Manifestation and growth of a transplantable osteosarcoma in mice: the effects of thymectomy and thymosin treatment.

Normal and thymectomized CBA mice treated with and without thymosin were injected with 2 x 10(6) viable osteosarcoma cells. The activity and weight of tumors, the cell-mediated and humoral-immune responses against the tumors, and the light microscopic characteristics suggested that thymectomy resulted in a significantly decreased tumor frequency. There were also significant increases in cell-mediated cytotoxic activity in vitro as measured by lysis of 51Cr-labeled tumor cells incubated with spleen cells, and in lymphocytic infiltration around the tumors. While thymosin treatment of thymectomized mice restored these parameters to those of intact animals, thymosin had no demonstrable effects on nonthymectomized animals.

Discriminative value of isozymes of amino acid naphthylamidase in the diagnosis of myeloid leukemias.

Isozymes of amino acid naphthylamidase (called B and C) with deviating electrophoretic mobilities were found in peripheral blood leukocytes of 25 out 25 untreated acute and chronic myeloid leukemias (AML and CML) and in 2 out of 2 cases of idiopathic myelofibrosis, while a normal pattern was found in 3 control groups and 5 cases of polycythemia vera. In the AML group, a correlation between electrophoretic mobility and the number of blast cells was found, and on remission, the B isozyme mobility was nearly normalized. Thus, deviating electrophoretic mobility of the B isozyme seemed to be valuable for diagnosis of myeloid leukemias, and in the AML group, the change in mobility might indicate the size of the leukemic clone.

Isozymes of amino acid naphthylamidase in human haematopoietic cell lines.

The occurrence of isozymes of the proteolytic enzyme amino acid naphthylamidase was investigated in a panel of 44 neoplastic and non-neoplastic haematopoietic cell lines by polyacrylamide gel electrophoresis. Two isozymes (A and C) were common for all lines, whereas different forms of a third isozyme with intermediate electrophoretic mobility (B) appeared in nine lines. Isozyme A seemed to be associated with cell proliferation and was not a marker for malignancy. The appearance of different B isozymes not present in normal peripheral blood lymphocytes, occurred in some Burkitt lymphoma lines and in two non-malignant lines maintained for a long time in culture. In three lines derived from myeloid leukaemia a variant of isozyme B seemed to be a marker for cell origin.

Staging laparotomy for Hodgkin's disease.

Splenectomy was performed in patients with Hodgkin's disease in a prospective study at the University Hospital of Umeå 1973-1978. Fifty-nine subjects were included and the laparotomy was made as a part of the pre-treatment investigation. The result of the surgical exploration was compared to other investigations, e.g., lymphangiography or cavography. The operation morbidity was 5% (3/59). The clinical stage was changed in 15/58 (26%). Fourteen upstaged and one downstaged. The macroscopical evaluation was poorly correlated to the histological involvement. It is concluded that if the staging procedure is performed by specialized surgeons the morbidity is low. No adverse effects have yet been observed after splenectomy. The staging laparotomy and splenectomy is considered superior to nonsurgical staging procedures from both treatment and cost benefit point of view.

Cell lines with spontaneous secretion of pregnancy-associated alpha 2-globulin.

Pregnancy-associated alpha 2-globulin (PA alpha 2G) was quantitated by radioimmunoassay in culture media from 34 exponentially growing human cell lines. Only 8/34 cell lines produced PA alpha 2G. The highest secretion was found in cell lines of histiocytic lymphoma origin while it was low in some carcinoma-derived lines. The results may support an assumption that PA alpha 2G is a normal cell product of B-lymphoid, monocytic and epithelial cells but may also simply indicate that ectopic PA alpha 2G production is especially common in tumors derived from such cells. The availability of PA alpha 2G-producing cell lines should facilitate studies of the immunoregulatory role of this protein.

Discrepant effects of interferon murine syngeneic ascites tumors and their solid metastasizing counterparts.

