Complete sequence of the Rous sarcoma virus env gene: identification of structural and functional regions of its product.

The amino-terminal amino acid sequences of gp85 and gp37, the envelope glycoproteins of Rous sarcoma virus (RSV), were determined. Alignment of these sequences with the amino acid sequence predicted from the complete nucleotide sequence of the Prague strain of RSV, subgroup C (PR-C), has allowed us to delineate the env gene-coding region of this virus. The coding sequences for gp85 and gp37 have been placed in an open reading frame that extends from nucleotide 5045 to nucleotide 6862 and predict sizes of 341 amino acids (36,962 molecular weight) for gp85 and 198 amino acids (21,566 molecular weight) for gp37. Carbohydrate makes a significant contribution to the observed molecular weights of these polypeptides--the amino acid sequence contains 14 potential glycosylation sites (Asn-X-Ser/Thr) in gp85 and two in gp37. Experiments aimed at estimating the number of carbohydrate side chains yielded results consistent with most or all of these sites being occupied. Although an initiation codon is located early (codon 4) in the open reading frame, it is likely that splicing yields an mRNA on which translation initiates at the same AUG as that of the gag gene to produce a nascent polypeptide in which gp85 is preceded by a 62-amino-acid-long leader peptide. This leader contains the hydrophobic sequence (signal sequence) necessary for translocation across the endoplasmic reticulum and is completely removed from the env gene product during translation. The polyprotein precursor, Pr95env, is cleaved to gp85 and gp37 at the carboxyl side of the basic sequence:-Arg-Arg-Lys-Arg-. gp85 is attached through a disulphide linkage to gp37, and although the positions of the cysteines involved in this linkage are not known, the presence of a 27-amino-acid-long hydrophobic region at the carboxy-terminus of gp37 is consistent with its role as a membrane anchor for the viral glycoprotein complex. The location of host range variable regions with respect to the possible tertiary structure of the complex is discussed.

Mortality of preoperative peritonitis in newborn infants without intestinal obstruction.

Within a period of 15 years 649 neonates were subjected to laparotomy; 60 (9%) of these patients died. Eighty-seven of the patients had a peritonitis already preoperatively without intestinal obstruction. Many of these were cases of ruptured omphaloceles or gastroschisis. In 17 infants a spontaneous intestinal perforation was the cause of the peritonitis. In 13 there was a preexisting meconium peritonitis. Seven children suffered from gangrenous intestine. Further causes for preoperative peritonitis were a complicated enteritis in 7 and a perforated appendix in 2 cases. Twenty-five or 29% of the children died. The highest mortality was found in children with ruptured omphalocele. It was 50% followed by gastroschisis with 36%. The mortality in patients with spontaneous intestinal perforation was rather similar, and the same high fatality rate was observed in infants with gangrenous intestine. In 28% of the children no cause for the peritonitis could be discovered. The high mortality rate is primarily due to the infants' bad general condition, i.e., low birth weight, prematurity and additional severe malformations.

Intravital microscopical studies of the tubular urine flow in the conscious rat.

A method was developed which allowed microscopical observation of the kidney in conscious and anesthetized rats. The left kidney was freed from surrounding tissue and placed in a plexiglas chamber with a protecting window which was sewed into the skin of left flank. During the experiments the covering glass was removed. Tubular transit time was measured in the conscious state and during inactin anesthesia in control rats as well as in unilaterally nephrectomized rats with compensatory hypertrophy. Transit time of lissamine green within proximal convolutions and Henle's loop increased significantly in both series during anesthesia. Luminal and outer diameters of proximal tubules in control rats were evaluated from microphotogramms of the kidney surface, they did not change due to anesthesia. The results could be mainly explained by a decrease in glomerular filtration rate due to anesthesia. Possible mechanisms are discussed which could explain the observed transit time in Henle's loops and distal convolutions in control and uninephrectomized rats.

Myofeedback for muscle retraining in hemiplegic patients.

The use of audiovisual displays of myoelectric potentials (myofeedback) has been suggested for retraining and strengthening of paretic muscles in patients with hemiplegia from stroke. This controlled study examines the specificity and efficiency of myofeedback in the strengthening of paretic muscles in hemiplegic patients. Eighteen patients with trace to fair-minus grade of deltoid muscle power resulting from strokes that had occurred at least six weeks earlier were randomly assigned to the six possible orders of accurate (true), positive noncontingent (placebo) and no feedback conditions. On three successive days, each patient received one session of each feedback condition, consisting of 20 isometric contractions of five seconds each with ten seconds of intervening rests. In addition, each patient's motivational level was rated. Averaged myoelectric potentials were quantified for each contraction for analysis. When the data were analyzed without grouping, no statistical differences among the three feedback conditions were noted. However, when the subjects were grouped according to age and motivational level, during true and no feedback sessions, the older and the poorly motivated groups were found to show progressive increment of myoelectric output, whereas the younger and the better motivated showed the opposite results. With placebo feedback, both groups showed progressive decrement of myoelectric output. It is concluded that the effect of myofeedback is nonspecific at least in its short-term application for the retraining of hemiplegically paretic muscles.
