Three-dimensional reconstruction of a human metaphase chromosome from electron micrographs.

A complete human metaphase chromosome has been reconstructed from a series of electron microscopical projections obtained by tilting the specimen stage at 3 degree intervals from -60 to +60 degrees. The reconstructed structure is about 3.0 microns long, 1.6 micron wide, and 0.8 micron thick. The mass distribution was fairly homogeneous within the chromatids and neither a hollow nor a dense core was observed. The distribution and course of fibers observed are most consistent with a looping model of chromosome structure.

[31P NMR spectroscopy of muscular diseases: correlation with MR imaging].

The spectra of normal and abnormal muscle were examined by 31P MR spectroscopy using a 2 Tesla system. Exercise tests on normal and abnormal muscle were also carried out. Abnormal muscles showed characteristic changes in the phosphorus spectrum, when compared with normal muscles, both at rest and after exercise; these were compared with the data from the MR images. The advantages and limitations of the method are discussed.

[Importance of MR tomography for the diagnosis of bone marrow diseases in childhood].

As a diagnostic and follow-up method of bone marrow diseases in children, magnetic resonance imaging (MRI) possesses special importance. It is characterised by high sensitivity in detecting bone marrow lesions, high soft tissue--bone tissue contrast, sharp demarcation of intraosseous lesions and visualisation of vascular structures without application of contrast medium. Thus it is able above all to take on the findings of radionuclide bone scanning, computed tomography and angiography. In the detection of pathophysiological alterations of the corticalis and of calcifications it is less reliable than x-ray images and CT.

Magnetic resonance imaging of aneurysms and thrombi.

This is a report of the first systematic investigation of the qualitative and quantitative diagnosis of aneurysms in the regions of the left ventricle and thoracic and abdominal aorta plus proof of intracavitary thrombi in the heart and the aorta, as well as aneurysms in the superior and inferior vena cava. For diagnosis of the heart, ECG gating is an absolute necessity, but for the analysis of abdominal aortic aneurysms it only leads to a considerable improvement of the spatial resolution. For differential diagnosis of the blood flow and intracavitary clots in the heart and the aorta, use of a second or even multiple echoes is needed. Also, digital subtraction between the first and second echoes (magnetic resonance digital subtraction) can assist in assessing flow. When dissecting aortic aneurysm is suspected and in cases when risk of perforation of ventricular and aortic aneurysms is present. MR offers particular advantages, since it is noninvasive and few scans can provide all the information that is required.

[Nuclear resonance tomography of the breast--diagnosis, differential diagnosis, problems and solutions. I. Study procedures].

The experience gained from performing nuclear resonance tomography on the breast, using a special surface coil, is described. The technical problems, causes of artefacts and the advantages of tomography in three planes are discussed. The procedure is slow. Images obtained by various repetition and echo delay times in several planes are illustrated. Problems in quantifying T1 and T2 images and methods for improving the technique are described in detail. During the course of 85 examinations on 72 patients, it has been possible to achieve technical improvements.

[Nuclear magnetic resonance tomography of the breast: diagnosis, differential diagnosis, problems and possible solutions. II: Diagnosis].

Eighty-five MR examinations were carried out on both breasts of 72 patients. A special surface coil was used. The results were compared with the findings of mammography and with the post-operative findings. There were 16 fibrocystic mastopathies, seven isolated cysts, 22 fibro-adenomas and 25 carcinomas. Differentiation between fibro-adenomas and carcinomas is only possible on the basis of morphological criteria. Cysts and fibrocystic mastopathies can be identified with certainty. Typical patterns of the various pathological conditions are described, but these will have to be confirmed by the study of a larger number of patients, in order to make them statistically significant. The advantage of MR consists in the simultaneous demonstration of the thoracic wall and the axilla.

[Nuclear magnetic resonance tomography in childhood].

To our present knowledge, MR tomography is an imaging procedure with neither ionizing radiation nor invasiveness requiring in children a good preparation and sedation. Especially in the neuro-pediatric field, and in the diagnostics of diseases of the soft tissue the high sensitivity is just as it is of clinical significance in bone tumours and their differential diagnosis. Extensive clinical-empirical examinations are still required to learn about the specificity of the different diseases.

[Percutaneous transluminal recanalization of chronic stenoses and occlusions of peripheral arteries].

It is reported on indications and results of percutaneous transluminal angioplasty using the Grüntzig- and Olbert-type balloon catheter technique. It is also referred to the expansion of PTA to the treatment of multiple obliterations in different layers with vascular surgical procedures. Today the essential indications for PTA are iliac stenoses on one or both sides, stenoses of the femoral and popliteal artery as well as short occlusions of the superficial femoral and popliteal artery of less than 10 cm.

Molecular resolution electron micrographs of monolamellar paraffin crystals.

A liquid helium-cooled cryoelectron microscope, operated to expose the specimen to only a very low electron dose, was used to obtain structural images of monolamellar n-tetratetracontane (n-C44H90) crystals at 0.25-nanometer resolution. These results are in contrast to earlier predictions that such extremely beam-sensitive materials could not be studied directly at this level of detail. Analysis of the resultant lattice images gives direct evidence for crystal bending as well as direct visualization of edge dislocations in this material.

[ECG-triggered NMR-tomography of the heart. Pathological findings].

Initial systematic examinations of the heart by ECG-triggered NMR are reported. Twenty-three abnormal findings were observed amongst 46 patients examined. Special indications are the search for cardiac aneurysms, intracardiac thrombi and changes from pressure and volume stress.
