[Application of CH50 assay to the detection of circulating immune complexes (author's transl)].

Detection and titration of circulating immune complexes appear as an essential tool for the diagnosis and prognostic evaluation of immune complex diseases. Authors propose a simple, rapid, senstive and quantitative method based on the evaluation of complement by soluble immune complexes measured by 50% hemolytic activity. This technique was compared to the I125 C1q binding test and may be recommended for clinical laboratory use.

[Value of the detection and characterization of circulating immune complexes in rheumatoid arthritis].

The authors analyse the value of research into and identification of immune complexes (IC) in chronic inflammatory rheumatisms (CIR) and in rheumatoid polyarthritis (RP) in 93 patients hospitalise in a Rheumatology Department. Comparative levels of ICs were established simple and quick technique of measuring the 50% haemolytic complement (HC 50) used up by the ICs. The rheumatoid factor (RF) is identified at the PEG IC precipitates with the Waaler-Rose test. The existence of raised levels of IC containing RF in both seropositive and seronegative PRs and to a similar degree in a non-negligable percentage of clinically non-classable RICs conforms the diagnostic value of this research. On the other hand there is no relationship between the levels of ICs and either clinical signs or age of the illness. In RPs other than CIR circulating ICs are found whose nature is still unknown.

Circulating immune complexes in schistosomiasis.

Circulating immune complexes (CIC) were investigated by the [125I]Clq binding test, the complement fixation test (CFT) and optical density measurement after redissolving 3% polyethylene glycol precipitates of serum from patients infected by Schistosoma mansoni. A highly significant correlation was obtained among these three techniques. More than 60% of the patients demonstrated significantly higher values than control individuals. The level of CIC was found to be higher in the mild than in the hepatosplenic form of the disease. Parasite antigen, IgG, IgM and IgE were characterized in these CIC. In experimental schistosomiasis in mice, maximum levels of CIC, evaluated by the CFT, were observed between the 40th and the 70th day of infection.

The use of the radioimmunoprecipitation-PEG assay (RIPEGA) to detect anti-IgG antibodies.

A new procedure, the radioimmunoprecipitation--P.E.G. assay (RIPEGA) is proposed for the detection of anti-IgG antibodies (anti-IgG Ab). This sensitive and reproducible test was performed by incubation of radio-labelled human IgG (125I-IgG) with patient's sera. Separation of free from complexed 125I-IgG was achieved by a 7% P.E.G. precipitation. Preliminary results demonstrated anti-IgG Ab in 84.4% of the cases of sero-positive rheumatoid arthritis (R.A.), 85% of seronegative R.A., 43% of other forms of inflammatory arthritis, 50% of unclassified arthritis and in 10.5% of the control subjects.

Detection and characterization of circulating immune complexes in rheumatoid arthritis.

Circulating immune complexes (IC) were detected and quantified in serum of patients with rheumatoid arthritis (RA) by three methods: radiolabeled Clq binding test, anticomplementary activity and optical density of 3% PEG precipitated sera. Rheumatoid factor (RF) is characterized in PEG precipitates by the Waaler-Rose reaction. The relationship between IC, complement and RF levels are analyzed.

Evaluation of spleen cell population and effect of splenectomy on granuloma modulation in BALB/c mice infected with Schistosoma mansoni.

A kinetic study of the cells present in the spleen of BALB/c mice infected with Schistosoma mansoni was carried out. The lymphocytes were evaluated phenotypically with monoclonal antibodies and the effect of splenectomy on the modulation of periovular granuloma was also investigated. The infected mice had proportional increases in the numbers of neutrophils, plasma cells, macrophages and eosinophils in the spleen. The largest number of neutrophil, plasma cells and macrophage were found between the 8th and the 12th week of infection, while the amount of eosinophils were higher later on, around the 20th week. The lymphocytes phenotypically characterized as Thy 1.2, Lyt 1.2 (CD4) increased mildly in proportional numbers. However, the percentage of lymphocytes with the Lyt 2.2 (CD8) phenotype, which is characteristic of suppressor and cytotoxic T cells, increased significantly with the progress of the disease. The numbers of B lymphocytes, which comprise 50% of the mononuclear cells present in the spleen, increased significantly till the 16th week when they began to decrease. The mean diameters of periovular granulomas were comparatively similar in both experimental groups (splenectomized and non-splenectomized mice). However, the evolutive types of granuloma (exudative, intermediate and fibrous) in splenectomized mice were proportionally different from those of non splenectomized mice in the 16th and 24th week of infection. It is inferred that lymphnodes or other secondary lymphoid organs, in the absence of the spleen, assume a modulating action on periovular granulomas, although the evolution of the granulomas is somehow delayed in splenectomized mice.

