Comparison of inhibitory effects of prolinal-containing peptide derivatives on prolyl endopeptidases from bovine brain and Flavobacterium.

The inhibitory effects of proline-containing peptides and their derivatives on prolyl endopeptidases from Flavobacterium meningosepticum and bovine brain were compared. Replacement of the carboxyl terminal proline in N-blocked peptides with prolinal resulted in remarkable decreases in Ki values for both prolyl endopeptidases. Further reduction of the prolinal to prolinol led to a decrease in their inhibitory effects. Z-Pro-, Z-Val-, and Suc-Pro-prolinals were similarly inhibitory for both the enzymes with Ki values of nM order. However, the inhibitory effects of Z-Pyr-prolinal and Boc-Pro-prolinal on these enzymes were significantly distinguished: they strongly inhibited the mammalian prolyl endopeptidase with Ki values of nM order, while the Ki values of these compounds for the microbial enzyme were only of microM order. These results suggest that there are some structural differences in the S2 and S3 subsites between the two enzymes, though their substrate specificities are apparently indistinguishable.

Lack of effects of thyroid hormones on human erythrocyte acetylcholine esterase.

Effects of thyroid hormones and their metabolites such as L-T1, L-T2, L-T3 and L-T4 on human erythrocyte acetylcholine esterase were studied. The activity of the enzyme of intact erythrocytes was not affected by these hormones, though studied under various conditions. The physiological significance of the binding of these hormones to erythrocyte membranes remains unclear. Our results indicate that the acetylcholine esterase is not a suitable enzyme for cytochemical bioassay for thyroid hormones.

Cushing's syndrome associated with bilateral adrenal adenomas.

The fifth case of Cushing's syndrome with bilateral adrenocortical tumours is described. By the hormonal dynamic studies both tumours have been shown to be autonomous in the secretion of cortisol. Histopathologically, both tumours were identified as benign adrenocortical adenomas without nodules, and the right one was a so-called black adenoma. A high concentration of cortisol was obtained from both tumour tissue extracts. The differential diagnosis of Cushing's syndrome due to bilateral adrenocortical adenomas from primary adrenocortical nodular dysplasia is briefly discussed.

[Intrascrotal malignant fibrous histiocytoma: a case report and review of the literature].

A 75-year-old man visited our clinic on November 11, 1982 with the complaint of a painless mass in the right scrotum. The mass was hen's egg sized and hard. Surgery was performed under the diagnosis of testicular tumor. Right hemiscrotectomy with right inguinal orchiectomy was performed because of a scrotal skin invasion of the tumor. The spermatic cord, testis and epididymis were grossly normal. The tumor was located just beneath the testis, and it was considered to be scrotal tunicus in origin. The tumor was elastic and hard, and the cut surface showed a yellowish white and lobulated appearance. The histological diagnosis was malignant fibrous histiocytoma. Postoperatively, radiotherapy for a total dosage of 4,910 rads was given. The patient is alive and well without any sign of recurrence 19 months after operation. The 20 cases reported in Europe, America and Japan, including our case, were tabulated and some discussion is made.

[Extrarenal nephroblastoma: report of a case and review of the literature].

A 3-year-old girl with an extrarenal nephroblastoma arising from the right retroperitoneal space is described. She was admitted to our hospital with the chief complaint of abdominal pain. On physical examination, she was found to have a large (12 X 10 cm in size), firm and nontender mass in the right upper quadrant of the abdomen. The mass did not extend beyond the midsagital line. The physical examination did not reveal any particular findings or any congenital anomalies. Urinalysis and hematological data were within the normal limit. Radiological examinations including CT scan showed that the solid tumor was related to the right kidney. Under the diagnosis of right nephroblastoma, 15 micrograms/kg/day of actinomycin D was given intravenously for 5 days from October 18, 1982. The regression rate of the tumor was 78 percent on CT scan after chemotherapy. On November 22, 1982, transperitoneal nephrectomy was performed through a right paramedian incision. The tumor was found to adhere tightly to the upper pole of the kidney. The surgical specimen was 76 g in weight. A section of the surgical specimen showed an extrarenal tumor located completely outside the kidney and separated from the renal cortex by a thickened renal capsule. Histological diagnosis was extrarenal nephroblastoma showing renal capsular invasion by epithelial tumor cells. No teratomatous components were encountered in the tumor.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of huge diploic epidermoid of the temporal bone with intratumoral hemorrhage].

A case of huge diploic epidermoid which occurred in the right temporal bone and occupied almost all of the middle fossa and a part of the posterior fossa. CT of the tumor showed a low density lesion with multiform isodensity spots and a rim enhancement by CECT. Histologically, multiple old blood clots were recognized in this tumor contents. A review of the literature was made as regards CT findings of diploic epidermoid.

Primary mucoepidermoid carcinoma in the thyroid gland. A case report including an ultrastructural and biochemical study.

An extremely rare mucoepidermoid carcinoma of the thyroid gland was found in a 44-year-old woman. By light microscopic examination, the tumor consisted of mucous cells and squamoid cells, and was similar to that occurring in any other organs. By electron microscopic examination, the mucous cells displayed prominent mucous droplets, and showed no evidence of colloid production in their cytoplasms. Abundant tonofilaments in aggregates were observed in the squamoid cells. Biochemically, thyroxine (T4) and triiodothyronine (T3) were not detectable in the tissue of this tumor. These findings indicate that this mucoepidermoid carcinoma does not retain the functional properties of thyroid follicular cells; namely, no evidence was found to substantiate a follicular epithelial origin of this tumor.

Clinicopathological studies of minimal thyroid and ordinary thyroid cancers.

Clinicopathological studies were carried out on 27 patients with minimal thyroid cancer and 56 with ordinary thyroid cancer at Kanazawa University, from April, 1979 to December, 1982. There was a significant difference in the rate of preoperative diagnosis between the minimal and the ordinary cancer groups. Subtotal thyroidectomy with modified neck dissection was usually performed in both groups. The histological types in minimal cancer group included 16 papillary carcinomas, 7 nonencapsulated sclerosing carcinomas, one follicular carcinoma, one anaplastic carcinoma with squamous cell metaplasia and two multiple mixed carcinomas. There was no significant difference in the rates of intrathyroidal and lymph node metastases between the two groups. While the metastatic lesions of sclerosing carcinoma were localized to the central cervical lymph nodes, the ordinary cancer in general and the papillary variant of minimal cancer metastasized not only to the central cervical lymph nodes but also to the ipsilateral and even to the contralateral jugular lymph nodes.

Effect of cadmium-feeding on tissue concentrations of elements in germ-free silkworm (Bombyx mori) larvae and distribution of cadmium in the alimentary canal.

Silkworm (Bombyx mori) larvae were reared on an artificial diet containing cadmium (Cd) at concentrations of 5 and 80 micrograms/g wet diet from hatching to the fourth instar and then for 5 days at the fifth instar, respectively. Concentrations of Cd and other elements in the alimentary canal, Malpighian tubes, silk gland, fat body and other organs were determined simultaneously by inductively coupled argon plasma-atomic emission spectrometry. Cd was accumulated in the alimentary canal and Malpighian tubes at concentrations of 1100 and 470 micrograms/g dry wt, respectively. The distribution of Cd in the supernatants of the two highly accumulated organs were determined on an SW column by high performance liquid chromatography-atomic absorption spectrophotometry. Cd was primarily bound to inducible high molecular weight Cd-binding proteins.
