The influence of embryonal bursectomy on benzpyrene-induced sarcoma of the chicken.

160 chickens hormonally bursectomized before hatching, and 160 controls were twice given a dose of 10 mg 3,4-benzo(a)pyrene 2 weeks and 6 weeks after hatching. After 24 weeks the controls had developed muscle sarcomas at a significantly higher rate than the bursectomized animals (49.6--32.1%). The tumors were identified as malignant polymorphocellular rhabdomyosarcomas in light and electron microscopy. When the experiment was stopped after 27 weeks, the controls showed more tumors than the bursectomized chickens (65.1% versus 58.9%). The effect of bursectomy lies in a prolongation fo tumor latency. The frequency of metastases is equally decreased to a significant degree (41.7% versus 21.7%). These results are interpreted as an illustration of the disturbed equilibrium between the T and B cell responses.

A possible reason for the phalloidin tolerance of hepatoma cells.

In contrast to normal liver cells, AS-30D hepatoma cells are insensitive to phalloidin. The lack of the typical phalloidin response in the latter cells is not due to a deficiency of contractile proteins. Actin prepared from hepatoma cells is able to form filamentous structures and is stabilized in a manner similar to muscle actin. Isolated liver cells were exposed to a medium containing phalloidin and removed after 20 min by centrifugation. The supernatant was incubated again with fresh cells. The procedure was repeated four times. The phalloidin response decreased to about 19% of the control because of the uptake of phalloidin during each incubation. When the same procedure was carried out with AS-30D hepatoma cells, and aliquots of the supernatants were tested with hepatocytes no marked decrease of the phalloidin response was seen. This indicates that hepatoma cells do not consume the toxin as do normal liver cells.

Inhibitory effects of 4,4'-diisothiocyano stilbene-2,2'-disulfonic acid (DIDS) in the response of isolated hepatocytes to phalloidin.

4,4'-Diisothiocyano stilbene-2,2'-disulfonic acid (DIDS) inhibits the typical development of protrusions, regularly seen after treatment of isolated hepatocytes with phalloidin. The degree of inhibition depends on the time of preincubation and on the concentration of DIDS, but not on the concentration of phalloidin. DIDS is more effective than H2DIDS. The inhibition by both compounds is irreversible. The binding capacity of hepatocytes for H2DIDS is much higher than that of the phalloidin-insensitive hepatoma cells. Gel electrophoresis of lysates from cells, pretreated with 3H2DIDS demonstrates that actin binds very little of the inhibitor. Our results suggest that a protein structure on the surface of hepatocytes, needed for the response to phalloidin, is influenced by DIDS or H2DIDS.

Histochemical identification of T and B areas in paraffin-embedded lymphoid tissues by demonstration of alpha-naphthyl acetate esterase (ANAE) activity.

A strong activity of acid alpha-naphthyl acetate esterase (ANAE) in T lymphocytes and macrophages within mouse lymphoid organs is demonstrable even after paraffin-embedding, if a rapid embedding procedure with acetone as dehydrating agent is used. The lymphoid tissue structure is better preserved in paraffin sections procured after this rapid embedding procedure than in frozen sections commonly used for histochemical demonstration of ANAE. In the paraffin sections the pattern of the ANAE reaction allows a clear delineation of the T and B areas in the lymphoid organs, and the intracellular location of the focal ANAE activity in T lymphocytes is readily discernible.

[Experience in application of electronic data processing in routine medical microbiology (author's transl)].

Experiences in a two years application of electronic data processing in medical routine microbiology are reported. Optical mark reader forms serve as request forms as well as workprotocol. On the left two thirds of these forms (Fig. 1 and 2) the patients data including clinical statements, and the kind of specimen to be investigated (sputum, smear etc.) as well as results not essential for the printout of laboratory reports are filled in. On the right third of the optical mark reader form, in which area the Bell & Howell document reader can read pencil marks, the patients identification number, the tests requested and all results are marked right at the work bench. As shown in the flow diagram (Fig. 3) after completion of the investigation the date on the mark reader forms are read mechanically and the test results are printed (Fig. 5) by means of the computer. In addition, the data processing system, which proved to be very flexible, performs all necessary types of administrative work as filing of laboratory data in a coded form on cards for permanent storage (Fig. 7) statistics and the printout of bills (Fig. 6). The processing of routine microbiology date is achieved by a Siemens DVA 404/3 (64 kbt) connected for magnetic disc device (2993 Mio bytes), and a line printer (60,000 lines per hour) and dialog display monitors, which are used simultaneously for the on line and off line handling of all the data in clinical chemistry-, serology- and hematology-departments of the same institute.

