Stem Extension Rate in Light-Grown Plants : Effects of Photo- and Thermoperiodic Treatments on the Endogenous Circadian Rhythm in Chenopodium rubrum.

Low temperature pulses have two effects on the circadian rhythm exhibited by stem extension rate of green Chenopodium rubrum plants. First, low temperature pulses have the same effect on the phasing of the rhythm as a dark period interrupting continuous light. Second, low temperature pulses stimulate stem extension rate during the 10 hours immediately following the end of the pulse. A difference in temperature between soil and air increases this effect. In any case, it is the change in temperature which is essential and not a specific temperature. Effects of light and temperature on phasing and amplitude of the rhythm explain why the maximal stem growth is observed under normal photo-thermoperiodic conditions, i.e. a high temperature during the photoperiod and a low temperature during the dark period.

Direct demonstration that the abundant 6-kilobase herpes simplex virus type 1 mRNA mapping between 0.23 and 0.27 map units encodes the major capsid protein VP5.

The two partially colinear 6-kilobase (kb) and 1.5-kb mRNAs mapping between 0.23 and 0.27 map units on the herpes simplex virus type 1 genome were precisely located. The 5' end of the 6-kb mRNA was located 28 bases downstream of the sequence ATATATT and was 10 bases to the left of the BamHI site at 0.268. This position is ca. 90 bases to the left of our earlier reported sequence (R. J. Frink, K. G. Draper, and E. K. Wagner, Proc. Natl. Acad. Sci. U.S.A. 78:6139-6143, 1981). We used a polyclonal antibody made against purified herpes simplex virus type 1 VP5 to demonstrate that the 155,000-dalton translation product of the 6-kb mRNA is this capsid protein. The antibody did not react with the 35,000-dalton translation product of the 1.5-kb mRNA. We also confirmed our identification of VP5 as the translation product of the 6-kb mRNA by comparison of tryptic peptides of the in vitro-translated protein and authentic VP5.

Effects of a pyridine derivative thromboxane synthetase inhibitor and its inactive isomer in endotoxic shock in the rat.

1 We investigated the effects of a pyridine derivative thromboxane synthetase inhibitor and its inactive ortho isomer on arachidonic acid metabolism and pathophysiological sequelae of endotoxic shock. In vehicle-treated rats, 30 min after intravenous S. enteritidis endotoxin (15 mg/kg), plasma iTxB2 (the stable metabolite of thromboxane A2) increased from non-detectable levels (less than 100 pg/ml) to 763 +/- 250 pg/ml (n = 10). Plasma i6-keto-PGF1 alpha (the stable metabolite of prostacyclin, PGI2) increased to 1850 +/- 426 pg/ml, (n = 9) and plasma iPGE increased to 2350 = 560 (n = 5). Pretreatment with the pyridine active (PA) meta isomer (30 mg/kg i.p.) significantly (P less than 0.05) suppressed iTxB2 to 390 +/- 31 pg/ml (n = 10) although 6-keto-PGF1 alpha levels (1294 +/- 358 pg/ml, n = 5) and plasma iPGE (2847 +/- 1103 pg/ml, n = 5) were not significantly different from the shocked control values. In contrast, pretreatment with, the pyridine inactive (PI) ortho isomer did not significantly affect endotoxin-induced iTxB2 (1431 +/- 194 pg/ml, n = 5) or i6-keto-PGF1 alpha synthesis (628 +/- 266 pg/ml, n = 5). 2 Pretreatment of rats with the Tx synthetase inhibitor, PA, significantly enhanced (P less than 0.05) survival and prevented splanchnic infarction relative to both endotoxin shocked control rats and those pretreated with the PI isomer. 3 Significantly reduced lysosomal labilization, hepatocellular dysfunction and elevations in serum fibrin/fibrinogen degradation products were seen only in groups pretreated with the PA isomer. 4 The beneficial effects of the latter compound in endotoxic shock thus appear to be due to inhibition of Tx synthesis, since its ortho isomer did not inhibit TxA2 synthesis nor did it protect against endotoxic shock.

The subcapsular hydrostatic pressure of allogeneic kidney grafts: a new clinical method for postoperative transplant monitoring.

A method for the measurement of the subcapsular hydrostatic pressure in human kidney grafts is described. This method permits follow-up studies over periods up to 4 weeks. First results indicate that the method is a useful tool to monitor the condition of renal allografts in the early postoperative period. It allows the early recognition of acute rejection episodes and predicts their course and reversibility. The subcapsular hydrostatic pressure is a particularly valuable parameter in the differentiation of acute rejection and acute tubular necrosis.

