Clinical and EEG studies of zotepine, a thiepine neuroleptic, on schizophrenic patients.

The overall effect of zotepine was a "slightly improved" or better response in 20 patients (64.5%), "unchanged" in 10 (32.3%) and "worsened" in 1 (3.2%). Zotepine exhibited some degree of improvement in 54.5% of patients unresponsive to prior drugs. The onset of effect of zotepine was within one month in 19 patients. The improvement rate in the hebephrenic type (66.7%) was almost the same as in the paranoid type. The improvement rate classified by psychopathology was highest for hypobulia, followed by restlessness-excitement and hallucination, depressive mood, hypochondria and delusion. The side-effects were subjective complaints, such as general fatigue, dryness of mouth, sleepiness or fainting in a small number of cases. There was a slight increase in S-GPT in one patient and a slightly increased blood platelet count, also in one patient. Serial EEG changes associated with zotepine studied in another 17 chronic schizophrenics could be classified into three groups: those with increased slow waves, those with enhanced alpha waves and those with unchanged EEGs. There was a positive correlation between the incidence of slow waves and higher plasma levels of zotepine.

Erythorbic acid content in tissues of guinea pigs administered erythorbic acid.

The erythorbic acid (ErA) content in the tissues of guinea pigs administered ErA was compared with that of ascorbic acid (AsA). Guinea pigs were administered 1, 5, 20, and 100 mg ErA/day or 1, 5, and 100 mg AsA/day for 16 days and then sacrificed. The liver, adrenal glands, spleen, and kidneys were removed to determine the contents of ErA and AsA using HPLC. Only a small amount of ErA was found in four tissues of the animals administered 20 mg or more of ErA/day. On the contrary, AsA was found in the tissues of all animals administered 1 mg or more of AsA/day. The ErA content in the tissues was much lower than that of AsA even when the amount of ErA administered was the same as that of AsA. However, the body weight gains of animals administered ErA were similar to those of animals administered AsA. These observations suggested that the mechanism of the retention of ErA in the tissues was much different from that of AsA and that the vitamin C activity of ErA might be more than one-twentieth that of AsA.

Detection of antibodies to Sendai virus in mouse sera by enzyme-linked immunosorbent assay (ELISA).

Enzyme-linked immunosorbent assay (ELISA) for diagnosis of Sendai virus infection in mice was evaluated. A large-scale survey of infected mice showed that ELISA is approximately 100 times more sensitive than the hemagglutination-inhibition or complement-fixation test. Although a few SPF mice showed false-positive reactions at a serum dilution of 1:40, further dilution to 1:80 eliminated the non-specific reaction. It was shown that ELISA is a highly satisfactory method for examination of Sendai virus infection in mice.

Activation of N-nitrosodialkylamines by near-ultraviolet irradiation: formation of directly-acting mutagens and DNA-damaging products.

On near-ultraviolet (UVA) irradiation in a phosphate buffer, N-nitrosomorpholine (NMOR) and N-nitrosopyrrolidine (NPYR) were converted into directly-acting mutagens. The activated NPYR was fractionated, and the active product was isolated. The compound was shown to be identical to alpha-phosphonooxy NPYR on the basis of several properties: retention times in high-performance liquid chromatograms, mutagenic specificity and potency, ultraviolet spectrum, and inactivation by phosphatase treatment. Photoactivation was inhibited by superoxide dismutase, and therefore superoxide is implicated as playing a key role in mutagen formation. N-Nitrosoproline (NPRO) and eighteen other N-nitrosodialkylamines were irradiated with UVA in the presence of phi X174 RFI DNA. The DNA underwent single-strand breaks to give RFII DNA, indicating that N-nitrosodialkylamines in general have this property. DNA chain cleavage was inhibited both by superoxide dismutase and hydroxyl-radical scavengers. These results provide new information on the genotoxic mechanism of action of N-nitrosodialkylamines.

