[A case of acute promyelocytic leukemia complicated with severe esophageal stenosis caused by aclacinomycin A].

Aclacinomycin A (ACM) is different from other anthracycline antibiotics in its antitumor activity and clinical effect. We report a case of acute promyelocytic leukemia complicated with severe esophageal ulcer by BH-AC . AMP therapy. The patient was 24 years old. In 1983, he achieved complete remission with BH-AC . DMP therapy but we confirmed relapse in April, 1984. ACM caused nausea, vomiting, diarrhea, alimentary tract bleeding and sore throat, together with a complication of esophageal stenosis. In about half of 10 cases receiving BH-AC . AMP therapy in our hospital, we noticed severe bleeding in the urinary treat, genital organs and alimentary tract. Care should therefore be taken with regard to mucosal injury when ACM therapy is used.

Loss of vision-guided adaptation of the vestibulo-ocular reflex after depletion of brain serotonin in the rabbit.

Brain serotonin in pigmented rabbits was depleted by intraventricular injection of 5,7-dihydroxytryptamine (5,7-DHT). Before depletion, an adaptive increase in the gain of the horizontal vestibulo-ocular reflex (HVOR) could be induced regularly by continuously rotating the animals in combination with optokinetic stimulus. After the depletion, such an increase in HVOR gain did not occur, even though dynamic characteristics of the HVOR and of the optokinetic eye movement were not altered. 5,7-DHT treatment also reduced brain norepinephrine, but depletion of norepinephrine to a similar extent by intraventricular injection of 6-hydroxydopamine in other rabbits did not affect the HVOR adaptation. These results suggest that brain serotonin plays an important role in maintaining adaptive modifiability of the HVOR.

Functional representation of eye movements in the flocculus of monkeys (Macaca fuscata).

The flocculus of the monkey was mapped with microstimulation methods in chronic preparations. Three types of low threshold, disconjugate eye movements were elicited: (1) a lateral deviation of the ipsilateral eye, (2) a downward deviation of the ipsilateral eye and (3) counterclockwise rotation of the ipsilateral eye. Horizontal eye movements were obtained from folia 2, 3, 4 and 6-10 of the flocculus, and the region eliciting horizontal eye movements was traced in the sagittal plane over a distance of 4 mm in one animal. However, sites flanking these horizontal movement sites produced either vertical movements, rotatory movements or failed to produce a movement of either eye. These findings suggest a zonal organization of flocculus functions in monkeys that is consistent with the differential projections of flocculo-vestibular connections revealed by retrograde tracing studies [2].

Neuronal events correlated with long-term adaptation of the horizontal vestibulo-ocular reflex in the primate flocculus.

The activity of flocculus Purkinje cells was examined in Japanese monkeys during sustained vestibular-visual stimulation which caused adaptation of the horizontal vestibulo-ocular reflex (H-VOR). In the floccular area related to the H-VOR by microstimulation. Purkinje cells consistently changed their simple spike responsiveness to head rotation in parallel with the adaptive H-VOR gain change. Similar changes occurred even after the H-VOR had been extinguished by lesioning of the vestibular nuclei. The complex spike discharge, on the other hand, modulated during vestibular-visual stimulation with a reciprocal pattern to the adaptive changes in the simple spike discharge. These results support the hypothesis that the flocculus adaptively modifies the H-VOR through their simple spike activity under the influence of visual climbing fiber signals.

Bioconcentration potential of polydimethylsiloxane (PDMS) fluids by fish.

The water solubilities (S), octanol/water partition coefficients (KOW) and bioconcentration factors (BCF) of four polydimethylsiloxane (PDMS) fluids covering a wide range of molecular weight were measured. It is shown that a previously described correlation between S and KOW for organic chemicals may be invalid for PDMS fluids; an alternative correlation is proposed. Some PDMS fluids tend to have a bioconcentration potential in silver carp.

Changes of platelet membrane associated Ca2+ during storage and by 37C incubation with fresh plasma.

The relationship between platelet aggregability and membrane associated Ca2+ measured by the use of chlortetracycline was studied. When platelet concentrates were stored at room temperature for 3 days, the relative value of platelet membrane-Ca2+ amount increased. But when stored platelets were incubated at 37C with fresh plasma, platelet membrane-Ca2+ decreased, and at the same time, ADP-induced aggregability increased. The relative value of membrane-Ca2+ amount and ADP-induced aggregability exhibited reverse correlation. According to these results, it is supposed that one of the factors which cause the decreased platelet aggregability during storage and partial recovery by 37C incubation with fresh plasma, is the change of the distribution of platelet Ca2+.

Monoclonal antibodies to the HN glycoprotein of Newcastle disease virus. Biological characterization and use for strain comparisons.

