Correlations of monocyte phagocytic receptor expressions with serum immune complex level in systemic lupus erythematosus.

The expression of Fc gamma RI, Fc gamma RII, and Fc gamma RIII (the IgG receptors CD64, CD32, CD16) as well as CR3 (the C3bi receptor, CD11b) on monocytes in the blood of patients with systemic lupus erythematosus (SLE) was investigated. The relationship between the receptor expression and the serum immune complex (IC) concentration was analysed. The decrease in mean fluorescence intensity (FI) of the Fc gamma RII of patients' monocytes stained by specific monoclonal antibodies (MoAb IV3) was very close to statistical significance (P = 0.052). The expression (FI) of CR3 (using MoAb OKM1) on monocytes of patients was also decreased, but not significantly. The detected decrease of Fc gamma RII and CR3 was inversely correlated with the high circulating immune complex level in patients' sera. At the same time, Fc gamma RI expression on SLE monocytes (using MoAb 32) was significantly elevated and this change was in parallel with the serum IC concentration.

Thyroid hormone-induced alterations in phospholamban protein expression. Regulatory effects on sarcoplasmic reticulum Ca2+ transport and myocardial relaxation.

The aim of the present study was to determine the changes in phospholamban protein levels and their regulatory effect on sarcoplasmic reticulum (SR) Ca2+ uptake and left ventricular function in hypothyroid and hyperthyroid rat hearts. Hypothyroidism was associated with decreases in basal left ventricular function (+dP/dt and -dP/dt), whereas in hyperthyroidism these parameters were elevated compared with values for euthyroid hearts. The maximal SR Ca2+ uptake rates were 12.8 +/- 1.1, 15.5 +/- 1.2, and 21.4 +/- 1.4 nmol Ca2+ per milligram per minute, and the EC50 values for Ca2+ were 0.76 +/- 0.09, 0.41 +/- 0.07, and 0.30 +/- 0.05 mumol/L assayed in homogenates from hypothyroid, euthyroid, and hyperthyroid hearts, respectively. The relative tissue level of phospholamban was increased (135%) in hypothyroidism and decreased (75%) in hyperthyroidism compared with euthyroidism (100%). An opposite trend was observed for the SR Ca(2+)-ATPase, which was depressed (74%) in hypothyroid hearts but increased (134%) in hyperthyroid hearts. Consequently, the relative ratio of phospholamban to Ca(2+)-ATPase was highest in hypothyroid and lowest in hyperthyroid hearts, and these changes correlated with changes in the EC50 of the SR Ca2+ uptake for Ca2+. Stimulation of hearts with 0.1 mumol/L isoproterenol revealed that the relaxant effects were lower in hyperthyroid hearts and higher in hypothyroid hearts compared with euthyroid hearts, consistent with the alterations in the phospholamban levels. The maximal increases in the speed of relaxation, elicited by isoproterenol stimulation, correlated with the changes in the relative ratio of phospholamban to Ca(2+)-ATPase in these hearts.(ABSTRACT TRUNCATED AT 250 WORDS)

[Formaldehyde determination using the colorimetric method with acetylacetone. II. Formaldehyde determination in cosmetic emulsions and certain household products].

Total formaldehyde content was determined by the colorimetric method with acetylacetone in cosmetic emulsions and household products. The commercial products not containing formaldehyde were fortified with 150 micrograms of the investigated compound. Recovery in this method was 93.3-102.6%. The method was found useful in routine determinations of formaldehyde in cosmetics and household products and can be used by the State Sanitary Inspections.

Immunological aspects of the effect of pentoxifylline (Trental) (a brief review).

