Regional localization of polymorphic markers on chromosome 10 by physical and genetic mapping.

The human vimentin gene and a random DNA segment (D10S39) were mapped to the short arm of human chromosome 10 by linkage analysis. A panel of somatic cell hybrids and monosomy cell-lines, which divide chromosome 10 into seven regions, was used to localize 10 polymorphic markers on this chromosome. The physical map locations obtained correlate well with linkage maps of chromosome 10. Two markers which have been shown to be closely linked to the gene for multiple endocrine neoplasia type 2A map distal to a translocation breakpoint in band 10q11.2.

Efficacy of concomitant early summer treatment with fenbendazole and clorsulon against Fasciola hepatica and gastrointestinal nematodes in calves in Louisiana.

The efficacy, safety, and compatibility of fenbendazole (FBZ) and clorsulon (CLN) were tested after oral administration of label recommended and of higher (5x) dosage rates to calves naturally infected with gastrointestinal nematodes and Fasciola hepatica. Results for 42 calves allotted to 4 treatment groups indicated a similar efficacy against mature F hepatica by FBZ (5 mg/kg of body weight) and CLN (7 mg/kg) in a combined oral suspension, compared with CLN (7 mg/kg) alone (100 vs 99% reduction). A lesser efficacy was observed against immature flukes (88.6 and 84.9% reduction, respectively). Calves given 25 mg of FBZ/kg and 35 mg of CLN/kg had nearly complete reduction of both mature (99.6%) and immature flukes (99.1%). Fasciola egg counts were reduced by greater than 99.5% in all treated groups. Against Ostertagia ostertagi, the percentage of efficacy of the combined FBZ (5 mg/kg) and CLN (7 mg/kg) treatment was 94.3% against adults and 81.3% against inhibited larvae. Efficacy against all other nematodes was 100%, except against Cooperia spp adults (98.3%) and immature Oesaphagostomum radiatum (88.0%). At 5 x dosage rates for FBZ and CLN, percentage of removal of adults and inhibited larvae of O ostertagi was 99.3 and 99.0%, respectively, and 99 to 100% for other nematodes. Results indicate that FBZ and CLN are compatible when mixed together and administered as an oral suspension to cattle and that the efficacy is similar to that of the drugs individually. On the basis of further results, we suggest that summer treatment may be superior in preventive value for gastrointestinal nematodes and F hepatica, compared with spring treatment, because of seasonal infection dynamics of the major cattle parasites in Louisiana.

Lack of correlation between HLA types and anti-idiotypic production in family members of a lupus patient.

Correlations of anti-single-stranded (ss) DNA, anti-F(ab')2, and anti-idiotypes to HLA types of 16 healthy family members of a lupus patient were studied. High levels of anti-ss DNA (63%) and anti-F(ab')2 (69%) were detected. Of the 12 family members who expressed HLA-DR2 antigen, 8 had anti-ss DNA and anti-F(ab')2 antibodies. One out of 3 family members who shared the same HLA phenotypes, A1B8DR2, of the proband had high levels of anti-idiotype directed against the proband's F(ab')2 anti-DNA. Though a high prevalence of A1B8DR2, of anti-ss DNA, and of anti-F(ab')2 in healthy family members of a lupus patient was found, anti-idiotypes against anti-DNA were not dependent on HLA-A, B, Dr.

Ovarian cysts decrease the success of controlled ovarian stimulation and in vitro fertilization.

Forty cystic structures, 16 to 60 mm, of probable ovarian origin were found in 126 cycles during an ultrasound scan on day 3 of a cycle in which in vitro fertilization was planned. The response of these patients to exogenous gonadotropin stimulation, oocyte capture, fertilization, cleavage, and pregnancy was studied. Patients with cystic change had significantly lower peak estradiol (E2) levels. Pregnancy rates were lower, but not significantly so. Those patients with structures 16 to 29 mm had more cycles canceled due to precipitous drops in E2, consistent with an untimely surge of luteinizing hormone. Patients with structures 30 to 60 mm had increased numbers of cycles with poor response to stimulation. The importance of an early follicular phase ultrasound scan is stressed, and the management of ovarian cysts in this patient population is discussed.

Le Fort I maxillary osteotomies in cleft palate cases. Surgical changes and stability.

Surgical and postsurgical changes in maxillary position following transpalatal osteotomy at the le Fort I level in 30 patients with clefts of lip and palate were evaluated cephalometrically. By superimposing on natural reference structures, a more accurate and detailed evaluation of change in maxillary position was possible than by using conventional cephalometric analyses. The mean horizontal advancement was 9 mm, with a mean vertical change of 3 mm. While it is customary to express postsurgical relapse as a percentage of surgical change (in this case 7% mean horizontal relapse and 23% mean vertical relapse), the most remarkable finding to emerge from this study was the variability in surgical and postsurgical change which would be obscured by concentrating on descriptive statistics. Postsurgical change was related to the amount of surgical change, but the correlation coefficients are quite low, and so other factors must be responsible for a significant proportion of any relapse.

High frequency of benign mediastinal uptake of gallium-67 after completion of chemotherapy in children with high-grade non-Hodgkin's lymphoma.