Urinary metabolites of N-nitrosodibutylamine and N-nitrodibutylamine in the rat: identification of N-acetyl-S-alkyl-L-cysteines.

N-Acetyl-S-butyl-L-cysteine, N-acetyl-S-3-oxobutyl-L-cysteine and N-acetyl-S-3-hydroxybutyl-L-cysteine have been isolated and identified (as their methyl esters) from the urine of rats given N-nitrosodibutylamine (NDBA), N-nitrodibutylamine (NO2DBA) and their 1-acetoxy derivatives. Greater amounts of these N-acetyl-S-alkyl-L-cysteines were detected in the urine after administration of NDBA than of NO2DBA, and greater urinary excretion of the three N-acetyl-S-alkyl-L-cysteines was observed with 1-acetoxy NDBA than with 1-acetoxy NO2DBA. This suggests that the markedly different biological activities of NDBA and NO2DBA might be due, in part, to a difference in their alkylating abilities in vivo.

[The effects of hyperoxemia on cerebral blood flow in subarachnoid hemorrhage patients].

The effects of oxygen inhalation at atmospheric pressure (1 ATA.O2) on CBF were studied in subarachnoid hemorrhage (SAH) patients due to ruptured intracranial aneurysms to prove the usefulness of the "O2 response" test for the evaluation of the cerebral vascular response. "O2 response" means the CBF (or ICP) decrease during hyperoxemia. CBF was measured by 10 m Ci 133Xe intravenous injection method using rCBF analyzer BI-1400 (Valmet). Two-compartmental analysis was used for the calculation of initial slope index (ISI). Studied cases were 53 postoperative SAH patients and 100 times examinations were done under the condition of Rest-1 ATA.O2. The incidence of global disturbance of the O2 response was 33% from day 0 to 3, 31% during day 4 to 7, 30% during day 8 to 14, and 23% at day 15 to 30. But, after day 31, there was no case which revealed an impaired O2 response. It was not possible to detect focal or hemispheric abnormalities of the O2 response because of the limitation of noninvasive two-dimensional CBF measurement method employed. The causes of O2 response abnormality, within a month after the onset, were increased ICP, hydrocephalus and diffuse brain damage. Multiple regression analysis proved that elevated PaO2 is the major factor in reducing CBF. CBF and ICP correlation study showed that the higher the Rest ICP, the fewer the CBF decreases during pure O2 inhalation. This may indicate that the increase in ICP in the acute stage changes the O2 response according to the level of increased ICP.(ABSTRACT TRUNCATED AT 250 WORDS)

Directly-acting mutagens formed from N-nitroso-N-(formylmethyl)alkylamines.

Directly-acting mutagens formed from N-nitroso-N-(formylmethyl)alkylamines (I) were isolated and identified as N-nitroso-N-alkyl-1-hydroxyimino-2-oxoethylamines (II). Their structures were elucidated on the basis of nuclear magnetic resonance spectra and confirmed by leading to their crystalline 2,4-dinitrophenylhydrazone. II (alkyl = ethyl and n-butyl) were strongly mutagenic to Salmonella typhimurium TA1535 and Escherichia coli WP2 hcr- without metabolic activation, while II with a tert-butyl group was not mutagenic. The formation of II from I is considered to proceed by the nitrosation of I, indicating a possible involvement of a formylmethyl metabolite in the carcinogenesis of nitrosamines with a 2-hydroxyethyl group.

Aging and urinary bladder carcinogenesis induced in rats by N-butyl-N-(4-hydroxybutyl)nitrosamine.