Pentoxifylline (methixanthine derivate) has haemorheological activity, thought to based on its ability to reduce blood viscosity and increase the filterability of blood cells and widely used for the treatment of various types of vascular insufficiency. Recent studies provide evidence for the effect of this drug on immune functions. Pentoxifylline is able to influence in vitro as well as in vivo production of cytokine including tumour necrosis factor alpha. This drug has been shown to attenuate the symptoms of endotoxin shock and adult respiratory distress syndrome. Its beneficial effect on tumour-induced cachexia, organ transplantation and AIDS can widen the indications of clinical administration. Nevertheless, the chronic continuous administration of Pentoxifylline in arterial insufficiency is to be considered due to its potentially enhanced susceptibility to malignant disorders.

Soluble intercellular adhesion molecule-1 (sICAM-1) in Graves' disease.

The aim of this study was to determine the level of sICAM-1 in sera of hyperthyroid and euthyroid patients with Graves' disease and find correlation between thyroid function, anti-TSH receptor as well as anti-thyroid antibodies. Comparative determination of sICAM-1, TSH and thyroid hormones, anti-TSH receptor- as well as anti-thyroglobin-, anti-thyroid peroxidase antibodies was made in sera of Graves' patients. Thirty patients with untreated hyperthyroid Graves' disease (12 had infiltrative ophthalmopathy) were studied before and after methimazole treatment. sICAM-1 was measured by ELISA using monoclonal antibodies to human ICAM-1. The sICAM-1 was significantly elevated in hyperthyroid Graves' patients (738.9 +/- 151 ng/ml) compared to controls (212 +/- 42 ng/ml) (p < 0.0001). Both in hyperthyroid and euthyroid Graves' groups with infiltrative ophthalmopathy the sICAM-1 proved to be higher than those without ophthalmopathy. Correlations were observed between sICAM-1 and serum thyroxine, triiodothyronine and anti-TSH receptor antibodies (r = 0.91, 0.83 and 0.61, respectively). There was, however, no association of serum ICAM-1 levels with anti-thyroid peroxidase or anti-thyroglobulin antibodies. It was concluded that elevation of sICAM-1 can be the consequence of hyperthyroidism and the autoimmune processes mediated by anti-TSH receptor antibodies as well as cytokines. The elevated level of sICAM-1 might be important both in regulation of autoimmune mechanism and "homing" phenomenon of lymphocytes in Graves' disease.

Effect of thyroid status on basal phosphorylation of cardiac myofibrillar phosphoproteins in rats.

The effect of thyroid status on myocardial function and accompanying alterations in the expression of specific genes has been well defined in animals. However, the effects of thyroid hormones on the basal phosphorylation of key cardiac regulatory proteins, which may also contribute to alterations in myocardial function, have not been defined. The present study concerns the phosphorylation status of myofibrillar proteins in hearts from hyperthyroid, euthyroid and hypothyroid rats. Hyperthyroidism was produced by daily subcutaneous injections of L-triiodothyronine, while hypothyroidism was induced with an iodine-deficient diet and KClO4. Two different approaches were used to study changes in the basal phosphorylation levels of troponin I and C protein: 1) direct measurement of the 32P-label associated with these proteins, using intact, beating hearts perfused with [32P]orthophosphate-labeled Krebs buffer; 2) indirect measurement by the back-phosphorylation technique with [gamma-32P]ATP and the catalytic subunit of cAMP-dependent protein kinase in vitro. Measurements of left ventricular contraction (+dP/dt and -dP/dt) were significantly higher in hyperthyroid than in euthyroid animals and this was associated with increases in basal phosphorylation levels of both troponin I and C protein in the myofibrils. In hypothyroid animals, both +dP/dt and -dP/dt were significantly lower than in euthyroid animals and this was associated with decreases in basal phosphorylation levels of troponin I and C protein. The changes in the phosphorylation status of troponin I or C protein correlated with the changes in the speed of myocardial relaxation (-dP/dt) in response to the altered thyroid states.(ABSTRACT TRUNCATED AT 250 WORDS)

Biochemical consequences of 5-fluorouracil gastrointestinal toxicity in rats; effect of high-dose uridine.