[Halothane and antituberculous drugs--a hepatotoxic combination? (author's transl)].

A case report is given on two patients receiving halothane anesthesia while beeing treated with isoniacid, ethambutol and rifampicin. Following halothane anesthesia, both patients developed a severe liver disease with encephalopathy grade III. We observed a moderate increase of bilirubin and SGOT and a more severe increase of serum ammonia. Histologically, both patients had alterations compatible with drug hepatitis. Within 14 days remission occurred spontaneously. The two case reports do not fit with typical isoniacid hepatitis or typical halothane hepatitis. The possibility of combined drug toxicity on liver during halothane and isocianid treatment is discussed.

Malignant nephrosclerosis during pregnancy and in the postpartum period (the uremic hemolytic syndrome).

Histologic, immunohistologic, and ultrastructural features are presented of two cases with malignant nephrosclerosis during pregnancy. Primary malignant nephrosclerosis emerges as a clinical entity which can be distinguished from toxemia of pregnancy in the midtrimester and post partum. The first description of malignant nephrosclerosis dates from 40 years ago, but only a few cases were reported associated with pregnancy. Although disseminated intravascular coagulation seems involved, the morphology is different from that of toxemia. Malignant nephrosclerosis reveals a close similarity to the hemolytic uremic syndrome. Early diagnosis by renal biopsy and proper treatment may prevent a lethal outcome due to progressive failure.

Development of tumors in the glandular stomach of rats after oral administration of carcinogens. I. Histological findings.

Experimental carcinomas in the glandular stomach of rats were induced by oral administration of MNNG (M-methyl-N'-nitro-N-nitrosoguanidin) for 35 weeks or ENNG (N-ethyl-N'-nitro-N-nitrosoguanidin) for 20 weeks. Rats were killed at different times after beginning of carcinogen treatment and tissue specimens were prepared for histologic investigation. Particular interest was placed on the development of tumors and on pathological findings possibly contributing to early diagnosis of stomach cancer. During the development of tumors, several dysplastic reactions were observed in the antral mucosa. They could be classified into 4 groups: One was regenerative hyperplasia (1) that meant irregular glandular proliferations without cell atypism at the margin of erosions and ulcers. This lesion was mainly found 1-9 weeks after administration of MNNG. In glandular hyperplasia (2) either crypts or glands were extended and mucosal layers were thickened. No signs of cell atypism were observed. This lesion was mainly found 12-17 weeks after administration of MNNG. Dysplasia (3) was combined with considerable structural modifications and cellular atypism. However, this lesion was limited to the mucosal layer. Neoplastic changes (4) were characterized by marked cellular atypism and extension to tunica submucosa and tunica serosa. Some tumors showed the histological patterns of benign tumors, but most of them were adenocarcinomas. In some cases metastases into pancreas, liver and lymph nodes and in one case into the 12th rib were observed. No particular enzyme patterns were found by histochemistry.

Development of tumors in the glandular stomach of rats after oral administration of carcinogens. II. Different cell types in antral carcinoma as revealed by electron microscopy.

In the present study electron microscopic investigations of tumors of the glandular stomach of rats induced by oral administration of MNNG or ENNG are dealt with. The aim of the study was to describe the different cell types found in the carcinoma and to elucidate the possible relationship between the undifferentiated carcinoma cells and the more differentiated cell types found in the tumor. The results show that besides undifferentiaded carcinoma cells, several differentiated cell types such as goblet cells, endocrine cells, cells with lamellated inclusions in their cytoplasm, and squamous carcinoma cells can be observed. The most conspicuous findings were carcinomatous gland cells with well differentiated microvilli on their luminal surface and typical tonofilamentous structures in their cytoplasm. These cells exhibited signs of the squamous epithelium and of gland cells. Therefore they may be considered as intermediate variants between adenocarcinoma cells and squamous carcinoma cells. The occurrence of such intermediate steps points to the possibility of differentiation by cell division of adenocarcinoma cells into several metaplastic cell types in the experimental stomach carcinoma induced by MNNG or ENNG.

[The so-called spontaneous nodular panniculitis (Weber-Christian lipodystrophy)].

Enlarged lymph nodes and a tumor, the size of a man's fist, were found in the abdomen of a female patient of 63. Histologic diagnosis determined a non-suppurative panniculitis with typical proliferation of foam cells and reactive inflammatory infiltration attaining the stage of granulomatosis. The disease is seen as granulomatous histiocytosis; a sure explanation of its pathogenesis is not yet possible.