The effect of age on induction of carcinogenesis by N-butyl-N-(4-hydroxybutyl)nitrosamine [(BBN) CAS: 3817-11-6] in the urinary bladder epithelium was examined in 130 male and 130 female F344 rats. Rats of both sexes 6, 52, and 98 weeks old were given 0.025% BBN in their drinking water for 20 weeks. Then approximately half the rats were sacrificed, while the rest were maintained without further treatment for 10 weeks. Examination of the rats revealed an age-related increase in the induction of urinary bladder carcinoma, although the total intakes of BBN and urinary excretions of its proximate carcinogen were not age related. Rats treated with BBN at 98 weeks of age developed more squamous cell carcinomas and invasive carcinomas than the 2 younger groups. This study demonstrated an increased risk of urinary bladder carcinogenesis with age in animals.

[Reproducibility of somatosensory evoked potentials and EEG in normal humans under hyperbaric oxygenation].

Hyperbaric oxygenation (HBO) was reported to have a favorable influence on ischemic brain dysfunction, and also appeared to be useful in improving brain edema. Somatosensory evoked potentials (SEP) and EEG would be available as indicators of brain function. The purpose of this paper is to study reproducibility of the N1-amplitude of SEP and EEG under HBO. Materials were 10 normal volunteers, from 21 to 31 (mean age: 26) years old. Two stimulating needle electrodes were inserted into the skin at the wrist and somatosensory stimulations were applied to the median nerve with 1 msec duration square pulses. Stimulation pulses were generated regularly every 1 sec., and stimulus intensity was adjusted just above the thumb twitch threshold. Monopolar EEGs were recorded from 16 silver-cup electrodes on the scalp, and SEPs were recorded by averaging 250 responses in those EEGs using computer technique. N1-amplitude was determined by the vertical distance between peaks of the P1 and N1 components. EEGs were analyzed by Fast Fourier Transform and the square root of the averaged power of each frequency band (delta, theta, alpha 1, alpha 2, beta) was obtained and evaluated as the equivalent potential. EEGs under the influence of sleeping or artifacts, such as electromyograms, eye lid movements and wandering of the base line, were excluded in evaluating the EEGs. SEPs and EEGs recorded consecutively. The first record was taken before HBO under air breathing at 1ATA, the second during HBO under pure oxygen breathing at 2ATA, and the third after HBO under air breathing at 1ATA once again.(ABSTRACT TRUNCATED AT 250 WORDS)

[Effects of hyperoxia, glycerol and ventricular drainage on ICP and CBF in patients with increased ICP due to CSF circulatory-absorbance disturbance].

The effects of hyperoxia, glycerol and ventricular drainage on intracranial pressure (ICP) and cerebral blood flow (CBF) were studied in cases with cerebrospinal fluid (CSF) circulatory-absorbance disturbance due to subarachnoid hemorrhage (SAH) or intraventricular hematoma. Ventricular fluid pressure (VFP) was monitored through a controlled ventricular drainage (CVD) tube using a Statham P-50 pressure transducer. CBF was measured by the 10mCi133Xe intravenous injection method using a Valmet BI-1400 rCBF analyzer. ISI (initial slope index) was used for the CBF value. Studied cases were 19 SAH patients and 2 hypertensive intracerebral hematoma cases with intraventricular hematoma. Hyperoxia included hyperbaric oxygen (HBO), reduced ICP and CBF. During pure oxygen inhalation at atmospheric pressure (1ATA . O2), there was a tendency toward a relationship showing that the higher the resting state ICP, the lesser the likelihood of a CBF decrease. This may indicate that increased ICP impairs cerebral vascular reactivity to hyperoxia in relation to the degree of elevation. The effects of hyperoxia on ICP and CBF were temporary and they promptly returned to the resting state after cessation of oxygen inhalation. In some cases, there was a rebound phenomenon in ICP after HBO. Glycerol administration reduced ICP and increased CBF. There was no clear correlation between resting state ICP and CBF increase after glycerol administration. The effects of glycerol were also temporary. ICP control by opening CVD increased CBF. There was a correlation between the level of resting state ICP and the CBF increase after the opening of CVD. In cases with CSF circulatory-absorbance disturbance, elevated ICP reduced CBF and may further worsen the cerebral damage.(ABSTRACT TRUNCATED AT 250 WORDS)

Pancreatic pleural fistula: demonstration by computed tomography after endoscopic retrograde pancreatography.