Selective protection of the normal host tissues from the toxic effects of anticancer agents would allow the use of higher, probably more effective, doses of the drugs. It has been demonstrated that delayed high-dose uridine administration after 5-fluorouracil decreases the extent of myelosuppression and causes faster regeneration of the bone marrow. We studied the biochemical consequences of the gastrointestinal toxicity caused by 5-fluorouracil and the potential of high-dose uridine treatment to influence these adverse effects. 5-Fluorouracil caused dose-related decreases in the biochemical parameters (thymidine kinase, sucrase, maltase, alkaline phosphatase) selected as early markers of the impaired metabolic activity of the intestinal mucosa. The nadir of the biochemical changes was reached between 24 h and 72 h after 5-fluorouracil treatment, and complete regeneration of the mucosa took 6-7 days. Delayed high-dose uridine administration failed to mitigate the severity of the gastrointestinal damage that ensued after 5-fluorouracil treatment, but caused significantly earlier regeneration of the mucosa.

Triggering of respiratory burst by phagocytosis in monocytes of patients with systemic lupus erythematosus.

The triggering of the respiratory burst by phagocytosis via different receptors in monocytes of patients with systemic lupus erythematosus (SLE) was investigated. The superoxide anion synthesis was assayed by reduction of ferricytochrome C that was inhibited by superoxide dismutase. The mononuclear cell suspensions were triggered by IgG-coated latex, C3 complement fragment-coated and uncoated yeast (Saccharomyces cerevisiae). Superoxide generation induced by phagocytosis via Fc gamma R was decreased in monocytes of patients with SLE. On the other hand, MoAbs against Fc gamma RI, Fc gamma RII and especially CR3 could also induce superoxide anion synthesis. At the same time, superoxide generation induced by anti-CR3 could be inhibited with C3-coated yeast.

Developmental analysis of primary and secondary somatic embryogenesis in soybean tissue culture.

Somatic embryogenesis from immature soybean cotyledons was induced on Murashige & Skoog medium, supplemented with 2,4-dichlorophenoxyacetic acid. The full developmental analyses of primary and secondary somatic embryogenesis was monitored by scanning electron microscopy. The developmental stages of embryogenesis from the first unequal cell division through the three- and four-cell embryos, and multi-cell globular and heart shape embryos, to the fully developed plants were determined. When the globular stage embryos were incubated on the embryo maturation medium for 4 to 6 weeks a secondary embryogenesis was observed and scanned. Chimeric embryos with epidermis zones similar to mature leaf were characterized.

Effect of thyroid status on phosphatidylinositols in rat heart.

The incorporation of 32Pi into phosphatidylinositols and inositol trisphosphates was studied in Langendorff-perfused hearts from hypothyroid, euthyroid and hyperthyroid rats. The hearts were perfused with modified Krebs buffer containing [32P]orthophosphate and the degree of 32P-labeling of phosphatidylinositol, phosphatidylinositol 4-monophosphate, phosphatidylinositol 4,5-bisphosphate, inositol trisphosphates and phosphatidic acid was measured. Hyperthyroidism was associated with increases in rates of rise and fall of left ventricular systolic pressure, sarcoplasmic reticular Ca(2+)-ATPase activity and 32P-labeling of phosphatidylinositols, inositol trisphosphates and phosphatidic acid. These measurements were significantly decreased in hypothyroid hearts. The tissue levels of inositol 1,4,5-trisphosphate isoform were found to be significantly higher in hyperthyroid hearts and lower in hypothyroid hearts than in euthyroid ones. Examination of phosphoinositide-specific phospholipase C activity in the perfused hearts revealed that hyperthyroidism was associated with an increase in the membrane-associated enzymatic activity, assayed at physiological calcium concentrations, while hypothyroidism was associated with a decrease in this activity as compared with control hearts. These findings indicate that alterations in the thyroid state of the myocardium may be associated with changes in basal phosphoinositide turnover which may contribute to alterations in myocardial contraction.

