Kinetic study of the malonaldehyde-azulene reaction determination of malonaldehyde in human plasma.

The influence of physicochemical conditions in the formation of derivatized malonaldehyde (MLD)-azulene was studied, and kinetic and conventional spectrophotometric data were compared. Conventional and kinetic spectrophotometric procedures were used to study inference of several compounds. An advantageous kinetic method was used to determine MLD in human plasma. The proposed method is faster than the thiobarbituric acid (TBA) method, because 150 s is sufficient to develop the reaction and the kinetic measurements. A comparative study of the results obtained using the proposed kinetic method and those reported in the literature for MLD determination by using the TBA reaction is presented.

Determination of sulphanilamide in milk by first-derivative and second-derivative spectrofluorimetry.

First-derivative and second-derivative spectrofluorimetric methods have been developed for the analysis of sulphanilamide (SAN) in milk, using an ethanol-water (60:40, v/v) medium. Linear calibration plots were obtained over a concentration range of 30-120 ppb, with correlation coefficients greater than 0.997. Relative standard deviations were within the range 1.6-3.6%. Limits of detection were 0.7-2.7 ppb. Recoveries of 90-102% were obtained. The derivative fluorescence methods are simple, rapid and sensitive and there is no interference from fluorescent impurities in the determination of low levels of SAN in milk.

Determination of malonaldehyde in human plasma: elimination of spectral interferences in the 2-thiobarbituric acid reaction.

A selective, derivative spectrophotometric method has been developed for the determination of malonaldehyde (MLD), based on a reaction with 2-thiobarbituric acid (TBA). The proposed method has been applied to the determination of MLD in human plasma. A study to eliminate several spectral interferences is described. A comparative study of the results obtained using the proposed derivative method and a conventional TBA method applied to human plasma is presented and the advantages of the proposed method over the conventional method for the determination of MLD in human plasma are evaluated.

Multicomponent determination of flavour enhancers in food preparations by partial least squares and principal component regression modelling of spectrophotometric data.

Three multivariate calibration methods, partial least squares (PLS-1 and PLS-2) and principal component regression (PCR), were applied to the simultaneous determination of three flavour enhancers (inosine 5'-monophosphate, guanosine 5'-monophosphate and monosodium glutamate), in mixtures by ultraviolet/visible absorption spectrophotometry. The absorption and first- and second-derivative absorption spectra of the ternary mixtures were used to perform the optimization of the calibration matrices by the PLS and PCR methods. The results obtained by the application of the different chemometric approaches are discussed and compared. No significant advantages were found for the prior differentiation step. The proposed method was applied satisfactorily to the determination of inosine 5'-monophosphate and guanosine 5'-monophosphate in several food preparations, in the presence of monosodium glutamate.

[Treatment of hepatic and intra-abdominal hydatidosis with mebendazole: prospective comparison of the cyclic and the continuous regimen].

AIM: To communicate the results of two different schedules (cyclic and continuous) of mebendazole therapy for hepatic and intraabdominal hydatid disease. DESIGN: Prospective and randomized. PATIENTS: 26 cases of abdominal hydatid disease. RESULTS: No significant differences were registered between both groups. CONCLUSION: Cyclic mebendazole therapy does not improve the results of continuous treatment in abdominal hydatid disease.

[Adrenal pheochromocytoma and metastatic intestinal carcinoid tumor in the liver. A purely accidental association?].

A patient with hepatic metastasis of an intestinal cancer tumor is presented. Six years previously the patient had been diagnosed of HTA secondary to pheochromocytoma of the left adrenal gland and surgery. The clinical findings are described and the results of scintigraphy with 123I-MIBG used for tumor detection are analyzed. The literature was revised and this unique presentation confirmed following the study and exclusion of a possible association with pluriglandular adenomatosis. Finally, following the poor results obtained with different therapeutic procedures in metastatic cancer tumors, the systematic, periodic determination of 5-HIAA, among other investigations, is suggested as a useful alternative in the early diagnosis and potential surgical treatment patients diagnosed of pheochromocytoma.

Magnetic resonance spectroscopy of inflammation associated with the temporomandibular joint.