We have isolated nine hybridomas secreting monoclonal antibodies directed against the HN glycoprotein of D26 strain of Newcastle disease virus (NDV) and classified them into three groups by their biological reactivities. The group I antibodies from six clones had high hemagglutination inhibition (HI) and neutralization titers, and exhibited neuraminidase inhibition (NI) activity with substrates, both bovine fetuin and N-acetyl neuraminlactose, although the levels of maximum inhibition were greater with the former substrate. The group II antibodies from two clones also had high HI activity but showed a strongly substrate-dependent NI, with efficient inhibition against fetuin and a total lack of inhibition against neuraminlactose. These antibodies had neutralization capacity but the titers were much lower than those of group I antibodies. One clone (group III) secreted antibody which exhibited HI and NI with either substrate but no detectable neutralization activity. Frequency of isolation of antigenic variants and competitive binding assays have shown that group I and group II antibodies may recognize distinct regions not overlapping each other whereas antibodies placed within each of the two groups are to the respective same overlapping regions. The site of group III seemed to be close to or overlap the group I determinant but distinct from group II region. Using the isolated monoclonal antibodies we compared seven heterologous strains by HI and neutralization tests as well as by enzyme-linked immunosorbent assay. The results indicated that the regions recognized by group I antibodies would be conserved among the strains whereas those by the other two antibody groups might undergo considerable changes.

High-grade tumor-specific immunity induced by L1210 leukemia variants obtained from the culture of L1210 cells fused with Lesch-Nyhan fibroblasts.

A highly immunogenic variant of the murine L1210 leukemia cell (L1210/LN-1) for generation of tumor immunity has been obtained from culture of L1210 cells originally fused with human Lesch-Nyhan fibroblasts. In L1210/LN-1 cells, no human chromosomes were identified and chromosomes M1 and No. 1 carried by the parent L1210 cells were missing. L1210/LN-1 cells displayed an intermediate morphology between L1210 cells and Lesch-Nyhan fibroblasts. Most of the CDF1 mice that were inoculated with less than 2 X 10(6) L1210/LN-1 cells survived over 60 days without evidence of tumor, whereas the original L1210 cells killed all the mice tested in about 2 weeks. When inoculated with more than 5 X 10(6) L1210/LN-1 cells, CDF1 mice developed tumor. The CDF1 mice which rejected 2 X 10(6) L1210/LN-1 cells were protected very effectively against challenge of otherwise highly aggressive 1-5 X 10(5) L1210 leukemia cells; 40 out of 43 primed mice tested survived for 60 days or longer after the tumor challenge. Even the CDF1 mice primed with irradiated or mitomycin-treated L1210/LN-1 cells survived against a challenge of 10(5) L1210 leukemia cells. They were, however, not protected against P388 leukemia or Meth A sarcoma, indicating that the immunity was specific to L1210 leukemia. The immunity induced by L1210/LN-1 cells was transplantable by immune spleen cells into syngeneic recipients. Thus, the L1210/LN-1 cells we obtained seem to be very useful as an immunogen for generation of high-grade tumor-specific immunity against highly malignant L1210 leukemia.

A new method for detection of anti-zona activity in human sera using latex agglutination reaction.

A slide latex test for detection of anti-zona pellucida activity in human sera was developed using a latex agglutination reaction. The latex reagent, made of polystyrene particles coated with solubilizing zona antigen(s), was found to give results comparable in sensitivity as well as specificity to those of the indirect immunofluorescence method as tested with anti-pig-zona antiserum and with human sera. Thus, the slide latex test was judged to be adequate for use instead of the indirect immunofluorescence technique.

Changes of platelet cell volumes in hypotonic solution.

Changes of platelet cell volumes in hypotonic solution were studied by the use of a new apparatus for cell volume analysis. Continuous mean cell volume analyzer (CMA) is a computerized instrument which continuously counts the mean cell volume and records the results. Moreover, it also shows patterns of expansion or shrinkage. Adequate amounts of platelet rich plasma were added into 0.8 or 0.4% of NaCl solution containing 5mM phosphate buffer. The mean platelet volumes were measured continuously every 2 seconds for 200 seconds. In isotonic solution, platelets changed their volumes little, but in hypotonic solution, the volumes increased for about 20 seconds and then, decreased gradually. Platelet volume expansion ratio and shrinkage ratio were calculated from these patterns. The shrinkage ratio well corresponded to the values of hypotonic shock response measured by spectrophotometry (r = 0.861). Expansion and especially shrinkage ratios decreased during storage, and they were dependent on stored temperature and pH.

Heme oxygenase activity in liver microsomes of the bullfrog, Rana catesbeiana, and its change during the tadpole development.

Heme oxygenase in the liver microsomes of tadpole and adult bullfrog, Rana catesbeiana, was studied in relation to hemoglobin metabolism during tadpole development. The results obtained are as follows. 1. The specific activity of the enzyme of premetamorphic tadpole liver was comparable to that of adult frog liver. The apparent Km for methemalbumin was about 25 microM for both tadpole and frog enzymes. 2. The enzyme activity was stimulated in vivo by the injection of methemalbumin, phenylhydrazine and triiodothyronine to the animals. 3. A marked increase in the enzyme activity was found in the liver of tadpole during prometamorphic stage and metamorphic climax.
