Nafazatrom (Bay g 6575) has been shown to be a potent inhibitor of tumor metastasis in preclinical models. It is believed to work by stimulating endogenous prostacyclin production. The drug has also been shown to inhibit the growth of certain experimental tumors, to be cytostatic for certain cell lines in tissue culture, and to induce differentiation in HL-60, neuroblastoma, and Friend erythroleukemia cell lines. Furthermore, no toxicity has been seen in animals or human volunteers. We report here a clinical trial of oral nafazatrom at five dose levels in patients with advanced cancer. Thirty patients with a wide variety of advanced malignancies were treated for 26-638+ days (median 82 days). No tumor responses were seen. Toxicity included two cases with mild skin rashes, one case with nausea and vomiting, and one case with diarrhea. Nafazatrom is a safe and well-tolerated agent. Maximum activity would be predicted to occur in the adjuvant treatment of cancer and we feel that further efforts should proceed to identify the appropriate dose for such a trial.
There is increased emphasis nowadays on the role of the general practitioner in preventive care for children. Accordingly, an analysis of the child's perception of the doctor and of the factors which influence and mould that perception are important. In New Zealand children's paintings have been used for such analyses. The present study looked at children's paintings of their family doctors in a Devonshire village. The findings show that boys are more likely than girls to depict their doctors as a gruesome or threatening character, and suggest that medical contact has little, if any, influence on character portrayal of doctors. Other factors that might influence character portrayal and are now thought worthy of detailed study, include television viewing patterns, personality, age, home background and artistic ability.

Nafazatrom is rapidly and almost completely absorbed after oral administration. However, the plasma levels of unchanged nafazatrom are very low, suggesting an extensive biotransformation during a first passage through the liver. The concentrations of nafazatrom in the plasma therefore, may only reflect indirectly the effective concentration at the receptor site. Concentrations, half-life and distribution of nafazatrom between aqueous and liquid compartments suggest that the cellular membrane may be the site of action.
A double-blind study was conducted in 60 female patients with moderate or severe insomnia, hospitalized for gynaecological surgery. After an initial 2-day placebo selection phase, 30 subjects received 15 mg midazolam and the remaining 30 received 1 tablet Vesparax (= 50 mg hydroxyzine, 150 mg secobarbital, 50 mg brallobarbital) for 5 nights. This verum phase was immediately followed by a 2-day placebo withdrawal phase in order to study the occurrence of rebound phenomena. Both verum compounds were effective in hastening sleep onset, increasing sleep duration, and improving sleep quality, without causing residual effects on the following day. There was no difference in effect between the two agents. Neither active drug caused rebound effects on withdrawal.

Four morphologically measurable conditions of epithelial cells proved safely recordable by electron-microscopic examinations of the human hydrosalpinx: 1. Epithelium largely normal, with clearly differentiated cells; 2. Epithelial areas with necrobiosis; 3. Regeneration of epithelial complexes; 4. Indifferent regeneration epithelium without definitely indentifiable function of the individual cell. -- Results so far obtained are likely to suggest that functional restoration of partially damaged epithelium of the fallopian tube can be expected, owing to the established regenerative potential of epithelial cells, once pressure has been removed from the hydrosalpinx.
The granulosa cell tumour is of epitheloid structure. Two characteristic cell types can be distinguished. One of them appears "brighter" in electron-optical investigation and may have originated from theca formations. These is a highly developed variety of organelles, including endoplasmatic reticulum, mitochondria, and ribosomes, which is just a strong in synthesis as steroidogenic cells. The existence of an intercellular drainage system may be presumed, in similarity to corpus luteum, to carry steroids from cell to blood capillary. Amitotic cell division has been noticed as a peculiarity of reduplication of tumour parenchyma.
1. Thirty pregnant women aged 22-34 yr presenting with pre-eclampsia were treated with guanfacine for periods of 16-68 days. 2. Elevated blood pressure was lowered to a statistically and clinically significantly extent in the first week of treatment, falling almost to normal levels before parturition in 24 cases. Four days after delivery all the patients had normal blood pressures. 3. Oedema (localized or generalized) was not reduced to any extent by guanfacine and additional treatment was necessary in 17 cases. There was little effect on proteinuria. 4. Twenty-five patients reported side-effects attributable to the treatment (sedation in 25 cases, dryness of the mouth in 15, and feelings of dizziness in 6 cases). 5. No notable changes were observed in either foetal or maternal heart rate during the period of treatment. Of the 30 deliveries, 19 were spontaneous, 5 by vacuum extraction, and 6 were by Caesarean section because of suspected placental insufficiency. Six of the infants were 'small-for-date babies', but all developed normally later.
