the Campylobacter presence in the small intestine of diarrhoeal calves, a contribution of this bacteria within the pathogenesis of calf diarrhoea is possible. Final evaluation of their pathogenesis importance is only positive by means of virulence tests.

[Histological studies of the occurrence of organisms shaped like Campylobacter in the abomasum of calves].

The mucosa of the abomasum is a strongly acidic and therefore germ poor, but not entirely germfree environment. Only in isolated areas of the fundic glands zone were straight rods in the foveolas, and even more seldom spiral shaped bacteria could be encountered. They had no relation to the inflammatory infiltrates. On the other hand, Campylobacter-like organisms were more often observed in the pyloric region and, preferably, in a narrow zone following the cutaneous mucosa of the omasum. These organisms appeared to have a different size (1.3 to 2.4 microns length and 0.4 to 0.8 microns width) and they occurred deep inside the glands in an almost pure fashion. Whereas they caused no visible reaction of the tissue in the narrow cardiac zone, their occurrence in the pyloric region was several times connected with neutrophilic-rich infiltration. It ought to be tested, whether Helicobacter are among these Campylobacter-like organisms, which cause a disease leading up to ulcerations in calves similar to gastritis B in humans.

[Iris reconstruction: surgery, laser or contact lenses with iris structure].

We report the results of 126 iris reconstructions from 1979 to 1989 at the Rostock University Eye Clinic: 68 were operative, 34 laser treatments, and 24 Hema contact lenses with an iris structure. We developed four variations of the iridopexy: (1) an iridopexy with an iris micronhook; (2) an operation with an iris catching device; (3) the procedure using anterior chamber forceps according to Barraquer; (4) the suction cannula technique. These four variations permit damage to the epithelium to be avoided--unlike the McCannel or Makkensen techniques. Four fixation of the iris, in phakic eyes two pupil punches were patented. For aphakic eyes, we prefer to fix the iris via a combination of argon and neodym-YAG laser surgery. To date, the most important indications for fitting the Hema contact less with an iris structure have been aniridia, albinism, iris coloboma, essential iris atrophy, and leucoma corneae.

[Lysozyme in calves].

Relatively high lysozyme concentrations, depending on age, were recorded from intestinal content, spleen, liver, kidney, lung, lymph nodes, and mucosa of calves. Yet, only minor quantities of lysozyme were found in blood serum or plasma and in granulocytes. Physicochemical characterisation, precipitation, using polyclonal antisera, and crosswise neutralisation of lysis reaction were likely to suggest occurrence of immunological relationship reactions as well as differences between organ lysozymes, on the one hand, and intestinal lysozymes, on the other.

Tumorigenesis and eye abnormalities in transgenic mice expressing MSV-SV40 large T-antigen.

Transgenic mice which expressed SV40 large T-antigen under the control of the MSV enhancer and the SV40 promoter were generated. In animals containing an intact MSV enhancer, total lens cataracts and neuroectodermal brain tumors, originating in the pineal organ were observed. In contrast, 5' deletion of the MSV enhancer to a residual 53 bp resulted in a different spectrum of pathologies. Whilst lens cataracts still occurred, no brain tumors could be detected. Instead, fibrosarcomas and adenocarcinomas of the kidneys were induced. In addition, tumors of the endocrine pancreas were observed with both transgene constructs. We conclude that the MSV enhancer element is sufficient to direct the expression of the viral reporter gene to the lens and the pineal organ in transgenic mice. Deletion of the MSV enhancer correlates with the loss of DNA elements responsible for the pineal cell specific expression of SV40 large T-antigen.

[Carl Wilhelm von Zehender (1819-1916)--authentic data from his life].

The life and work of Carl Wilhelm von Zehender are discussed in the light of recent information. As a pupil and friend of Albrecht von Graefe, he took part in the foundation of the Heidelberg Ophthalmological Society, which was, right from its start, an international association. He was the first editor of "Klinische Monatsblätter für Augenheilkunde" (1863). In 1862, he was nominated professor of ophthalmology and otology at Berne, Switzerland, but already in 1866, he accepted a call to the newly created chair of ophthalmology at Rostock, Mecklenburg. There, he initiated ophthalmic microsurgery and--in the field of social medicine--scientific expert judgment of damage to the eyes. Hirschberg's rather depreciating criticism of von Zehender appears no longer valid.

