Hemocyanins in spiders, XVII. A presumptive active-site sequence of arthropod hemocyanins.

Peptides containing the sequence -His-His-Trp-His-Trp-His- have been isolated from the subunits e and a of tarantula (Eurypelma californicum) hemocyanin and III B of Limulus polyphemus hemocyanin. In view of the published spectroscopic and titration data on the copper-polypeptide complex and the strong quenching effect of oxygenation on hemocyanin fluorescence, it is concluded that this sequence represents the major part of the active site of Eurypelma, Limulus, and possibly other arthropod hemocyanins.

Structure of the linkage-region between polysaccharide chain and core protein in bovine corneal proteokeratan sulfate.

Peptidokeratan sulfate from bovine cornea was degraded by a combination of desulfation, exo-enzymic digestion and finally digestion with endo-beta-N-acetylglucosaminidase D. The same procedure was carried out both with [3H]fucose-labelled and [3H]mannose-labelled peptidokeratan sulfate. Data obtained by methylation analysis of peptidokeratan at the different degradation steps, as well as action of endo-beta-N-acetylglucosaminidase D, showed that the binding-region in proteokeratan sulfate from bovine cornea is identical with a structure found in various GlcNAc(beta 1-N)-Asn-linked mannosyl glycoproteins. The existence of a chitobiose unit between asparagine and mannose was proved by action of endo-beta-N-acetylglucosaminidase D. The existence and position of an (alpha 1 leads to 6)-linked fucosyl residue at the Asn-bound GlcNAc was demonstrated by action of alpha-fucosidase, endo-beta-N-acetylglucosaminidase D and by gel chromatography on Bio-Gel P-4. By gas chromatography/mass spectrometry studies, the existence of a 1,4,6-trisubstituted beside a 1,4-disubstituted GlcNAc in the binding-region oligosaccharide was shown. Other results reported here are according to analytical data previously published (Keller, R., Stein, T., Stuhlsatz, H.W., Greiling, H., Ohst, E., Müller, E. & Scharf, H.-D. (1981) Hoppe-Seyler's Z. Physiol. Chem. 362, 327-336).

Treatment of human echinococcosis with mebendazole. Preliminary observations in 28 patients.

Preliminary results of a long-term, prospective therapeutic trial with mebendazole in 28 patients mainly with inoperable echinococcosis are reported. The course of disease was monitored closely and plasma mebendazole levels were checked regularly. A major problem is the lack of reliable methods for defining and measuring early success or failure of therapy. Most of the patients improved clinically. No marked change of parasitic lesions was observed except in one patient with constantly low plasma mebendazole levels who deteriorated. No serious side effects occurred. The preliminary results are encouraging and suggest that mebendazole has a "parasitostatic" effect on larval growth.

Sodium flux and lipid spectrum in the erythrocyte membrane in essential hypertension.

In the erythrocyte membranes of normotensive individuals and patients with essential hypertension, sodium flux behavior, cholesterol content, phospholipid spectrum, and the fatty acid composition of the phospholipids were measured. There was no difference between normotensives and hypertensives in sodium influx in the erythrocyte. In both groups, 1 microM norepinephrine resulted in a significantly equal increase of sodium influx after an incubation period of 15 min. Compared with normotensives, erythrocytes in essential hypertensive patients showed a decreased sodium efflux. The cholesterol/phospholipid quotient of the erythrocyte membrane of hypertensive subjects increased in comparison with that of normotensive individuals. No differences between the two groups were found in the phospholipid pattern and in the fatty acid composition of phosphatidylserine/-inositol, -choline, and sphingomyelin. However, in phosphatidylethanolamine of the erythrocyte membrane in essential hypertensives in contrast to normals, a decreased content of the fatty acid fraction 22:0/20:3 and in increased content of the fatty acid fraction of 20:5/22:2/22:3 was found. The dependence of the Na turnover on the lipid spectrum of the erythrocyte membrane is discussed. The altered lipid composition of the erythrocyte membrane in essential hypertensives does not seem to cause the altered Na efflux behavior.

[Complex airway resistance as a means of characterizing the strength of asthmatic reaction in the guinea pig].

45 guinea pigs sensitized to ovalbumin and 9 controls were challenged with antigenic aerosol. Before and during the challenge oesophageal pressure (Poes), airflow (V) and tidal volume (VT) were measured in anaesthetized (urethane) spontaneously breathing animals. Inspiratory and expiratory lung resistance (RI, RE) and dynamic lung compliance (Cdyn) were obtained according to the method of Neergard and Wirz. Total pulmonary resistance was calculated by different methods. 18 animals responded with a significant asthmatic reaction with an increase of expiratory resistance and a decrease of dynamic compliance. There exists a close hyperbolic relation between RE and Cdyn before and during the asthmatic reaction. The best parameter to characterize the strength of asthmatic response in the anaesthetized guinea pig is delta Poes/VT.

Enzyme-histochemistry of the juxtaoral organ in man ("organ of Chievitz").

