Human nephrectomy specimens with ischemic glomerular changes were studied by colloidal iron staining and immunofluorescence with respect to glomerular polyanion, immunoprotein deposition and proteinase inhibitor deposition. In the sclerotic area of ischemic glomerular changes, polyanion loss and deposition of IgM, Clq, C4 and C3 were observed. These findings were common in cases with and without chronic pyelonephritis showing a considerable similarity to the glomerular changes of experimental focal sclerosis in rats or focal sclerosing glomerulonephropathy in human. alpha 1-Antitrypsin and alpha 2-macroglobulin (proteinase inhibitors) were also found in the sclerotic area. In particular, alpha 1-antitrypsin was always present in the sclerotic portions of every atrophic nephron and an important role of alpha 1-antitrypsin in atrophy of nephrons was suggested.

The nuclear areas, perimeter, diameter and form factor, and nucleolar area and diameter of about 50 cells in each in 38 colonic epithelial lesions including normal epithelium (6), Peutz-Jeghers polyp (2), adenoma (11), dysplasia (6) and carcinoma (13) were measured at the electron microscopic level by using an image analyzer. The area of nucleoli was larger in dysplasia and carcinoma than the normal (statistically significant). The area of nuclei was larger in carcinoma than the normal. The form factor was also largest in the nuclei of carcinoma, indicating that these cells had the most spherical shape. These results suggest the possibility of quantitative diagnosis of epithelial lesions by measuring the area and perimeter of some 50 nuclei and nucleoli.

As of March 1978, the prognosis was confirmed in 715 non-Hodgkin's lymphoma cases reported during the 12-year period from 1965 through 1976 in the Prefecture of Kagoshima. Included were 222 cases of extranodal lymphoma. Patients with the initial tumor site in the gastrointestinal tract and Waldeyer's ring had a relatively good probability of survival. Among the patients with cutaneous lymphomas, those with the mycosis fungoides type had a poor survival rate. In Kagoshima Prefecture, the survival curve for cases of nodal lymphomas is characteristically much poorer than those reported in the medical literature. Of the nodal lymphomas, the nodular type was found in 128 cases and the five-year survival rate was 15%, while the diffuse type accounted for 365 cases and the five-year survival rate was 8%. The paracortex type, considered to be of T-cell origin, accounted for 55 cases and the five-year survival rate was 0%. These differences were statistically significant. Based on our data, the classification of nodular lymphomas into subtypes according to the degree of nodularity was found to be useful since it was related to clinical stage and survival. Rappaport's cellular classification of diffuse lymphoma is related to prognosis and was found to be useful, but problems still remain in determining T- and B-cell types by means of cellular morphology and thus further study is warranted.
An inhibin-like substance, a follicle-stimulating hormone (FSH) inhibitor, was isolated from porcine follicular fluid and purified. Serum FSH levels of unilaterally ovariectomized mice which received injections of the inhibitor were lower than those of unilaterally ovariectomized mice which received injections of saline. The inhibitor also suppressed FSH binding to granulosa cells in vitro. These results suggest that this protein is an FSH inhibitor and that it has two modes of action: suppression of FSH levels in serum and suppression of FSH binding to granulosa cells.

The glycosaminoglycan (GAG) content of stomach carcinoma tissue was compared with that of non-neoplastic mucosa. GAG synthesis was also studied, by an analysis of 35S-labelled material after incubation of tissue segments in medium containing 35SO4. No significant difference was found between the amount of GAG and its components in the medullary carcinoma tissue and in non-neoplastic mucosa, but GAG synthesis of the carcinoma tissue was at a much higher rate than that of the non-neoplastic mucosa. In the autoradiograph, high 35S uptake in the carcinoma cells was observed. The GAG content of the scirrhous-carcinoma tissue was about twice that of medullary carcinoma.

Two cases of so-called multilocular cyst of the kidney are presented. Although both cases satisfied all of the criteria which characterize the multilocular cyst of the kidney, one had cystic lesions and neoplastic lesions (nephroblastoma-like lesions) and the other had only cystic lesions and was complicated with hamartoma. We prefer the term "cystic partially differentiated nephroblastoma" as the diagnostic term for the former and "multilocular cyst of the kidney" for the latter. A study of 40 reported cases of multilocular cystic lesions of the kidney revealed that cases having only cystic lesions were distributed in all ages from 4.5 months to 71 years and that cases having neoplastic lesions were seen in infants from 4 months to 2 years.

We developed a new beta-adrenergic blocking, antiarrhythmic compound S-596 and compared the findings with those of propranolol or practolol. S-596 antagonized the positive chronotropic and inotropic actions of adrenaline in isolated guinea-pig atria, and blocked the relaxant response to adrenaline of isolated guinea pig tracheal strips. In anesthetized mongrel dogs, S-596 given intravenously inhibited increases in heart rate and myocardial contractile force and decreases in systemic blood pressure induced by isoproterenol. In conscious dogs, the oral administration of S-596 reduced the isoproterenol induced tachycardia and the maximal effect was attained one hour after administration. In this regard S-596 was about 5 times more potent than propranolol, and S-596 was significantly longer-acting than propranolol. Thus, S-596 has greater beta-blocking activity than propranolol. S-596 has a lesser degree of myocardial depressant action than propranolol in spontaneously contracting rat or guinea pig atria and no intrinsic sympathomimetic activity in reserpinized rats. S-596 also has a weaker local anesthetic and antiarrhythmic activity than propranolol, as determined in guinea pigs and rabbits.
In order to grade objectively and characterize dysplastic and precancerous esophageal epithelium its DNA content was measured by cytofluormetric methods and compared to normal and cancerous esophageal epithelium. This yielded the following results. With transition of the esophageal eipthelium from mild dysplasia to severe dysplasia and finally to in situ carcinoma, Feulgen-DNA values showed patterns characteristic of a tetraploid population. They lacked prominent peaks which were usually observed with invasive carcinomas. Dominant near-tetraploid population and definite tetraploid-octoploid populations were characteristic of severe dysplasia or carcinoma. The mean Feulgen-DNA values were significantly larger in severe dysplasia than in the lesser grade of dysplasia as well as the normal epithelium. However, this was not the rule in the full blown carcinomas. It would appear that the esophageal cytophotometric patterns are analogous to those previously observed in the skin and uterine cervix.
An analysis was carried out on glycosaminoglycan produced in pleomorphic adenoma, adenoid cystic carcinoma, sialadenitis and normal tissue of the salivary gland. After incubation of the tissue segments in a medium containing 35SO4, a radioautograph of the tissue section was made to observe the localization of 35SO4 incorporation, and 35S-labelled materials were purified from the tissues, and analyzed. High 35S-radioactivity was observed in the ductal cells of the inflammatory gland tissue and in the acinar cells of normal palatinal gland, but little radioactivity was observed in the interstitial components in these tissues, and the amount of 35SO4 incorporated in the tumor cells was also significant. Eighty to 90% of the 35S-radioactivity incorporated could be detected as 35S-glycosaminoglycans in all tissues except for the normal palatinal gland, which contained a large amount of 35S-sulfated glycoprotein. No significant difference in the synthetic activity of 35S-glycosaminoglycans and in their components were observed between nonneoplastic and neoplastic cells. These results suggest that glycosaminoglycan-producing cells in pleomorphic adenoma as well as in adenoid cystic carcinoma are derived from the tubuloacinar cells of the salivary gland.