[Ergometric stress test using Tl-201 myocardial perfusion. Tomographic or planar imaging?].

The clinical performance of the planar vs. tomographic ergometric stress 201-Tl myocard perfusion scintigraphy was compared. The results of the investigations of 80 patients were analysed retrospectively. In 59 patients planar, in 21 tomographic imaging was performed. As a consequence of clinical decision making cardiac catheterization was made in all of the patients, within 3 months after the perfusion scintigraphy. The sensitivity of the planar method was 23/27 (85%), the specificity 18/32 (56%), the accuracy 41/59 (69%). The respective values of the SPECT technique were: Sensitivity: 12/12 (100%), specificity 6/9 (66%), accuracy 18/21 (86%). It is concluded, that in comparison to the planar imaging the SPECT technique is superior in the detection of the perfusion abnormalities of the myocard due to significant coronary narrowings.

Inhibition of metabolic activity of polymorphonuclear granulocytes by thyroid stimulating antibodies.

Both thyrocytes and polymorphonuclear granulocytes (PMNs) are known to have on their surface thyrotropin receptor (TSH-R). Anti-TSH-R antibodies having stimulating and blocking effects on thyroid function have been detected in sera of Graves' patients. Thyroid stimulating anti-TSH-R antibodies (TSAb) are involved in the pathomechanism of thyrotoxicosis. The basic question is in this study whether TSAb exerts effects on metabolic activity of PMNs. Immunoglobulin G (IgGs) of 32 patients with thyrotoxicosis had significant inhibitory effect (p less than 0.001) on chemiluminescence of healthy PMNs compared to controls. An inverse correlation was observed between inhibitory effect of IgGs on function of PMNs and TT4 (r = -0.61) in contrast to anti-TSH-receptors antibodies (r = 0.09). TSAbs of eight hyperthyroid patients were measured by determination of cyclic AMP in suspension and slices of porcine thyroid gland. Function of human PMNs separated from healthy donors was determined by luminol-amplified chemiluminescence. Photon-emission of PMNs induced by adherence was significantly lower than that of controls. None of patients' IgGs contained anti-PMNs antibodies or other toxic compounds. It was found an inverse correlation between chemiluminescence of PMNs and capability of TSAb to increase intracellular cAMP in thyroid slices and the thyroid hormone levels of patients with thyrotoxicosis (p = 0.0005). In conclusion, TSH-R on PMNs did not belong to "mute" receptors and metabolic changes in PMNs induced by TSAb might have pathogenetic as well as methodological implications.

Plasma renin activity and sleep-wake structure of narcoleptic patients and control subjects under continuous bedrest.

In order to determine if renin release would be affected by a dysfunction of the circadian and ultradian organization of sleep, 24-hour profiles of plasma renin activity (PRA) concomitant with sleep stages were established in 10 normal subjects and nine narcoleptic patients, with 10-minute blood sampling intervals. Mean PRA levels were similar in control subjects and narcoleptic patients. Individual 24-hour profiles revealed that the previously described association between renin oscillations and sleep stage alternations was preserved. Increased PRA release was observed during the transition from rapid eye movement (REM) sleep or waking periods to nonrapid eye movement (NREM) sleep, and REM sleep occurred as PRA levels were decreasing. Thus, PRA curves exactly reflected the irregularities and disturbances in the sleep structure of the narcoleptic patients. The 24-hour PRA profiles of the patients did not show the general upward trend during nighttime sleep, which is probably induced in the control subjects by the repetitive recurrence of longer episodes of undisturbed NREM sleep. Because of marked sleep fragmentation in the patients, the duration of NREM sleep was often insufficient to allow for the occurrence of a significant PRA increase. Because sleep onset REM (SOREM) episodes, characteristic of narcolepsy, are not preceded by NREM sleep and its associated increase in PRA, no relative PRA decline occurred during this type of REM sleep. In conclusion, the 24-hour PRA profiles of the narcoleptic patients reflected exactly their sleep stage distribution, confirming previous findings that PRA oscillations appear to be inseparable from the NREM-REM sleep cycle.(ABSTRACT TRUNCATED AT 250 WORDS)

[Determination of formaldehyde using the colorimetric method with acetylacetone. I. Determination of formaldehyde in selected cosmetic products].

