Impaired development of the rat perineurium by undernutrition. An enzyme histochemical study.

Enzyme activity of adenosinetriphosphatase, NADH2-tetrazolium reductase and lactate dehydrogenase was studied in the rat perineurium 10, 20, 30, and 90 days post partum. The effect of under-nutrition was shown by a lack of enzyme activity in rats 30 days post partum in contrast to control animals, 30 days also corresponds to the time when, in normal rats, the perineurial diffusion barrier to exogenous macromolecules becomes established. In undernourished rats this diffusion barrier does not appear. At 90 days post partum raised enzyme activities indicate a high metabolic turnover in the perineurium even in animals with undernutrition.

Effect of vasotocin on plasma free fatty acid level in the migrating anadromous sea lamprey.

The effect of injections of arginine vasotocin (AVT) on plasma free fatty acid (FFA) levels was studied in anadromous sea lampreys collected in the St. John River, New Brunswick, during their upstream spawning migration. Plasma FFA was significantly higher in lampreys injected with a single dose of 1 000 mU vasotocin/kg body weight than in those receiving only the vehicle solution, the difference being the greater at 90 than at 30 min post-injection. The significance of AVT in migration is discussed.

Chronic calcific pancreatitis in association with celiac disease.

Painless pancreatitis with extensive calcification and villous atrophy of the small intestine was observed in a nonalcoholic female patient. The etiology of villous atrophy could not be established. The pathophysiological significance of this association is discussed.

[Surgical treatment in deep venous thrombosis of the legs and pelvis (author's transl)].

The only effective causal therapy in thromboses of the deep pelvic and femoral veins is fibrinolysis or operative thrombectomy. Concerning the contraindications, fibrinolysis is successful only up to the 5th day. After that time patients with deep vein thrombosis were submitted to thrombectomy and at the same time, to prevent renewed thrombosis secondary to a slowing down of the flow, a peripheral arteriovenous fistula was temporarily positioned. Between 1974 and 1976, 14 patients were treated operatively this way. In 6 cases of pulmonary embolism a vena cava umbrella filter was inserted.

Spectrum of methyldopa liver injury.

The administration of alpha methyldopa resulted in serious liver injury to seven patients, requiring hospitalization. All patients presented with symptoms indistinguishable from viral hepatitis. Histological changes ranged from those compatible with acute hepatitis to chronic active hepatitis and subacute hepatic necrosis. One patient with subacute hepatic necrosis rapidly progressed to hepatic failure and died; autopsy revealed postnecrotic cirrhosis. Inadvertent rechallenge after recovery resulted in dramatic recurrence of symptoms in one patient, confirming the fact that a hypersensitivity phenomenon is involved. A high state of awareness among physicians of this unpredictable toxic effect of the drug may reduce its unrecognized perpetuation.

Effects of a novel anti-aggressive agent upon two types of brain stimulated emotional behavior.

The effects of anti-aggressive agent Sch 12679 were evaluated upon stable baselines of rage and predation elicited by electrical stimulation of the hypothalamus in cats. Sch 12679 depressed approach and terminal aspects of both forms of attack. This is consistent with previous reports, and suggests the drug is effective in reducing many forms of aggression including brain stimulated emotional behavior.

Effects of para-chlorophenylalanine upon brain stimulated affective attack in the cat.

Previous investigations of the effects of parachlorophenylalanine (PCPA) on affective attack have produced equivocal results. The present report examined the effects of PCPA upon attack elicited by electrical stimulation of the ventromedial hypothalamic nucleus in the cat. Using a within-animals design and postdrug testing clear potentiation of affective attack patterns was observed for a number of the components of attack behavior (approach, use of forepaws, and biting). The present results may point to possible serotonergic involvement in the inhibitory control of several aspects of affective attack.

Paroxysmal nocturanal hemoglobinuria and marrow failure treated by infusion of marrow from an identical twin.

An 18-year-old white Canadian male patient with paroxysmal nocturnal hemoglobinuria and refractory marrow failure received a marrow infusion from his normal identical twin brother without prior ablation of the patient's marrow by drugs or irradiation. After 2 years of follow-up, the patient is well with no evidence of clinical disease or significant hematological abnormality. The results suggest that normal marrow stem cells can have a selective advantage over the abnormal paroxysmall nocturnal hemoglobinuria clone.

Chemical and Drug Induced Liver Injury↗

Methyldopa-induced liver injury. Rapid progression to fatal postnecrotic cirrhosis.