The effects of interferon on mice with syngeneic tumors growing either intraperitoneally or as solid tumors were analyzed. The same dose that depressed the growth of ascites tumors, enhanced growth and in one case metastasizability of the corresponding solid tumors. The possibility that this was due to some host reaction is discussed. It seems important to develop animal models for analyzing interferon effects before treatment is initiated in human tumor disease.

Effects of leukocyte and fibroblast interferon on events in the fibroblast cell cycle.

Serum-depleted human foetal skin fibroblasts were stimulated by addition of 10% foetal calf serum to proliferate synchronously for at least one cell cycle. This proliferation was suppressed by leukocyte or fibroblast interferon (IF), which prolonged the G1 phase and diminished the rate of DNA synthesis during the S phase in a dose-dependent manner. When used in identical concentration, as judged in terms of units of antiviral activity, fibroblast IF had more pronounced effects on cell cycle events than leukocyte IF. Interferon exerted its effect in early G1, before the cells were irreversibly committed to DNA synthesis.

Clinico-pathologic correlation in non-Hodgkin's lymphoma. I. Retrospective analysis using the Lukes and Collins classification.

A retrospective review of 302 patients with non-Hodgkin's lymphoma is presented. The tumors were classified according to a modified Lukes and Collins scheme. Clinico-pathologic correlation was analysed for the subtypes. Certain distinct features were found with respect to age, sex, clinical stage and sites of involvement at admission. Prognostically, two major groups were found with significantly differing survival curves. The pathologic and clinical significance of the modified Lukes and Collins classification is discussed.

Clinico-pathologic correlation in non-Hodgkin's lymphoma. II. Clinical and prognostic significance of nodularity.

A retrospective review of 302 patients with non-Hodgkin's lymphoma is reported. The tumors were classified according to a modified Lukes & Collins scheme and their growth pattern was analysed. Nodular growth was scored in 3 degrees. Nodularity was found to be correlated with certain clinical features and of great relevance for prognosis. The growth pattern was also correlated with cell type, but even within a certain cytologic type, nodular tumors were associated with better survival than diffuse tumors.

The effect of zinc tape upon wound healing. A biochemical, histochemical and histological study in rats.

The healing of excisional wounds in rats, which were treated with zinc tape, gauze sponge or porcine skin, was studied. Wound closure was completed earlier in zinc-tape-treated wounds than in wounds treated with a gauze sponge or porcine skin. Wound contraction was more pronounced in gauze- and porcine skin-treated wounds than in zinc-tape-treated wounds. More foreign body giant cells were seen 14 days after operation in gauze-treated contra zinc tape-treated wounds, whereas hydroxyproline concentration was higher in tape-treated wounds. Histochemically the acid phosphatase activity was pronounced in macrophages, foreign body giant cells and fibroblasts. The alkaline phosphatase activity was pronounced in granulocytes and fibroblasts. Quantitatively a reduction in alkaline phosphatase activity was seen in gauze-treated wounds from 7 to 14 days. A decrease was seen in albumin concentration in gauze- as well as zinc-tape-treated animals which was most pronounced after 7 days in the gauze-treated animals.

Transplantable osteosarcoma in mice. Structural characterization of a transplantable osteosarcoma obtained in an allogenic system.

Light microscopic, histochemical and ultrastructural studies of a transplantable mouse osteosarcoma were carried out. The osteosarcoma grew in CBA mice after injection of cultured cells derived from a Dunn osteosarcoma. The tumour differed from the original Dunn osteosarcoma with respect to metastatic potential and structural features. The transplantable tumour was an anaplastic, richly vascularized fibroblastic osteosarcoma with alkaline phosphatase activity and rather sparse osteoid formation, resulting in death of the animals within 6 to 8 weeks. Virus particles were found intracellularly, mainly localized to cisterns of rough endoplasmic reticulum, and extracellularly often close to plasma membranes and collagen fibres. Sign suggestive of formation of collagen fibres by tumour cells were observed. A possible viral influence upon the tumour was suggested also by its growth behaviour in vitro. The results indicate that this new transplantable tumour, obtained in an allogenic system, represents a clonal derivative of the original Dunn osteosarcoma.