Total formaldehyde content was determined by the colorimetric method with acetylacetone in shampoos and foam baths. The commercial products containing formaldehyde were fortified with a solution of this compound of various amounts. Recovery in this method was 91.7-98.7%. The method was found useful in routine determinations of formaldehyde in shampoos and foam baths and can be used by the State Sanitary Inspection.

Disinfection of regenerated dialyzers with ozone.

Regenerated, ozone-disinfected dialyzers were use for a total of 190 hemodialyses in 35 patients without the occurrence of any complication or pyrogen reactions; the efficiency of hemodialysis was identical with that of new or formalin-disinfected dialyzers. The priming fluid contained in both compartments of ozone-disinfected dialyzers proved to be sterile. The ozone did not damage the dialyzer membrane or the supporting structure of dialyzer. Ozone can effectively substitute formalin and other chemical sterilants as a disinfectant of regenerated capillary dialyzers. The process can also be automated.

[Diagnosis of femoral pseudoaneurysm and factors contributing to its incidence after heart catheterization].

Two-hundred patients who underwent heart catheterization between 30 November 1989 and 30 September 1990, were followed up to establish the frequency of the vascular complications (mainly femoral pseudoaneurysm) of the procedure. Factors were evaluated which may promote the occurrence of femoral pseudoaneurysms. This complication was diagnosed in 6 cases. Two-dimensional and Doppler echocardiography (conventional and colour-coded) played an important role in the diagnosis. Systemic hypertension, anticoagulant therapy, obesity and technical difficulties (multiple punctures and catheter guidance problems) proved to be factors promoting femoral pseudoaneurysms. Serial echocardiograms revealed a tendency to spontaneous healing even in cases of relatively large pseudoaneurysms.

Effects of chronic alcohol ingestion on myocardial lipid and fatty acid composition in adult turkeys.

STUDY OBJECTIVE: The aim was determine whether alcohol induced left ventricular dilatation and myocardial lesions in turkeys are associated with changes in the phospholipid and fatty acid compositions of the cardiac tissue. DESIGN: Triglyceride, phospholipid, and cholesterol concentrations and the fatty acid compositions of phospholipids and triglycerides were measured in the myocardium of control and alcohol fed turkeys. Tissue samples were taken from the anteroapical wall of the left ventricle. SUBJECTS: Seven week old domestic Nicholas turkeys were used (10 controls and 10 ethanol fed); the alcohol, comprising more than 20% of the dietary energy, was given in the drinking water for 16 weeks. MEASUREMENTS AND MAIN RESULTS: Left ventricular function was assessed by echocardiography at the end of the treatment. Different phospholipid fractions were isolated by two dimensional thin layer chromatography, and individual fatty acids were identified by gas chromatography. Non-invasive evaluation of the cardiac function revealed a dilatation of the left ventricle and an impaired contractile function in the alcoholic birds. Increased amounts of triglyceride, phosphatidylinositol, sphingomyelin, lysophosphatidylcholine, and lysophosphatidylethanolamine were found in the left ventricle of the alcoholic turkeys. Examination of the fatty acid compositions of different phospholipids and the triglyceride fraction revealed an increase in the saturated/unsaturated ratio following ethanol treatment. In the phospholipid fractions of the alcoholic birds the arachidonic acid (20:4n-6) content was significantly decreased as were the linolic (18:1) and linoleic (18:2n-6) acid contents in the triglycerides. CONCLUSIONS: These alterations in the myocardial phospholipid and fatty acid compositions may contribute to the electrophysiological and functional derangements of the left ventricle in alcoholic cardiomyopathy.