[Damage to the corneal endothelium following exposure to sulfur hexafluoride gas].

Damage to the corneal endothelium caused by sulfur hexafluoride was analyzed in 14 rabbits. Fourteen eyes were used for testing, while the other 14 served as controls. Three stages of endothelial reaction resulting from long-term contact with sulfur hexafluoride were established by morphologic and morphometric analyses. The first stage - acute necrosis of the endothelial cells - covers all stages of destruction up to total obliteration in the exposed area of the endothelium, which is clearly demarcated from the rest of the endothelium. The second, repair stage, begins as early as 24 hours after the operation and includes the diverse forms of endothelial cell spread, transformations of the hexagonal endothelial cells, and endothelial cell migration with polymorphia and formation of rosettes. The third, proliferative phase of the endothelial cell reaction follows directly after the second phase and is characterized by the production of more giant cells, the production of giant rosettes, compaction of cell membranes and filling of the intercellular space with amorphous substances. The results of this research are of major practical relevance, since they show that a 40/60% sulfur hexafluoride/air mixture cannot be regarded as inert.

[Follow-up study of auditory evoked brain stem potentials--a contribution to the differential diagnosis of facial paralysis].

The case report of a 5-year-old girl suffering from an acute peripheral facial paralysis is used to discuss the clinical picture of Bell's paralysis. In the present case the sign was due to a brain-stem encephalitis, the diagnosis being based on brain-stem acoustically evoked potentials. Although not significantly influencing further practical procedure the authors consider it worth mentioning that apparently banal "peripheral" facial paralyses can actually be part of complex CNS lesions.

[Effect of metronidazole on the gastrointestinal flora of swine].

In veterinary medicine since the beginning of the seventieth several derivatives of imidazole are mainly and successfully used for therapy of pig dysentery. Daily metronidazole doses of 25-100 mg/kg body mass resulted in no provable effect on the normal quantitative composition of the germ flora of healthy porkers. Even in such high doses of 100 mg/kg body mass per day that substance revealed well tolerant. A singular occurrence of colienterotoxemia was disconnected with the metronidazole application.

Primary structure and nuclear localization of a murine homeodomain protein.

The murine homeobox Hox 1.1 (m6) is the first of a cluster of six boxes on chromosome 6. Using probes and synthetic peptides derived from the Hox 1.1 sequence, we were able to isolate cDNAs and antibodies that allowed us to characterize the product of this homeobox-containing gene. From the open reading frame on the cDNA clone B21, a protein could be predicted, made up of 229 amino acids and having a calculated molecular weight of 25,740. A unique feature of this protein is that it has 15 glutamic acid residues as its carboxyl terminus, which gives it a very hydrophilic and acidic carboxyl terminal structure, most probably folding onto an alpha-helix. A second domain of six amino acids is present on the Hox 1.1 protein, which is conserved in other homeodomain proteins. Antibodies generated against synthetic peptides from the homeobox region were used in the immunoblotting procedure and revealed a major protein band of Mr 31,000 in extracts from 3T3 cells and F9 teratocarcinoma cells induced by retinoic acid and cAMP. The nuclear location of the protein was established by immunofluorescence. The presence of this protein in F9 cell nuclei is in faithful accordance with the kinetics established for the 2.4-kilobase Hox 1.1 transcript during differentiation into parietal endoderm cells.

The murine homeo box gene product, Hox 1.1 protein, is growth-controlled and associated with chromatin.

In order to gain insight into the function of the Hox 1.1 gene, we studied the expression of the murine homeo box gene product, the Hox 1.1 protein. Monoclonal antibodies were raised against synthetic peptides of the Hox 1.1 protein to study the localization and expression pattern of this protein under various culture conditions. By means of indirect immunofluorescence we localized the Hox 1.1 protein to the nucleus in differentiated F9 and NIH 3T3 cells. During mitosis the protein was found to be associated with chromatin. Confluent NIH 3T3 cells harbored little if any Hox 1.1 protein. After "wounding" the cells in this confluent monolayer, we observed an induction of the expression of the Hox 1.1 protein. However, addition of insulin to F9 and contact-inhibited NIH 3T3 cells led to an increase of the Hox 1.1 RNA and protein expression. Thus, the induction of the Hox 1.1 protein is associated not only with the differentiation of embryonal carcinoma (EC) cells, but may also correlate with stages of cell growth.