The subclinical form of experimental visceral leishmaniasis in dogs.

Pathological aspects of a subclinical form of experimental canine leishmaniasis is reported here for the first time. Fifteen mongrel dogs were used in the present study. Eight dogs were infected and seven were used as control. Four of the control dogs were inoculated with spleen cells from non-infected hamsters. The eight mongrel dogs inoculated intravenously with amastigotes forms of Leishmania chagasi evolved for periods as long as 25 months without any clinical characteristic sign of classical Visceral Leishmaniasis (VL). Most of the laboratory test results were compatible to those of the seven control animals but culture of bone marrow aspirated material and serologic testing (IIF) demonstrated or provided evidence that the animals were infected. The most important and predominant histopathological lesion in infected animals were epithelioid granulomas presented in the liver, spleen, adrenal gland and lung of some animals. Channels containing erythrocytes in some granulomas of the liver suggest that these granulomas are formed inside sinusoidal capillaries. Despite the animals were proved to be infected and presented characteristic histologic lesions, they did not present external signs of disease. The granulomatous aspect of the lesions indicates a good immunologic reactivity and suggest that a host-parasite equilibrium does exist in the dog experimental model.

Evaluation of the use of iliac cancellous bone and anorganic bovine bone in the reconstruction of the atrophic maxilla with titanium mesh: a clinical and histologic investigation.

The present article describes a titanium mesh procedure used for bone augmentation in the treatment of severe maxillary atrophy. A mix of iliac cancellous bone and anorganic bovine bone in a 1:1 ratio is proposed for achieving the best bone quality at the time of implant placement, which is performed 5 to 6 months after the augmentation surgery. This procedure provides for 3-stage surgery using a titanium mesh (which is removed 4 to 5 months later) to retain the cancellous bone/Bio-Oss mixture. Bone specimens taken 5 to 6 months after the augmentation procedure showed bone regeneration and the presence of vessels, indicating bone vitality.

[Evaluation of a prevention and prophylaxis program for the handicapped].

The growing interest regarding the importance of oral health in subjects affected with physical and psychological handicaps has led the Authors to prepare a programme of oral hygiene aiming at the prevention of dental and periodontal diseases and demanding the participation of the other members of the families, the educators and the specialized staff put at the head of their assistance. Fifty-three subjects presenting different pathologies, who have undergone elementary operations of oral hygiene, have been examined. In order to value the efficacy of these systems, DI (Debris Index), CI (Calculus Index) and OHI (Oral Hygiene Index) of Greene and Vermillion have been periodically calculated. The results obtained have been encouraging and have emphasized the importance of the behaviour of family and institutions towards people with physical and psychological handicaps, in order to let a project of primary prevention succeed.

[Investigation of circulating immune complexes in some diseases associated with platelets hyperdestruction process (author's transl)].

A possible relationship between the presence of circulating immune complexes (CIC) and the platelet-bound IgG levels was investigated in 38 patients with some diseases associated with a no drug-related platelets hyperdestruction process of which 33 had idiopathic thrombopenic purpura (ITP). By using the 125I-C1q binding test, these CIC were only detected in 4 of the 42 tested sera. No correlation was noticed between the percentage of C1q binding and the platelet IgG levels evaluated by the quantitative hemolysis inhibition test. These results do not allow to consider the involvement of CIC in the thrombocytopenia genesis observed in these diseases.
