[Content of cholesterol extracted from human skin surface--a possible discriminant of atherosclerosis?].

Basing on the data of Bouissou et al. on an increase of cholesterol content in the skin biopsies in atherosclerosis patients, the cholesterol amount was evaluated after superficial atraumatic rapid treatment of patients palm skin. The optimization of the extraction conditions was performed: the surface skin cholesterol value was constant for individual subjects (variability factor 5%). The range of this value was 1.5-6 Mg/cm2 and it was higher for atherosclerotic patients. No correlations with any conventional plasma lipoprotein parameters were observed. At the same time positive correlation (r = 0.54. p < 0.01) was observed with dyslipoprotein coefficient--(Ch+TG/alpha-Ch). Possible diagnostic significance of this skin cholesterol parameter is discussed.

Diet-induced hypercholesterolaemia in the rabbit.

Intraspecies variation in diet-induced hypercholesterolaemia in rabbits, simulating atherosclerosis, was studied. The chinchilla rabbit population examined contained several subpopulations, as indicated by polymodal forms of histograms of plasma cholesterol levels. This finding indicates that the inclusion of subpopulations in an investigation can lead to erroneous conclusions, and that subpopulations should be identified before such work is undertaken. The relationship between the molar cholesterol/phospholipid ratio in rabbit erythrocyte membranes and the plasma cholesterol level in experimental atherosclerosis was also studied. A correlation was evident only over the range of 1-5 g cholesterol per litre of plasma.

Cholesterol oxidation on fluorocarbon emulsion surface leads to the formation of 7-peroxycholesterol.

Formation of biologically active oxidized derivatives of cholesterol as a result of its oxidation on the surface of fluorocarbon emulsions was studied. A single product of cholesterol oxidation, 7-peroxycholesterol, was found. It was shown that 7-peroxycholesterol and its derivative 7-keto-cholesterol inhibit the rosette formation between human T-lymphocytes and sheep erythrocytes. These substances exert a strong cytostatic action on the growth of procaryotic and eucaryotic cell cultures. Thus, oxidative modification of blood plasma components on the surface of fluorocarbon emulsion particles with the formation of highly active compounds must be taken into account when using the fluorocarbon emulsions in medicine.

[Formation of biologically active substances during cholesterol oxidation on the surface of fluorocarbon emulsions].

Oxidative modification of cholesterol on the surface of fluorocarbon emulsions was studied. The oxidation yielded one primary product--7-peroxycholesterol. It was shown that the obtained cholesterol C7 derivatives possess a high biological activity. It was concluded that the possibility of oxidative modification of plasma substances on the surface of fluorocarbon emulsion particles with the formation of highly active compounds must be taken into account when using the fluorocarbon particles in medicine.

[The effect of liposomes containing cholesterol or 7-ketocholesterol on the capacity of cultured cells to form colonies].

The experiments with the cultivated Chinese hamster cells showed that the incubation of cells with (phosphatidylcholine+cholesterol+7-keto-cholesterol)--containing liposomes (4:3:1 by weight) during two hours leads to a decrease in the effectiveness of colony-forming up to zero, while the (phosphatidylcholine+cholesterol-containing liposomes (1:1 by weight) reduce the effectiveness by 90%. Furthermore, the cholesterol-containing liposomes cause the decrease of the number of colonies with maximal size, accompanied by the increase of the number of colonies with middle size.

[Changes in the levels and composition of lipoproteins and apolipoproteins of blood plasma in patients with ischemic heart disease after hemosorption].

Prior to and following activated charcoal hemosorption, concentrations of lipids, apolipoproteins AI and B and lipid and protein composition in lipoprotein fractions isolated by ultracentrifugation were determined in the plasma from patients with coronary heart disease. The majority of the patients showed a parallel proportional decrease in plasma atherogenic parameters and all components of very low density lipoproteins and low density lipoproteins, triglycerides in particular. Antiatherogenic parameters, such as high density lipoprotein and apo-AI cholesterol, and all the components in high density lipoprotein subfractions were less reduced. In 19% of the patients, hemosorption failed to affect plasma lipids and apolipoproteins. The findings suggest that, in case of successful hemosorption, apoB-containing lipoproteins are chiefly eliminated as whole complexes from the plasma and that this procedure is most beneficial in hypertriglyceridemia.

[Possible approaches to creating phospholipid preparations--acceptors of membrane cholesterol].

The capability of liposomes with plant phosphatidylcholine to extract cholesterol from erythrocyte membrane was investigated. It was shown, that plant phosphatidylcholine extracted 42% of membrane cholesterol, without causing haemolysis. This cholesterol extraction was responsible for the decrease of membrane microviscosity and for the increase of Na+, K+-ATPase activity. It is suggested that it is possible to create the extractive agent of cholesterol with plant phosphatidylcholine.

The programmatic hemosorption in the comprehensive treatment of atherosclerosis of various localization. Hemosorption in the treatment of atherosclerosis.

