The ADH-reserve capacity in Brattleboro rats.

The water metabolism was studied in homo- and heterozygous Brattleboro rats suffering from hereditary hypothalamic diabetes insipidus. In homozygous Brattleboro rats the spontaneous water intake and urinary output and the diuretic reactions signficantly increased after water and salt loading. No antidiuretic activity was found in the urine, posterior pituitary or hypothalamus of these animals, and this state was not affected by hyperosmosis. For the heterozygous rats the spontaneous water intake and urinary output and the diuretic reaction exceed the respective control values, the posterior pituitary, the hypothalamus and the urine are of reduced antidiuretic activity and this activity is less mobilizable by hyperosmosis. It is concluded that the ADH-reserve deficiency is total in the homozygous Brattleboro rats, and partial in the heterozygotes. As a result of hyperosmosis, the vasopressin release is of a reduced extent, yet detectable in the heterozygotes.

Lymphocytotoxic antibodies in lightdermatoses.

Occurrence of lymphocytotoxic antibodies of the "cold type" was studied in 92 cases with various lightdermatoses. They were detected in the sera of 39% of the patients with cutaneous porphyrias and never in cases with polymorphous light eruption and other photodermatosis. Correlations between their presence and the duration of the porphyria as well as the severity of hepatopathy could be observed. The results indicate the importance of a long-standing tissue damage in the production of these antibodies. In addition the findings confirm the hypothesis according to which polymorphous light eruption does not belong to the lupus erythematosus entity.

Adenohypophysial necrosis in rats following destruction of the pituitary stalk. A histologic, immunocytologic and fine structural study.

Adenohypophyses of adult male rats have been investigated by light microscopy, immunocytology and electron microscopy 30 minutes, 1, 2, 4, 6 and 24 hours following electrolyte destruction of the pituitary stalk. Light microscopy revealed massive ischemic infarction of the adenohypophysis. Immunoreactive growth hormone, prolactin, TSH, FSH and LH were demonstrated up to 24 hours after surgery in necrotic adenohypophysial cells by the immunoperoxidase technique. Fine structural abnormalities were already noticeable 30 minutes following stalk lesion. The changes rapidly progressed and within 2 hours unmistakable signs of cellular necrosis became evident. Mitochondria, especially those of growth hormone cells, seemed to be affected at an early stage, whereas alterations in rough-surfaced endoplasmic reticulum membranes developed later. Nuclear changes, formation of cytoplasmic vacuoles and disruption of cell membranes were conspicuous findings. No evidence, indicating the participation of lysosomes in the development of cellular damage, was obtained. Secretory granules were prominent and well preserved even in cells which showed advanced necrosis. The failure of discharge and degradation of secretory granules in the necrotic cells suggest that factors accounting for their intracellular migration and extrusion are very sensitive to ischemia and are paralyzed at an early phase.

[Fracture-dislocation of the hip joint. II. Central luxation].

64 cases of central luxation are analysed by the authors. 4--10 years after the injury 31 patients appeared at the check-inspection. It has been stated by the authors that because of the severity of the injury and the age of the injured patients surgical intervention may be performed even in case of greater dislocation but rarely. The result of the conservative treatment is in the case of congruency of the loading surface satisfactory, but it is even in the case of incongruency generally better than it may be supposed on the basis of the roentgenogram. For the prevention of later occurring severe arthrosis the importance of the rehabilitation is emphasized by the authors.

[Diagnosis and treatment of postoperative infections following hip injuries].

The authors studied the diagnosis and treatment of posttraumatic infections of the hip joint, based on their own material. In case of latent infection the increased sedimentation rate and taking of comparative X-ray films are held important, that shows a narrowing and disappearance of the sclerosed edge of the acetabulum. Following dislocation of the hip joint ankylosis may be attained, but after fractures of the proximal and of the femur the infection cannot be cured, unless the nail and the head of the femur are removed.

Virus-like structures in lupus erythematosus discoides.

Biopsy specimens from typical skin lesions of DLE patients were studied by means of electron microscopy. Virus-like structures were discovered in the cytoplasm of fibrocytes, endothelial cells and keratinocytes. After prolonged chloroquine treatment a decrease in the frequency and size of these structures was observed.

Mechanism of chlorpropamide antidiuresis.

A study was made of the effect of chlorpropamide in rats with pituitary stalk lesion. It was found that the drug made the oliguric interphase more pronounced and protracted, while in the lasting polyuric phase in the absence of endogenous ADH it influenced water intake only moderately. Enhancement of the effect of exogenous ADH was observed after a 2-day chlorpropamide pretreatment. The results support the view that the presence of a small amount of endogenous ADH is necessary for chlorpropamide-antidiuresis. The drug presumably brings about the decrease in diuresis via ADH mobilization, but its peripheral effect is also enhanced.

Characteristics of phage AP50, an RNA phage containing phospholipids.

A bacteriophage specific for Bacillus anthracis was isolated and designated as AP50. The nucleic acid of phage AP50 is RNA and the virion contains five different phospholipids. Some physical and biological characteristics of the phage, including morphology, were examined. To the best of our knowledge, this RNA bacteriophage containing phospholipids is the first to be isolated for a Gram-positive host.

Study of the antidiuretic effect of clofibrate in rat.

The effect of clofibrate on the water metabolism was studied in rats with pituitary stalk lesion. It was found that this drug made the oliguria interphase much more marked and more prolonged. In rats anaesthetized with alcohol, clofibrate pre-treatment led to an enhancement of the effect of ADH. The experiments permit the conclusion that the presence of endogenous ADH is necessary for clofibrate antidiuresis. Its effect is presumably exerted via the mobilization of the ADH, but it can not be excluede that it also enhances the peripheral effect of ADH.

Studies on T and B lymphocytes in the peripheral blood of discoid lupus erythematosus patients with and without chloroquine treatment.

The proportions of T-B cells of peripheral lymphocytes in DLE patients as well as the effect of chloroquine treatment on T cell count were studied. In DLE, both the active and the total rosette-forming cell counts decreased, whereas the B cell count did not differ from normal values. Long-term chloroquine administration causes a further reduction in T cell count.

Studies on megacinogeny in Bacillus cereus. II. Bacillus cereus isolates characterized by prophage-controlled production of megacin A (phospholipase A).

Five out of a number of Bacillus cereus strains isolated from soil produced high titre specific bacteriocin (megacin A) in mitomycin C-induced cultures. In the course of cultivation with ethidium bromide, the strains gave off segregants not producing bacteriocin (cin-). The lysate of two wild strains formed plaques on the corresponding cin- bacteria. The two phages (wx23 and wx26) were identical in antigenic structure with phage wx was present in the lysate of B. cereus strain W, and converted cin- derivatives into cultures producing megacin A (phospholipase A). The phages produced plaques at 26 degrees C but not at 37 degrees C. In the lysates of the remaining three strains phages were not detected with biological and morphological methods; these cultures have been assumed to carry defective prophage genome. As the corresponding prophages are responsible for the determination of inducible phospholipase A production, phages named wx seem to form a separate group of B. cereus phages.