A 55-year-old woman developed symptoms suggestive of hepatitis 12 weeks after first receiving methyldopa for hypertension. Liver biopsy showed chronic aggressive hepatitis with subacute hepatic necrosis. Methyldopa was discontinued, but after exhibiting transient clinical improvement, the patient's condition progressively deteriorated until she died of hepatic failure, in spite of therapy with massive doses of corticosteroids and other nonspecific measures. During the terminal stage, a considerable decrease in the size of the liver was observed. At autopsy, the liver was found to be small, shrunken, and scarred; histological sections demonstrated postnecrotic cirrhosis. Such a rapid and relentless progression of methyldopa-induced liver injury is undoubtedly rare, but it may be prevented by careful supervision of patients who exhibit liver function abnormalities early in the course of therapy.

[Postoperative prophylaxis of thromboembolism. Present position.(author's transl)].

Only anti-coagulators and aggregation-stoppers may be said to be the medicamental prophylaxis of thromboembolism. The Cumarin-Indandion group is burdened with continuous laboratory controls. Contraindications excluding a general prophylaxis for this group as well as for Heparin and ASS exist. Infukoll, which is virtually always applicable, has the disadvantage that it has to be infused. Heparin- and Dextranspreparations however can already be used preoperatively, which means that a further decline of the coagulation complications can be expected.

[Basic-diagnostic-criterions of respiratory insufficiency--results of a community work of the working group "pathophysiology of breathing" (author's transl)].

11 laboratories of the working group "Pathophysiology of Breathing" cooperated with standardized methods to obtain generally admitted practical criterions for the definition of respiratory insufficiency. Function parameters of 2800 examined persons in different diagnostic groups were tested by manual statistical analysis and computed by data processing system. which were assessed with regard to their usefulness as criterions for respiratory insufficiency. The arterial O2-partial pressure proved to be the most crucial criterion of respiratory insufficiency permitting objective evidence of a manifest respiratory insufficiency by hypoxeamia under standard efforts with a bicycle-ergometer. The screening parameters of ventilation such as vital capacity and forced exspiratory volume are suitable as subjective rapid methods for a quantitative estimation of one type of respiratory insufficiency: the ventilation disturbance. Investigations, which were carried out with a higher apparatus expense like the estimation of residual volume, the functional residual capacity, and the arterial CO2-partial pressure permit the further differentiation of causes of the respiratory insufficiency and the separation between respiratory partial- and global insufficiency. The parameters of gas exchange: O2-consumption, respiratory quotient, and the screening parameter of cardiocirculation heart rate are not or only limited useful as criterions of respiratory insufficiency alone.

[Studies on endocrine and spermiogenic testis function. IV. Relations between the testosterone content of the testes cell nucleus volume of the Leydig cells and rat spermatogenesis (deduced from studies on the effect of the dietary protein quality on the function of the testes)].

The following relations were derived from large-scale testing of feed protein quality and its bearings on testicular function in growing Wistar rats: testicle weights and live weights r = 0.77 for juvenile animals and r = 0.36 for adult animals; nuclear volumes of Leydig's cells to testicle weights r = 0.84 for juvenile animals and r = 0.43 for adult animals; testosterone in testicular tissue, ng/g, to testicle weights r = 0.55 for juvenile animals and r = 0.80 for adult animals; testosterone per live weight, ng/100 g, to nuclear volumes of Leydig's cells r = 0.76 for juvenile animals and r = 0.46 for adult animals. Critical thresholds above which complete spermatogenesis is always ensured may be established for testosterone (50+/-5 ng/100 g live weight) and for the nuclear volume of Leydig's cells (30 mum3). Another conclusions drawn from the above results is that high testosterone levels in the testes of adult animals were not attributable to high testosterone biogenesis of Leydig's cells, as might have been assumed from high activities of that kind recordable from some nuclear volumes.

Enzyme-histochemical studies of "retrograde" reaction in motor neurones of immature rats.

Changes in immature rats in motor neurones after axotomy were studied by enzyme-histochemical methods. Increased activity of dehydrogenases in these neurones demonstrates enhanced metabolism and there was also increase of acid phosphatases. Decreased activity of acetylcholinesterase and indoxylacetate esterase in the neurones and their processes seems to indicate impaired neuronal function to transmit impulses. "Retrograde" reaction in the immature and the grown up animal is in general of the same kind but takes place quicker in the immature rat. However, in new-born and very young animals, it is difficult to recognize alterations in the anterior horn of the spinal cord. Therefore, nervous tissue of new-born animals seems not to respond as it does some days later in ontogenesis.

Effects of scopolamine and alpha-methylparatyrosine upon predatory attack in cats.

The effects of scopolamine were compared with the effects of alpha-methylparatyrosine methyl ester (AMPT) upon several aspects of electrically elicited predatory attack in cats. Thresholds of current necessary to elicit predation increased under both drugs. Furthermore, specific deficits developed in suprathreshold predatory patterns for both drugs compared with nondrug baselines. Scopolamine suppressed all aspects of attack, while AMPT suppressed approach of the attack object while leaving reflexive biting intact.