The inclusion of hemosorption through activated coals into the comprehensive treatment of 580 patients sick with atherosclerosis has made it possible to achieve a positive clinical effect among 81 per cent of the patients sick with the ischemic heart disease, 79 per cent of atherosclerotic discirculatory encephalopathy and 76 per cent of obliterating atherosclerosis of lower limbs. The positive dynamics is especially visible among patients who are administered the programmatic hemosorption, making it possible over a long period of time (3-5 years) to sustain remission in the clinical state and the normal level of the lipids in the blood. The direct effect is expressed in the improved microcirculation, disappearance of the sludge-syndrome and the reduction of ischemic disorders. The remote effect of hemosorption is connected with the repeated removals of cholesterin of the atherogenic lipoproteids, which leads, we believe, to the interruption or even regression of the atherosclerotic process.

[Value of hemosorption and essential phospholipids in the complex treatment of patients with ischemic heart disease].

Eighty-three patients were treated with hemosorption and essential phospholipids for coronary heart disease (CHD) and followed up for 3-5 years. The new approach to the treatment of CHD involves simultaneous removal of cholesterol-rich atherogenic lipoproteins by hemosorption and administration of essential phospholipids to increase cholesterol withdrawal from the cells. This combined treatment of coronary patients is shown to normalize cholesterol metabolism, improve microcirculation, increase stress tolerance and bring about an obvious clinical remission. The good clinical effect is corroborated by the results of bicycle ergometry and data on lipid spectrum, cholesterol metabolism, microcirculation and radio-isotope studies.

[Extraction of cholesterol from biological membranes with positively charged micelles of phosphatidylcholine].

The content of cholesterol in red cell and platelet membranes was lowered in rabbits with experimental atherosclerosis after intravenous injection of positively charged micelles of soybean phosphatidylcholine. That lowering was accompanied by a reduction in membrane microviscosity, rise of the activity of Na,K- and Ca-ATPases of red cells, and a decrease in the rate of the ADP- and collagen-induced platelet aggregation. Injection of phosphatidylcholine gave rise to an increase in the blood serum content of phospholipids and cholesterol in high density lipoprotein fractions, to a reduction in the content of triglycerides and the atherogenicity index, as well as to the lowering of the microviscosity of high density lipoproteins. The aortal area affected by atherosclerotic lesions was 2 times less in the group of animals given phosphatidylcholine.

[Structural and functional changes in thrombocytes in experimental atherosclerosis].

Electron paramagnetic resonance with the use of stearic acid derivatives (5- and 16-doxylstearate) as spin probes was applied to studies of the structural organization of rabbit platelets in experimental atherosclerosis. Substantial differences were established in the molecular packing of phospholipid plasma membranes, associated with a higher molar content of cholesterol in the cells. An increase in the aggregation properties of platelets was also observed, manifesting in a shorter time of the ADP-induced aggregation of platelets isolated from plasma. The data obtained confirm the primary part of membranotropic cholesterol activity in atherosclerosis, attesting to the validity of the "membraneous" hypothesis of the atherogenesis.

[Change in the properties of plasma membranes of normal and neoplastic cells after incorporation of cholesterol].

It is shown that cholesterol incorporation into the membranes of lymphocytes, lymphoblast leikose cells L1210 and into those of ovary tumor causes an increase in the membrane phospholipid bilayer microviscosity measured by pyrene as a fluorescent probe. An increase in the membrane lipid microviscosity resulted in a decrease in the activity of Na+, K+-ATPase and 51-nucleotidase of the normal and tumor cells. After the injection of the tumor cells with an elevated cholesterol/phospholipid ratio an increase of the life span in the experimental animals as compared with the control group was observed.

[Changes in blood lipoproteins in patients with peripheral atherosclerosis after hemosorption].

Concentrations of cholesterol, triglycerides and the molar ratio cholesterol/phospholipids were higher while the values of cholesterol in high density lipoproteins (HDL) was lower in blood of patients with obliterating peripheric atherosclerosis as compared with control values. After extracorporal haemosorption on columns with activated charcoal and preadministration of heparin all the patterns studied were decreased, thus indicating the sorption of all the lipoprotein classes, including anti-atherogenic HDL, simultaneously with lipolysis of very low density lipoproteins, triglycerides as well as the sorption and destruction of large erythrocytes containing relatively high amount of cholesterol. Within the subsequent period all the patterns studied returned to the initial level. At the same time, concentration of HDL cholesterol, which, as shown earlier, correlated inversely with severity of clinical manifestations in the patients, continued to increase, exceeding the initial level by 39% within 7-14 days; these data were also corroborated by polyacrylamide gel disc electrophoresis. The alterations found may be among the factors, responsible for favourable clinical effect of haemosorption in the patients with peripheric atherosclerosis. Possible reasons for the HDL concentration increase are discussed.
