Antibodies eluted from lymphoid cell membrane. Occurrence in certain varieties of scleroderma.

By means of acid elution two antibodies could be removed successfully from the circulating lymphocytes of 11 patients with certain varieties of scleroderma. One was specifically directed against nuclear antigen(s) of endothelial cells (NEC) of the dermal blood vessels, and another against nuclear antigen(s) of epidermal basal cells (NBC) of the involved and uninvolved skin of the patients. In two cases of acroscleroderma, the eluates failed to react with either endothelial or basal cells of involved or uninvolved skin. In none of 20 healthy controls involved in this study could an antibody be eluted from the circulating lymphocytes. The aforementioned antibodies do not bind complement in vitro and do occur in the serum of four patients. Circulating antinuclear antibody (speckled type) was detectable in two cases of scleroderma.

Buckley's syndrome.

An infant girl with chronic eczema, recurrent infections, elevated IgE and impaired neutrophil chemotaxis appeared to belong to the group of patients described by Buckley, Wray & Belmaker (1972). An hereditary influence is suggested by a similar defect found in the patient's mother. Levamisole improved the in vitro defect and the clinical picture.

Evaluation of the local antihistaminic activity of dimetindene maleate by a quantitative method.

The topical application of a 0.1% dimetindene maleate gel on the forearms of 11 subjects reduces the areas of both erythema and wheal produced by the intradermal injection of 0.025 mg of histamine hydrochloride to a statistically greater degree than 1% d-chlorpheniramine maleate cream (p less than 0.005 and p less than 0.0005, respectively). The quantitative method employed in this study has proved to be an excellent means for the evaluation of the antihistaminic activity of topical preparations.

Identification of sequence similarities between Mycobacterium leprae and the myelin proteolipid protein by computational analysis.

This study was undertaken under the assumption that antigenic mimicry plays a role in the pathogenesis of neuropathy in leprosy, a unique feature among mycobacterial diseases. The SWISS-PROT protein sequence databank was scanned using a computer programme based on an identity matrix algorithm, to identify common amino acid regions between human myelin and mycobacterial proteins. The highlighted motifs were back-tested against a database of MHC-binding peptides (MHCPEP). Of the 28 common sequences between mycobacterial and human myelin proteins, only two were found to yield some matches with MHC-presenting peptides. Both motifs were from M.leprae. The myelin proteolipid protein was the human protein containing the identified similarities. We believe that this theoretical approach can provide a way to predict potentially "mimetic" motifs by search for antigenic regions in protein sequence databases without screening a large number of synthetic peptides.

[Anti-cardiolipin antibodies in lupus erythematosus].

Anti-cardiolipin antibodies (ACA) were investigated in the serum of 79 patients with lupus erythematosus by means of ELISA method. 34 patients (43%) had ACA in their serum. 63.6% of the patients with ACA had 4 or more ARA criteria and 75% of them had malar rash. None of our patients had the "anti-cardiolipin antibodies syndrome" but three of them with thrombocytopenia had ACA.

[Behavior of plasma androgens in cases of adrenal pathology].

Following chromatographic separation, the RI method has been used to examine the behaviour of plasmatic androgens in a group of normal subjects, in two patients with Addison's disease and two patients with hypercorticoadrenalism, due respectively to corticoadrenal adenoma and bilateral adenomatous hyperplasia. A significant reduction in adrenal androgens was observed in patients suffering from Addison's disease. The most interesting information, however, was the paradoxical reduction in DHEA and delta 4-A in patients with corticoadrenal hyperfunction.