A case of chronic pancreatic pleural effusions is reported. The effusions were massive in the right chest at first and became bilateral. Endoscopic retrograde pancreatography showed a pancreatic pleural fistula toward the diaphragm. Computed tomography after endoscopic retrograde pancreatography revealed the entire course of the fistula and a mediastinal pseudocyst. Computed tomography and the operative fistulogram did not show immediate leakage of the contrast medium from the mediastinal pseudocyst.

Urinary metabolites of N-nitrosodibutylamine in the rat: identification of N-acetyl-S-butyl-L-cysteine derivatives.

N-Acetyl-S-(butyl, 3-oxobutyl and 3-hydroxybutyl)-L-cysteines have been isolated and identified (as their methyl esters) from the urine of rats given N-nitrosodibutylamine (NDBA), N-nitrodibutylamine (NTDBA) and their corresponding alpha-acetoxy derivatives, N-nitroso-N-butyl(1-acetoxybutyl)amine and N-nitro-N-butyl(1-acetoxybutyl)amine, respectively. Greater amounts of these L-cysteine derivatives were detected in urine after administration of NDBA than of NTDBA. This suggests that the markedly different biological activities of NDBA and NTDBA might be due, in part, to a difference in their alkylating abilities in vivo.

Solution structure of [d(A-T)5]2 via complete relaxation matrix analysis of two-dimensional nuclear Overhauser effect spectra and molecular mechanics calculations: evidence for a hydration tunnel.

Pure absorption phase proton two-dimensional nuclear Overhauser effect (2D NOE) spectra at 500 MHz have been obtained for [d(5'ATATATATAT3')]2 in deuterium oxide solution at several mixing times. The 100 nonexchangeable proton resonances have been assigned. The experimental 2D NOE spectra were compared with theoretical spectra calculated by using the complete relaxation matrix analysis method [Keepers, J. W., & James, T. L. (1984) J. Magn. Reson. 57, 404-426] and x-ray diffraction determined molecular coordinates of A, B, alternating B, left-handed B, C, D, and wrinkled D forms of DNA and of energy-minimized structures calculated from the most promising X-ray crystal structures by using the molecular mechanics program AMBER, in which all hydrogens, counterions, and hydration water molecules were included. The analysis of all features of the 2D NOE spectra played an important role in extracting the promising structures, and it was concluded that the wrinkled D form yields the best fit for the 2D NOE data of the A-T decamer. The molecular mechanics calculation indicated that this model structure, whose minor groove is comparatively deep and narrow, may be energetically more stable than the B form for alternating d(A-T) DNA. Interesting features of the structure include possible intra- and interchain sugar-phosphate attractions and a hydration tunnel inside the minor groove capable of accommodating three types of water molecules that aid in helix stabilization via hydrogen bonding. Counterions (sodium) serve to reduce interchain phosphate-phosphate repulsive effects.

Tissue-specific transcription enhancement of the fibroin gene characterized by cell-free systems.

Six cell-free extracts have been used to characterize the nature of DNA signals and trans-acting factors responsible for the transcription enhancement of the Bombyx mori fibroin gene. The upstream element of the fibroin gene involved in the enhancement can be divided into two regions. The proximal region, -72 to -32, is recognized as a common enhancing signal by all B. mori extracts from the posterior silk gland, the middle silk gland, the ovarian tissue, and an embryonic cell line. It is weakly recognized by an Antheraea silkworm cell line extract but not by a HeLa cell extract. The distal region, -238 to -73, appears to be a tissue-specific enhancing signal that is recognized more effectively by the posterior silk gland extract than by the middle silk gland extract. These observations suggest that the use of these cell-free systems can offer a means for the biochemical characterization of the trans-acting factors involved in the tissue-specific regulation of the fibroin gene.
