A morphological study of membrane lesions during natural killer-mediated lysis.

Early and late events occurring during cell-mediated cytolysis have been examined ultrastructurally using transmission, scanning and freeze-fracture electron microscopy. K562 erythroleukemic cells were used as target for human natural killer lymphocytes, a subpopulation involved in host defense against tumor, virus infected and allogeneic cells. Cell binding between target and effector is already evident after 15 min, but membrane lesions and target hydration appear after 30 min incubation. At 60 min K562 cells appear swollen and microvilli-deprived. Progressive membrane disruption is also evident and the underlying cytoplasmic material is clearly exposed after 120 min. Submicroscopic membrane lesions appear to be early events in comparison with 51Cr (isotope) release from damage cells.

Increased number of circulating Leu 11+ (CD 16) large granular lymphocytes and decreased NK activity during human ageing.

The phenotype and functional activity of circulating T and natural killer (NK) cells was investigated in a group of selected normal elderly donors in comparison with a group of normal young individuals. Old subjects exhibit a decline in circulating T cells and an impaired response to mitogens (Con A, PHA) associated with a relative increase in cells reacting with Leu 7 and Leu 11a monoclonal antibodies, directed against large granular lymphocytes with NK activity. The rise in circulating NK cells observed in the elderly was not associated with a concomitant increase in NK cytolytic activity against K 562 tumour cells, and time course kinetics of the cytotoxic reaction was similar to that observed in young subjects. When a greater than 95% pure population of Leu 11a+ cells from old individuals was sorted by flow cytometry and their functional cytotoxic activity examined, a significant decrease in NK cytotoxicity was detected. The target cell binding capacity of effector cells, morphologically evaluated at single cell level, did not differ in old and young subjects, even though bound cells from young donors have a higher lytic capacity. These data show that an increase in circulating NK granular lymphocytes occurs during human ageing, but suggest that in old subjects only a subset of NK cells is active in order to maintain a functional response similar to that observed in the young.

The long-term course of primary generalized epilepsy with persisting absences.

The long-term course of primary generalized epilepsy with absences persisting after the age of 30 to 61 years was studied in 26 patients, each followed clinically and by EEG for 20 to 37 years. Whether treated or not, absences became less frequent, but only rarely ceased, and 92% of patients with persisting absences eventually developed generalized convulsions. EEG background activity did not deteriorate, and the classic spike and wave complexes remained typical in 84% of patients. In 36% of cases, almost all of them women, psychomotor slowing was observed, which did not affect job performance. It was not related solely to duration or severity of epilepsy, or to antiepileptic drugs, and some hormonal mechanism may be partly responsible.

[Extraordinary variant of the alpha rhythm].

The authors describe a variant of alpha rhythm, which is remarkable because of its great amplitude (200-300 microV). It is observed quite exceptionally in children and adolescents with a severe encephalopathy of either ante- or perinatal etiology. They suggest that the surplus amplitude of this alpha rhythm could depend upon a deafferentation of the occipital cortex in patients with severe amblyopia.

Distribution of acid alpha-naphthyl acetate esterase among human T lymphocyte subsets.

Human T lymphocyte subsets, identified by means of OKT3, 4 and 8 monoclonal antibodies, were isolated by a fluorescence activated cell sorter (FACS IV) and analyzed for distribution of alpha-naphthyl acetate esterase (ANAE) activity. As compared to OKT8+ lymphocytes a higher proportion of OKT4+ lymphocytes was ANAE-positive exhibiting a spot or dot-like pattern in the cytoplasm. OKT8 and 4 positive subsets showed a similar ANAE distribution in diffuse granular form. Although OKT4 and OKT8 populations presented a different ANAE dot-like reactivity, this marker did not allow as clear a distinction between them as that reported for TG and TM lymphocytes.

Analysis with OKT monoclonal antibodies of T-lymphocyte subsets present in blood and liver of patients with chronic active hepatitis.

The relative distribution of T lymphocyte subsets, as defined by the monoclonal antibodies OKT, was determined by cytofluorimetric analysis in peripheral blood and in cells isolated from liver biopsies of 31 patients with chronic active hepatitis (CAH). The percentage of peripheral blood lymphocytes binding OKT8 (directed against cytotoxic/suppressor T cells) was found to be elevated in patients with HBsAg and HBeAg positive chronic active hepatitis. Patients with CAH who had seroconverted to anti-HBe, had an increased number of OKT3-positive cells in their blood, which was directed against a common T cell surface antigen, associated with a decreased number of OKT8 positive cells. Lymphocytes isolated from liver biopsies of patients with CAH presented a general increase of OKT8-positive cells associated with a decreased number of OKT4-positive (helper/inducer) T cells. It is likely that OKT8-positive cells found in liver biopsies represent cytotoxic T cells directed against either viral or liver cell determinants.
