Dietary ascorbic acid and muscle carnitine (beta-OH-gamma-(trimethylamino) butyric acid) in guinea-pigs.

Tissue ascorbic acid (AA) contents of approximately 12 and 100% saturation respectively were produced in two groups of guinea-pigs. The 'low-AA' group had a significantly lower muscle carnitine concentrations than the 'high-AA' group. There was no concomitant emergence of the symptoms customarily regarded as characteristic of hypovitaminosis C. It is concluded that muscle carnitine (beta-OH-gamma-(trimethyl-amino)butyric acid) is a highly-sensitive indicator of tissue AA contents; this could account for the lassitude and fatique reported to precede the emergence of frank scurvy in man.

The inclusion of nonphysician health care practitioners in hospital quality assurance programs is being called for by the new JCAH standards and PSRO regulations/PSRO guidelines require that nonphysicians be reviewed by peers.

Tissue ascorbic acid and a liver dehydroascorbatase in guinea-pigs.

The extraction, partial purification and assay of a dehydroascorbatase from guinea-pig liver is described. There was no evidence that changes in dehydroascorbatase activity could account for the modified tissue ascorbic acid concentrations associated with aging or with the ingestion of fluoride, flavonoids or anthocyanin material.

A comprehensive client management system for crisis intervention services.

This paper presents and evaluates a comprehensive client management system for suicide and crisis intervention services. The management system consists of five service designations: self-injury, crisis, active support, general support, and single contact. These designations determine the extent and type of activity engaged in, as well as the scope of follow-up and monitoring. As intended, self-injury clients received more contacts, longer periods of involvement, and more time in direct service. There was little difference in the length of each contact and the ability to ascertain outcome. While the management system established valuable service guidelines and priorities, it did not resolve approaches to some of the idiosyncratic problems presented to suicide and crisis intervention services.

Effect of total body temperature on toxicity of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU).

One hundred eighty-two rats were divided into groups to test the effect of three different total body temperature levels on the toxic effect of three different dose levels of 1,3-bis(2-chlorethyl)-1-nitrosourea (BCNU) and to assess the effect of delayed total body temperature elevation on BCNU breakdown products. Results were tabulated on the basis of life survival figures. At depressed total body temperatures (28 degrees C), normally expected toxicity was avoided. Elevated body temperatures, on the other hand, enhanced the toxic effect of BCNU. Delayed total body temperature elevation (3 and 6 days after BCNU administration) created similar toxicity. This finding was not observed when total body temperature elevation was delayed 10 days. The results point toward an interesting interaction between BCNU and heat (immediate and delayed) on the basis of an elevated metabolic rate of tissue, a synergistic effect of two therapeutic modalities, interference with normal reparative processes by the combination, or an enhancement of retained serum protein-bound breakdown products of BCNU.

Estradiol-17 beta interference with meiotic maturation in Rana pipiens ovarian follicles: evidence for inhibition of 3 beta-hydroxysteroid dehydrogenase.

Increasing doses of estradiol-17 beta added to in vitro incubations inhibited pregnenolone-induced germinal vesicle breakdown in Rana pipiens ovarian follicles. The inhibition was reversed with increasing concentrations or pregnenolone added to the medium. Because no evidence of estradiol-17 beta inhibition or interaction with progesterone-induced GVBD was observed, the effect of estradiol-17 beta on the conversion of 3H-pregnenolone to 3H-progesterone was investigated. Estradiol-17 beta in doses as low as 10(-7) M significantly inhibited the conversion of 3H-pregnenolone to 3H-progesterone in follicles incubated in vitro. It is suggested that estradiol-17 beta is a feedback inhibitor of 3 beta-hydroxysteroid dehydrogenase-isomerase, the enzyme complex that converts pregnenolone to progesterone, a necessary step in the initiation of GVBD.

Deposition of IgM and complement at the dermoepidermal junction in acute and chronic cutaneous graft-vs-host disease in man.

The presence of cutaneous immunoglobulin and complement was investigated in 88 patients with and without graft-vs-host disease (GVHD) after transplantation of bone marrow from HLA identical siblings for the treatment of acute leukemia or aplastic anemia. For comparison, skin biopsies from the patients obtained before transplantation, from 58 healthy individuals (mostly marrow donors) and from four syngeneic marrow recipients were studied. A direct immunfluorescent staining technique was used. Dermo-epidermal IgM deposits were found in 11% of healthy individuals and patients before grafting but were present in 86% of patients with chronic and 39% of patients with acute GVHD. Patients with allogeneic grafts who never had GVHD or who had recovered from it and patients with syngeneic grafts showed findings not different from those in healthy individuals. Findings similar to those with IgM, although less striking, were made for C3, i.e., patients who had chronic or acute GVHD had a high incidence and intensity of C3 deposits at the dermo-epidermal junction. This observation raises the possibility that humoral immunity is involved in the development of GVHD.

Purification of methylmalonyl-CoA mutase from Propionibacterium shermanii using affinity chromatography.

A novel procedure for the purification of methylmalonyl-CA mutase from Propionibacterium shermanii has been described which employs affinity chromatography on a column of immobilized vitamin B-12 linked covalently to Sepharose. The method has the advantage of being simple and rapid, thus enabling the purification of the enzyme to near homogeneity with good yields.

Factors influencing nursing in Macaca fascicularis.

Nursing by two mother-infant pairs in a caged colony of Macaca fascicularis was monitored at 1-min intervals for 8 h beginning 8:30 a.m. BST, once a week for 3 months in the summer of 1973. Nursing occupied about 210 min in 8 daylight hours for the infants at 10 weeks of age, and the time spent nursing decreased at the average rate of 9.4 min per week until the infants were about 6 months old. The time spent nursing by the infants studied here resembles closely the times spent nursing by some other macaques and by baboons. In the course of a day the amount of time spent nursing varies significantly with a diurnal peak. If nursing by one mother-infant pair is independent of nursing by the other pair, then the time the two pairs spend nursing together would be a function of the product of the frequencies of nursing by each pair. The expected times for the pairs nursing together based on the hypothesis of independent events were significantly less than the observed times the pairs nursed together. Nursing, therefore, involves a positive influence or imitation of one nursing pair by the other. Nursing sessions involving both mother-infant pairs were longer on the average than sessions involving only one pair.

Maternal-fetal mixed lymphocyte reactivity in pre-eclampsia.

Two-way mixed lymphocyte reactivity (2-way-MLR) between maternal and fetal lymphocytes in vitro, and one-way mixed lymphocyte reactivity (1-way-MLR) of maternal lymphocytes (responders) to stimulation by mitomycin-treated fetal lymphocytes (stimulators), were investigated in normal and pre-eclamptic pregnancies. No relationship was observed between pre-eclampsia and the 2-way-MLR. The 1-way-MLR was lowest in normal pregnancy, higher in mild pre-eclampsia, and highest in severe pre-eclampsia, although none of these differences were statistically significant.