We observed increased gallium-67 uptake in the mediastinum after completion of chemotherapy in 10 of 62 patients with non-Hodgkin's lymphoma. All 10 were under 15 years of age, yielding a frequency of 43% in this age group. The interval between cessation of chemotherapy and the development of increased gallium-67 uptake ranged from 1 to 8 months, and the abnormality persisted for 2 to 59 months. Serial chest x-rays were performed in all patients, and four of the 10 had transient widening of the mediastinum that remained within normal limits for the children's ages. Three patients had chest computed tomographic (CT) scans at the time of increased gallium-67 uptake, and one of the three had serial scans that showed a mediastinal mass consistent with thymic enlargement. All of the patients were asymptomatic and none were biopsied. All 10 remained well, with a mean follow-up of 52.5 months. The phenomenon we describe is probably due to "rebound" thymic hyperplasia, which is a benign and transient condition. We conclude that abnormal mediastinal gallium-67 uptake after completion of chemotherapy is likely to be benign and transient if the patients are young, have small non-cleaved-cell histology, are without other evidence of lymphoma recurrence, and do not have initial mediastinal involvement. Progressive widening of the mediastinum on chest x-ray is cause for suspicion and requires further evaluation. Serum lactate dehydrogenase (LDH) levels may not be helpful in differentiating a benign from a malignant process.

The principles of bony spinal fusion.

Bony fusion is frequently required for anatomical reconstruction and stabilization of the spine. This review discusses bone structure, healing after fracture, and bony fusion, including the use of autografts and allografts. Research should be done on preparation of the bed, the nature and quantity of bone to be used, and how best to immobilize the spine.

Evaluating the success of deaf parents.

Nineteen parent-child pairs completed the Parental Strengths and Needs Inventory (PSNI) and were interviewed about their relationships. Parent and child scores on the PSNI were above average, with parents expressing high interest in information about childrearing. Interviews revealed issues specific to deaf parents, such as using children as interpreters and frustration when children had poor sign language skills. Children with good sign language skills said they were generally willing to interpret for their parents, except during conflict situations. Children also reported advantages to having deaf parents: they enjoyed being bilingual and their parents tolerated noise and loud music. Study results underscore the importance of triangulation strategies in cross-cultural research.

Peripheral serotoninergic receptor sensitivity in senile dementia of the Alzheimer type.

Platelet aggregation induced by 5-hydroxytryptamine (5-HT) was used as a measure of the functional responsiveness of peripheral 5-HT receptors in controls and drug-free patients suffering from senile dementia of Alzheimer type (SDAT). No significantly different aggregatory response (5-HT/ADP ratio) was noted between the two groups of subjects. If the receptor mediating this response is a 5-HT2 receptor, there is no evidence to suggest that the functional activity of this system is abnormal in SDAT.

Acute myocardial infarction after a wasp sting.

A 56 year old man with ischaemic heart disease and known allergy to wasp venom was stung in the mouth by a wasp and within four hours sustained an acute myocardial infarction. This complication has been described on only three previous occasions. Possible pathogenic mechanisms include the anaphylactic reaction itself, the action of wasp venom constituents, and therapeutic intervention with adrenaline.

The midline cartilages of the cranial base of the hypopituitary dwarf mouse (dw/dw). A histological and autoradiographic study.

The dwarf mice used in this study demonstrated severe growth retardation related to the 5 endochondral growth sites of the midline cranial base. The principal defect appears to be a dramatic reduction of activity as judged by comparing the histology of the dwarf mice with their normal litter mates. The principal defect appears, from the [3H]thymidine data, to be a complete cessation of mitotic activity in the proliferative zone. It is not possible, from the present study, to say whether activity in the matrixogenic zone is also reduced. The anomalous enlargement of hypertrophic cells by 80 days in dwarf mice is unexplained.

Effect of enhanced macrophage function on early wound healing.

Although the macrophage is important to wound healing, research has focused on its relationship to fibroblast and collagen synthesis. This study was designed to assess effects of enhanced macrophage function on early wound healing, before established collagen synthesis. Sprague-Dawley rats had dorsal incisions after one of three treatment regimens: (1) saline solution, 0.5 ml administered intravenously, (2) intravenous glucan, a macrophage stimulant, 20 mg; (3) topical glucan, 20 mg. Intravenous therapy was administered 24 hours before and after incision. Breaking strength was significantly increased (p less than 0.01) by both intravenous glucan (49.8 +/- 5.5 gm) and topical glucan (59.7 +/- 5.6 gm) on the fourth day after incision, compared with controls (22.0 +/- 2.6 gm). Similar results occurred on the seventh day after incision. Although formalin fixation significantly enhanced breaking strength in fresh control wounds (22.0 +/- 2.6 vs 39.5 +/- 2.2 gm), no increase occurred in wounds treated with intravenous glucan (49.8 +/- 5.0 vs 55.3 +/- 6.4 gm), indicating maximal cross-linking of collagen. Collagen synthesis, reflected by tritiated proline uptake, was no different in control versus glucan groups. Supernatants from control or glucan-activated macrophages were injected intraperitoneally or applied topically in the rat model. Activated supernatant, both intraperitoneal and topical, resulted in increased breaking strength on the fourth day after incision. Formalin fixation did not increase breaking strength in the activated supernatant groups. We conclude that enhanced macrophage function increases early wound breaking strength. This effect appears unrelated to collagen synthesis but may be related to increased cross-linking of collagen. Similar effects are seen with activated macrophage secretory products administered intraperitoneally or topically.

Protective effect of glucan-enhanced macrophage function in experimental pancreatitis.

In an effort to assess the impact of enhanced macrophage function in acute pancreatitis, mice were subjected to a choline-deficient diet supplemented by ethionine to induce necrotizing pancreatitis. Treatment with the macrophage stimulant glucan resulted in improved survival rates (58 percent versus 14 percent) and maintenance of pancreatic architecture. Glucan treatment also resulted in decreased plasma and peritoneal trypsin activity, as well as increased trypsin-binding activity in the blood and peritoneal cavity. Plasma interleukin-1, as well as macrophage production of interleukin-1, were increased in the glucan-treated mice, which indicated enhanced macrophage function. These composite findings suggest that by enhancing diverse aspects of reticuloendothelial function, clinical use of immunomodulators may have significant impact on the pathogenesis of acute pancreatitis.