Noninvasive early recognition and treatment of temporomandibular joint dysfunction remains a diagnostic challenge. This pilot study evaluated the use of phosphorus 31 magnetic resonance spectroscopy with magnetic resonance imaging to measure alterations in pH and high-energy phosphate metabolite ratios of muscle that is adjacent to an inflamed temporomandibular joint. Ten New Zealand white rabbits were used in this study. Two animals were used to develop signal acquisition protocols and to ensure that stable baseline data could be measured. In each of the eight animals used in the experiment, one temporomandibular joint was injected with a suspension of silica particles and the contralateral joint served as a control. Data were collected from control and experimental joints on days 0, 7, 14, 21, and 28, after the injection. At the end of the study, temporomandibular joints were block resected and histologically examined to confirm the presence of an inflammatory response. Results indicated that pH and metabolite ratios could be obtained by 31P-magnetic resonance spectroscopy. Changes in pH and some metabolite ratios in experimental joints showed statistical significance (p < 0.001). Differences were seen on day 2 and day 7 (p = 0.040 and p = 0.008, respectively) in the phosphocreatine/alpha-adenosine triphosphate ratios. This contrasts with phosphocreatine/beta adenosine triphosphate ratios that showed significance that began at day 7 (p = 0.022) and continued to day 14 (p = 0.025). Histologic examination indicated that the tissue response within the joint capsule was less than the granulomatous reaction expected.(ABSTRACT TRUNCATED AT 250 WORDS)

Simultaneous cardiac mechanics and phosphorus-31 NMR spectroscopy during global myocardial ischemia and reperfusion in the intact dog.

To investigate the high-energy phosphate metabolic correlates of left ventricular (LV) dysfunction during the onset and recovery from severe, global myocardial ischemia in vivo, seven preinstrumented closed-chest dogs had ECG-gated phosphorus-31 (31P) NMR-spectroscopy (NMR-S) studies performed and LV micromanometer and sonomicrometer data measured before, during, and every 5 min following severe occlusive global myocardial ischemia. Ischemic LV + dP/dtmax fell from 2396 +/- 576 mm Hg/s at baseline to 2185 +/- 478 mm Hg/s (p less than 0.05) and did not normalize until after 30 min of reperfusion. LV ejection fraction (EF) decreased significantly (0.32 +/- 0.07 EF units to 0.12 +/- 0.13 EF units; p less than 0.05) and did not recover by 30 min of reperfusion (0.27 +/- 0.09 units; P less than 0.05 vs baseline). Simultaneous 31P NMR-S studies demonstrated excellent beta-ATP signal-to-noise (10 +/- 4:1). Myocardial acidosis occurred during global ischemia (delta pH = -0.22 +/- 0.23 units; p less than 0.05), with recovery at 30 min of reperfusion. Inorganic phosphate/phosphocreatine ratio (Pi/PCr) increased significantly during ischemia (0.46 +/- 0.07 to 0.61 +/- 0.07; P less than 0.05), with delayed normalization of this ratio at 30 min of reperfusion. beta-ATP peak area did not change during ischemia. Pi/PCr and LV contractility (+dP/dtmax) were significantly correlated at baseline (r = -0.70) and during global ischemia (r = -0.78; p less than 0.01), but not during recovery (r = 0.006; p = NS). Therefore, the simultaneous evaluation of high-fidelity hemodynamic data and topical 31P NMR-S can be performed in the intact state.

Determination of salicylic acid and its metabolites in urine by derivative synchronous spectrofluorimetry.

The simultaneous determination of salicylic acid in binary and/or ternary mixtures and its two main urinary metabolites is proposed. Mixtures of salicylic, salicyluric and gentisic acids are resolved by synchronous spectrofluorimetry, in combination with first-derivative measurements. The urine is extracted with diethyl ether in acid medium. Salicylic and salicyluric acids are re-extracted into glycine-sodium hydroxide buffer solution of pH 11.6 and determined at that pH, and salicylic and gentisic acids are re-extracted into boric acid-sodium hydroxide buffer solution of pH 8.5 and determined at pH 6.

Extraction--spectrophotometric determination of oxalate in urine and blood serum.

An extraction--spectrophotometric method is described for the determination of oxalate, based on the formation of a mixed ligand vanadium (V)--mandelohydroxamic acid--oxalate complex. The complex was extracted into a solution of trioctylmethylammonium chloride (Adogen 446) in toluene and the absorbance measured at 535 nm. The experimental variables and interferences in this determination were studied. The detection limit is 0.5 microgram ml-1 and the range of application is between 2 and 8 micrograms ml-1. The method was applied to the determination of oxalate in urine and blood serum.

[Infected cyst of the seminal vesicle associated with malformation of the upper urinary tract and contralateral vesiculitis. Embryonal and clinical aspects].

Congenital cyst of the seminal vesicle is a rare anomaly related with developmental anomaly of the mesonephric duct that is frequently associated with unilateral malformation of the upper urinary tract. New imaging techniques (ultrasound, CT, MRI) have allowed us to rediscover and study seminal vesicle disorders and its adjacent area. These techniques have afforded a better definition of cyst characteristics, its contents and anatomic relations, thus permitting more effective surgical treatment.
