Thromboxane B2 (TXB2) formation in clotting whole blood of healthy and diabetic humans in vitro.

We investigated the ability of platelets from two groups of diabetics type I and two groups of healthy volunteers matched of age to generate thromboxane B2 (TXB2) during spontaneous clotting of whole blood. The serum concentration of TXB2, reflecting the ability of the platelets to generate TXA2 during clotting, was measured by gas liquid chromatography. Platelets from old diabetics with more than 40 years duration of diabetes mellitus formed significantly less TXB2 than those from old healthy controls. Platelets from juvenile diabetics (9 years duration of disease) formed nearly the same amount of TXB2 as those from young healthy volunteers. The importance of these results is discussed.

[Sex-specific differences in food intake, body weight and parameters of lipid metabolism (HDL-cholesterol, total cholesterol and triglycerides) in rats under various feeding conditions].

We compared the food intake, body weight as well as several parameters of the lipid metabolism in male and female rats fed with a diet high in carbohydrate content (HCD) or high in fat content (HFD). The absolute food intake was higher in male than in female animals. With regard to the relative food intake (per 1 000 g body weight), the sex-specific differences could be demonstrated only in the HCD rats. It was striking that the HFD rats showed the same relative food intake in both sexes but that the females of this group exhibited significantly less weight gain. The HDL-cholesterol serum level was significantly higher in the HFD females than in the males. In the HCD group, we could not find any sex-specific differences. The total cholesterol serum level of female rats was significantly elevated in both diet groups, whereas the triglycerides serum level was only higher in the females of the HCD group, as compared to the corresponding levels of male rats. In the HFD group we could not find any sex-specific differences.

[Fatty acid patterns of triglycerides in serum and skeletal muscles of rabbits on a cholesterol diet].

In 18 rabbits a hypercholesterolaemia was produced by a 12-week dietetic load with cholesterol. The hypercholesterolaemic rabbits had increased proportions of saturated and simply unsaturated fatty acids in the triglycerides of the serum and the skeletal muscle. The content of eicosapentaenic acid was lowered. The alterations in the fatty acid pattern are equal to those of patients with hyperlipoproteinaemia and arteriosclerosis. On the basis of these findings rabbits seems to be a suitable model, in order to investigate the connections between fatty acid patterns and the development of arteriosclerosis by experiments on animals.

[Surgical reduction of the kidney for inducing chronic kidney insufficiency].

By operative reduction of the renal tissue on 1/5 of both kidneys there are to be produced a chronic renal insufficiency in 21 black mini-pigs. The operative procedure was carried out at one time (nephrectomy and resection of the residual kidney) or at two times (in one group of it at first the nephrectomy and in the other group at first the resection of the kidney). Only 50% of the animals died with the symptom uremia in renal insufficiency. The nephrectomy--with a temporarily shifted resection of the residual kidney--effects the desired changes most approximately with very different reaction. This procedure appears as an animal model for the chronic renal insufficiency altogether not suitable.

Lysozyme (muramidase) and hemolytic complement (CH50U) in sera of beagle dogs and minipigs after longterm application of different preparations of estrogens and gestagens.

In studies for the chronic toxicity of estrogens and gestagens, lysozyme and hemolytic complement (CH50U) in sera of beagle dogs and minipigs were measured. Various doses of the sexsteroids were given: 0.01 mg, 0.1 mg and 1.0 mg/kg respectively. The influence of the steroids on the tested parameters after an application for 6 months was not significant, only in a single series with unphysiologically, high dosages of the preparations were some values changed significantly.

[Effect of long-term application of SUISYNCHRON-premix on swine].

Two groups of five female store pigs, weighing an average of 60 kg at the start of the experiment, were given either 1 or 10 mg metallibure zinc complex (SUISYNCHRON) per kg body weight once daily for six months, in the form of a 2% talcum premix added to concentrates. Clinical inspection, weight gain, haematological findings, blood sugar, serum transaminases, properties of urine and faeces, carcass examination and histological study (internal organs, endocrine glands, skeletal muscle) showed that the 1 mg/kg dosage had no toxic effect. The 10 mg/kg dosage resulted in considerable depression in appetite and some apathy in the pigs, but there was no evidence of a toxic effect.

[Clinical data on the use of estriol-testosterone combinations in small animal practice].

A combination of oestriol and testosterone oenanthate was tried in 14 different indications on 110 dogs and 5 cats of both sexes. Two formulations were used, containing in 1 ml either 0.15 mg oestriol and 2.25 mg testosterone or 1.15 mg of the former and 2.25 mg of the latter. Dosage was 0.2 ml per kg body weight, by i/m injection. The criterion for success was the degree of functional restitution, without taking into account pathological changes, which were generally unaffected. Good or very good results were obtained in hip dysplasia, arthitis and patellar luxation. None of the 50 bitches treated showed signs of oestrus or irregularities in the cycle. The formulation containing the lower drug concentration seemed to give the best therapeutic results.