Postoperative retinal fibrosis--a controlled clinical study of systemic steroid therapy.

In a prospective clinical trial, the effect of systemic steroids on the incidence and severity of retinal fibrosis following retinal detachment surgery was tested against placebo. Several grades of retinal fibrosis of various degrees of severity were defined prior to the study. The maximum postoperative incidence of eyes with any signs of retinal fibrosis was 65% (75.4% in the placebo group, 63.3% in the steroid group). A set of fibrosis criteria was identified that had a tendency to respond to systemic steroids with a statistical significance increasing with time. The majority of these steroid-responsive criteria was of a mild degree and located at the posterior pole. The mean visual acuity was significantly lower in eyes with retinal fibrosis. The effect of systemic steroids on retinal fibrosis tested under clinical conditions was considerably weaker than that known from experimental conditions with intravitreal injections of triamcinolone acetonide.

[Inner retinal traction breaks resembling a sharkmouth in areas of peripheral cystoid degeneration (author's transl)].

10 predominantly diabetic eyes from a mixed collection of 1,561 enucleated specimens showed extended, linear, ridgelike elevations of the inner retinal layers within areas of cystoid degeneration. These ridges lie peripherally and run parallel to the ora. The posterior slope of each ridge contains a chain of inner retinal holes which extend into the cavities of the cystoid degeneration. The changes, viewed from behind, resemble an open sharkmouth. The posterior border of the vitreous base adheres to the crest of the ridge behind which the vitreous itself is detached. The ridges and inner holes are thought to result from vitreous traction, chronic rather than acute, acting on peripheral areas of cystoid degeneration which can presumably be elevated more easily than normal retina.

4-hydroxyquinoline-3-carboxylic acids as inhibitors of cell respiration. 2. Quantitative structure-activity relationship of dehydrogenase enzyme and Ehrlich ascites tumor cell inhibitions.

Studies on dehydrogenase enzyme inhibition have been extended with the design, synthesis, and correlation analysis of 7-[(substituted-benzyl)oxy]-, 7-[(substituted-phenethyl)oxy]-, and 7([substituted-phenoxy)ethoxy]-4-hydroxyquinoline-3-carboxylic acids. Sixteen new congeners and the fifteen molecules previously synthesized have been tested against cytoplasmic malate dehydrogenase and lactate dehydrogenase, as well as against mitochondrial malate dehydrogenase. The lipophilic congeners show a clear specificity for inhibition of the mitochondrial enzyme. Correlation analysis of the data on the three enzymes allows a comparison of the binding sites in quantitative terms, while examination of the data on inhibition of ascites tumor cell respiration affords an indication of membrane transport. A newly developed high-pressure liquid chromatography based retention index is compared to the octanol-water pi constant as a model for hydrophobic interactions.

[Measurement of the subcapsular pressure of allogeneic kidney transplants: a new method for postoperative transplant monitoring].

The results presented here show the measurement of the subcapsular hydrostatic pressure (SHP) to be a suitable method for the postoperative monitoring of a allogeneic kidney transplants. The SHP of the kidney reflects the interstitial pressure of this organ. Disturbances in the graft of immunological or non-immunological origin induce pressure increases with, for the most part, specific characteristics. An examination of the height and course of the mean subcapsular pressure level enables one to recognise and differentiate disturbances in transplant function at an early stage as well as assess their severity and duration. The results obtained so far show that the measurement of the subcapsular pressure is of particular significance in determining the extent of ischemic tubular damage, in early recognition of acute rejection and i assessing the effectiveness of immunosuppresive treatment for rejection. Repeated pressure measurements with the method described here can be performed with little technical outlay and without any stress for the patient.

Early experience of transluminal coronary angioplasty (TCA) by the brachial artery (the Sones technique in transluminal angioplasty).

Sixteen transluminal coronary angioplasty procedures (TCA), eight right coronary artery (RCA) and eight left anterior descending coronary artery (LAD), by the brachial artery cut-down approach, were attempted with 9/16 (56%) immediate successes and 2/16(12%) early recurrences. The procedure success rate for RCA obstructive lesions, 6/8 (75%) was greater than for LCA obstructions, 3/8 (38%). In six unsuccessful procedures the balloon catheter could not be advanced into the lesion, and in one unsuccessful procedure dissection of the coronary artery proximal to the lesion occurred. The brachial (Sones) technique for transluminal coronary angioplasty permits the use of softer guiding catheters for selective probing and approach to the coronary lesion but may be more likely to induce coronary spasm. Complete and high-resolution pre TCA angiograms with multiple views to disclose the exact anatomy of the coronary artery and its lesion is essential to ensure successful dilatation.