The activities of several enzymes - hydrolases, oxidoreductases and carbonic anhydrase - were demonstrated histochemically in the epithelial parenchyma of the human juxtaoral organ. Two characteristics enzyme-activity-patterns provided morphological distinction between two different forms of the juxtaoral organ, independently of the sex or age of the patient. Provisionally we have called them type I and type II. Type I showed a strong activity of alkaline phosphatase and carbonic anhydrase and low activity of non-specific esterases, whereas type II showed just the contrary. The enzyme activities in the epithelial parenchyma displayed obvious similarities to those of the duct cells of salivary glands but they were different from those of the oral mucosa studied. Only alkaline phosphatase of the enzymes demonstrated, showed activity in the epithelia of the juxtaoral organ but none in the oral mucosa or salivary glands. On account of the multiple nerve endings present, a receptor function is presumed for these, whereas the function of the epithelial parenchyma is still unknown.

[The Meckel syndrome (author's transl)].

The Meckel syndrome is caused by an autosomal recessive gene. The three main features are encephalocele, polycystic kidneys and polydactyly. At least two of these should be present to establish the diagnosis. Several minor symptoms are observed in various combinations. The possibility of prenatal diagnosis is explained by a case report: A neural tube defect was diagnosed by ultrasonography and AFP-assessment in amniotic fluid whereas unfortunately the presence of polycystic kidneys was not recognized sonographically. The methods of today are able to detect the polydactyly but failed in this case. One can suggest that the complete prenatal diagnosis of the Meckel syndrome is not essential for clinical management since the presence of one major symptom will lead to legal abortion. The search for Meckel syndrome is important in cases of delivery of a child with multiple malformations, indicating a genetic counseling for each subsequent pregnancy.

Formation of N,N-Dimethylglycine, Acetic Acid, and Butyric Acid from Betaine by Eubacterium limosum.

Two bacterial strains that grow anaerobically on betaine were isolated from enrichment cultures and identified as strains of Eubacterium limosum. In a mineral medium supplemented with yeast extract and Casitone, the doubling time of E. limosum strain 11A on betaine was 6 h at 37 degrees C. The molar growth yield amounted to 9 g of dry cell mass per mol. Betaine was fermented in accordance with the following equation: 7 betaine + 2 CO(2) --> 7 N,N-dimethylglycine + 1.5 acetate + 1.5 butyrate. E. limosum also grew on methanol and choline. The former was converted to acetate and butyrate, and the latter was converted to N,N-dimethylethanolamine, acetate, and butyrate. The conditions for the quantitative determination of N,N-dimethylglycine by capillary tube isotachophoresis have been determined.

The production of monoclonal antibodies against glomerular and other antigens of the human nephron.

During our studies on human kidney tubular antigens we have applied the technique of cell fusion for the preparation of monoclonal antibodies. For this purpose, plasma membranes were prepared from human kidney cortex by homogenization, fractionated on density gradients and selected according to brush border marker enzyme activity. Spleen cells from Balb/c mice hyperimmunized with plasma membranes were fused (PEG) with NS1 plasmocytoma cells by standard procedure. Culture supernatants were tested for presence of specific antibodies by indirect immunofluorescence with fluorescein-conjugated rabbit anti-mouse Fab antibodies on human kidney slices. In two fusions (210 wells), 70 positive hybrids were found secreting antibodies for a variety of antigens in the kidney. Most of them were directed against tubular antigens. In addition, as a by-product, we detected hybridomas which secreted antibodies specific for antigens in other parts of the nephron, such as glomeruli, blood cloning, and monoclonal antibodies were produced in large amounts from ascitic fluid. Some of these antibodies are specific for antigens of the basement membrane, others for antigens of the mesangium. Some recognize antigens present on glomeruli alone, others recognize antigens present on glomeruli and tubules or on glomeruli and blood vessels. We are convinced that the new immunological technique will yield better information on the antigenic microstructure of the nephron. In addition, the monoclonal and, by definition, monospecific antibodies might be useful for diagnostic purposes: recognition and quantitation of the corresponding antigens in the serum and/or urine of patients suffering from kidney diseases.

Studies on the characterization of the linkage-region between polysaccharide chain and core protein in bovine corneal proteokeratan sulfate.

1) A new method of enrichment of the linkage-region in corneal proteokeratan sulfate is described, which consists of desulfation of peptidokeratan sulfate, followed by chromatography on Con A-Sepharose 4B and enzymatic degradation with beta-D galactosidase and beta-N-acetyl-D-glucosaminidase. 2) After permethylation, hydrolysis, reduction with sodium borohydrid and acetylation gas chromatography/mass spectrometry analyses were performed. The followings products could be detected as their peracetates: 2,3,4-tri-O-methylfucitol; 2,3,4,6-tetra-O-methylmannitol; 3,4,6-tri-O-methylmannitol; 2,4-di-O-methylmannitol; 2,3,4,6-tetra-O-methylgalactitol; 2,4,6-tri-O-methylgalactitol; 2,4-di-O-methylgalactitol. 3) The results point to the presence of a branched linkage region in the proteokeratan sulfate molecule with one mannose as the branching point and two mannose residues as the starting point of two disaccharide chains.
