Effect of thyroid hormones on activity of nonspecific phosphohydrolases in blood serum, tissues and liver mitochondrial fraction of white rat.

Activity of alkaline and acid phosphatase was determined by the method of Bodansky in blood serum, homogenates of liver, kidney, lung, brain, liver mitochondrial fraction of normal, methylthiouracil-induced hypothyroid, and Thyroideum-induced hyperthyroid rats. The following results were obtained: an increase in both enzymes in all studied materials in hyperthyroidism, and a decrease in most of the studied materials in hypothyroidism.

Acid Phosphatase↗

Studies on 5'-nucleotidase activity in blood serum, tissues and liver mitochondrial fraction of normal, hypo- and hyperthyroid rats.

Clinical and experimental studies have previously shown the effect of thyroid state on the activity of non-specific phosphohydrolases. This research was undertaken to determine the influence of thyroid hormones on the activity of specific phosphohydrolase-5'-nucleotidase. The activity of 5'-nucleotidase was estimated in blood serum, tissues (liver, kidney, lung, brain) and mitochondrial fraction of liver of normal, methylothiouracil-induced hypothyroid and thyroxin-induced hyperthyroid rats of Wistar strain. The experiments have shown that the hypothyroid state induced decrease of the activity of 5'-nucleotidase in all studied materials, and on the contrary, the hyperthyroidism induced the increase of the enzyme activity. The hypothetical mechanism of the effect of thyroid hormones on the processes of feed back regulation of oxydative phosphorylation and delivery of substrates and oxygen is presented. The key role of 5'-nucleotidase in this mechanism is discussed.

Perchloric acid-soluble proteins and protein-bound hexoses in serum and tissues of normal, hypo-and hyperthyroid rats.

The total content of perchloric acid-soluble proteins (PCA-SP) and protein-bound hexoses (PBH) were studied in blood serum and tissues (liver, kidney, lung, brain) of normal, hypo- and hyperthyroid rats. The results obtained indicate that the hyperthyroid state induced an increase in both PCA-SP and PBH in blood serum and studied tissue homogenates, and hypothyreosis causes a decrease of total glycoproteins content in all studied materials. The results were counted per 1 g of protein and in this way the nonspecific effect of thyroid hormones on the protein metabolism had no influence on the measurements.

Histopathological and histochemical studies of the skin of guinea pigs after long-term exposure to nitrogen dioxide.

Male guinea pigs were exposed to nitrogen dioxide (2 mg/m3) during 180 days (8 hours a day). Long-term exposure induced thickening of the corneal layer of the epidermis as well as inflammatory infiltrations in the proper skin. The following enzymes were estimated histochemically in skin samples of experimental and control animals: succinic dehydrogenase, NADH2-tetrazolium reductase, lactate dehydrogenase; alkaline phosphatase, acid phosphatase and adenosine triphosphatase. Chronic exposrue stimulated a decrease of NADH2-tetrazolium reductase in the epidermis and connective tissue components of proper skin and marked positive reaction of lactate dehydrogenase in epidermal cells and hair follicles. Increase of a diffuse reaction on adenosine triphosphatase in smooth muscles of the skin was found also in exposed animals.

Studies on the effect of long-term exposure to nitrogen dioxide on serum and liver proteins level and enzyme activity in guinea pigs.

Forty male guinea pigs were exposed to nitrogen dioxide in a concentration of 2 mg/m3, 8 hours daily for a period of 180 days. Forty male animals were used as a control group. The following changes were found in intoxicated animals: the decrease of total protein and seromucoid concentration in blood serum and the decrease of total protein, perchloric acid-soluble proteins, protein-bound hexosamines and sialic acids content, in liver tissue. Electrophoretic examination of the serum proteins showed the increase of alpha 1- and beta 2-globulins and the decrease of albumin concentration. Changes in the level of glycoproteins fractions and protein-bound carbohydrates in blood serum were described also. Estimation of enzymes activity showed the decrease of alanine and aspartate transaminase activity in blood serum caused by the strong decrease of the cytoplasmic fraction of these enzymes. However the simultaneous increase of the mitochondrial fraction of transaminases activity was observed. The decrease of the activity of choline esterase was found also. Similar changes of enzymes activity were found in liver tissue. Histopathological studies were done for the further clearing the influenze of nitrogen dioxide on serum and liver proteins concentration and enzymes activity. It was found that after long-term exposure to nitrogen dioxide the destruction processes may be observed in the liver. The possible mechanism of the nitrogen dioxide-induced damage of protein metabolism is discussed.

Collagen content in the heart of patients operated on for valvular heart disease caused mainly by infective endocarditis.

Collagen from tissues of the heart valve, the wall of the left auricle, palatine tonsils and periodontium was studied. The material was taken during surgical procedures, performed for therapeutic reasons, in patients with acquired valvular heart disease (avhd) and congenital heart disease. The obtained results pertain to the content of total collagen as well as to its soluble and insoluble fractions. An increase of the soluble collagen fraction, as compared to the insoluble one, provides support for collagen degradation, and this fact has prompted the authors to conclude that collagen plays a part in the etiopathogenesis of a.v.h.d. The authors suggest that estimation of collagen metabolites, and the amount of soluble collagen in tissues, and particularly in palatine tonsils, may be helpful in identifying persons susceptible to a.v.h.d.

Investigations upon collagen metabolites in blood serum and urine of patients with acquired valvular heart disease.

In 45 patients suffering from acquired valvular heart disease (a.v.h.d.) and in 20 controls blood serum levels of copper (Cu2+), ceruloplasmin (Clp), hydroxyproline (Hypro), hydroxylisine (Hyliz), collagen-like protein (Col-p), and daily urine excretion of Cu2+, Hypro, Hyliz and glycosamino-glicans (GAG) were determined. The comparison of the obtained mean values has shown that blood serum levels of Cu2+, Clp, Hypro, Hyliz, Col-p in the studied patients were statistically significantly higher than those found in controls, and the values of daily urine excretion of Cu2+, Hypro, Hyliz, GAG behaviour were similar. There were no significant differences between active and inactive processes and between infective and rheumatic endocarditis excluding GAG urine excretion. These elevated collagen degradation products provide evidence for disturbed collagen metabolism and confirm our previous biochemical, histopathological and ultrastructural examinations of connective tissue in patient with a.v.h.d. They also indicate that general collagenopathy might be the primary cause of a.v.h.d.

Influence of thyroid hormones and methylthiouracil upon elastin content in the liver of rats with carbon tetrachloride-induced hepatic fibrosis.

The effect of administration of thyroid extract and methylthiouracil upon the elastin content of the liver was investigated in healthy and CCl4-poisoned rats. Hyperthyroidism induced a decrease in liver elastin content and hypothyroidism led to an increase of liver elastin. Similar changes were found in rats with carbon tetrachloride-induced hepatic fibrosis. The possible mechanisms of these changes are discussed.
