Differential sensitivity of T and B cell areas in mouse lymph nodes to exogenous cortisol. A histomorphometrical study.

Intraperitoneal injection of a single high dose of cortisol acetate into young adult BALB/c mice resulted in severe prolonged involution of the axillary lymph nodes, which started to regenerate only three weeks later. Histological and histomorphometrical analysis revealed that cortisol-induced lymphocyte depletion and diminution of lymph nodes was more pronounced in its B cell areas than in the paracortical T cell areas. Lymphocyte-depleted areas in the outer cortex i.e. in the B cell compartment, were characterized by a persisting diffuse reticular framework. Lymphocyte depletion in the paracortex resulted in partial transformation of its normal diffuse structure into "paracortical cords" with intervening patent sinuses. It was concluded that 1) T cell areas in mouse lymph nodes are more resistant to exogenous cortisol than B cell areas, and 2) paracortical cords may be interpreted as basic tissue units of the paracortical area in the mouse lymph node.

Precancerous conditions and epithelial dysplasia in the stomach.

A distinction can be made between a precancerous condition and a precancerous lesion. The former is a clinical state associated with a significantly increased risk of cancer, whereas a precancerous lesion is a histopathological abnormality in which cancer is more likely to occur than in its apparently normal counterpart. Up to the present time atrophic gastritis, gastric ulcer, pernicious anaemia, gastric stumps, gastric polyps, and Ménétrier's disease have all been considered as precancerous conditions and lesions of the stomach. Of these, only atrophic gastritis, pernicious anaemia, gastric stumps, and certain types of gastric polyp can now be regarded as having any really significant malignant potential. The precancerous lesion common to these is epithelial dysplasia which can occur in ordinary (foveolar) gastric epithelium as well as in intestinal metaplasia. The criteria for grading dysplasia in gastric epithelium into mild, moderate, and severe grades are given, and attention is drawn to the problems of differentiating inflammatory or regenerative change from mild dysplasia and intramucosal carcinoma from severe dysplasia. The clinical and epidemiological implications of gastric dysplasia are discussed with suggestions for further research.

BCG-induced changes in size of thymic cortex and thymus-dependent areas in spleen and lymph nodes of mice.

Significant changes in the size of thymic cortex and of thymus-dependent areas (TDA) in spleen and lymph nodes, were revealed by histomorphometry after intraperitoneal injection of BCG into young-adult BALB/c mice. Reactions of the thymic cortex were found to be biphasic with initial involution and subsequent hyperplasia. TDA in the spleen showed a transient involution with almost complete regeneration at the end of the experiment. The response of TDA in lymph nodes was characterized by pronounced and persistent hyperplasia. The functional significance of these BCG-induced changes of lymphoid tissue compartments is discussed.

Ultrastructure of telangiectatic osteosarcoma.

Recent investigations have shown that telangiectatic osteosarcoma has a poorer prognosis than other osteosarcomas. To elucidate the histogenesis of TOS two cases were investigated on the electron microscopic level. The results show that besides anaplastic, osteoblast-like, and fibroblast-like tumor cells angiosarcomatous components can be observed in this malignant bone tumor, which are characterized by endothelial cells with pinocytotic vesicles, tight intercellular junctions, fine fibrils, and so-called Weibel-Palade bodies in their cytoplasm. From these results, it is concluded that telangiectatic osteosarcoma is derived from multipotent mesenchymal cells with potential differentiation into various directions, such as osteoblast-like cells, and fibroblast-like cells.

Hepatic failure in a patient treated with dacarbazine (DTIC) for malignant melanoma.

A 55-year-old white woman received chemotherapy with DTIC after surgery for malignant melanoma (stage I, SSM IV, depth of invasion 12 mm). She died suddenly during the second treatment cycle. Autopsy revealed massive necrosis of the liver and thrombosis of the hepatic veins. The cause of the fatal outcome is attributed to the adverse toxic effects of DTIC.

Decreased sensitivity of isolated hepatocytes from baby rats, from regenerating and from poisoned livers to phalloidin.

Isolated hepatocytes, prepared from 5 day old rats, from regenerating livers of from livers after poisoning with carbon tetrachloride, are less sensitive to phalloidin in vitro than hepatocytes from untreated adult controls. The time course of the reduced susceptibility to phalloidin was compared with the ability of hepatocytes to take up bile acids under various conditions. SDS-electrophoresis of cell lysates gave no evidence for decreased levels of actin in cells with reduced sensitivity to phalloidin. In contrast, there was a good relationship between the active uptake of bile acids and the sensitivity of hepatocytes to phalloidin. The decreased response of hepatocytes from baby rats, from regenerating livers or from poisoned livers to phalloidin is more probably related to differences in phalloidin uptake than to a reduced endowment with microfilamentous structures.

Quantification of BCG-induced reactions of T and B areas in peripheral lymphoid organs of young adult BALB/c mice. A histomorphometrical and autoradiographical study.

Significant structural alterations of the axillary lymph nodes and spleens were revealed by histomorphometry following intraperitoneal injection of BCG into young adult BALB/c mice. The relative proportions of the germinal center areas were increased in lymph nodes as well as in spleens. The overall proportion of the B cell areas, however, showed a significant increase only in the spleens, compared to a significant reduction in the lymph nodes. By contrast, T cell areas appeared significantly reduced in the spleens, while their size was almost doubled in the lymph nodes. The labeling index in the lymph node paracortical areas was also doubled autoradiographically. The structural changes in the lymph nodes and spleens differed not only in quality, but also in their time course. In lymph nodes, the altered proportions of the T and B cell areas persisted from the second through the sixth week after BCG treatment. By contrast, structural changes in the spleen began to subside 2 weeks after treatment, and after 6 weeks the proportions of the T and B cell areas in the spleen had returned to normal. These quantitative morphologic findings suggest a difference in the functional roles of lymph nodes and spleen in the systemic immune response to BCG.

Comparative histologic, histochemical, and histomorphometric studies of T and B cell areas in peripheral lymphoid organs of normal young adult BALB/C mice.

T cell areas can be distinguished from adjacent B cell compartments in routinely stained histologic sections of paraffin-embedded mouse lymph nodes and spleens by their comparatively loose texture. The reliability of this morphologic criterion for identifying T cell areas has been proven in comparative histologic, enzyme-histochemical, and immunohistochemical investigations. It is thus possible to determine the size of functionally different compartments in the lymph nodes and spleen of the mouse with sufficient accuracy by the use of conventional histologic techniques. According to our subsequent histomorphometrical studies in two groups of inbred mice matched with respect to age and strain, the proportions of B cell areas in the peripheral lymphoid organs varied significantly depending on environmental conditions, whereas the proportions of T cell areas were stable.

Multifocal early gastric cancer of mixed type.

A report of three selected gastric resection specimens presents the problems of histological classification of individual early gastric cancer, illustrated in discrepancies between macroscopic or endoscopic, and histologic identification. Multifocal origin and different types of gastric cancer are demonstrated in all three cases. Problems related to exact histologic classification of this mixed type are discussed together with the prognosis and surgical consequences.

Malignant fibrous histiocytoma of bone and osteosarcoma. A comparative light and electron microscopic study.

Malignant fibrous histiocytomas are well-described tumors of the soft tissues. Recent investigations have shown that malignant histiocytoma may also occur as a primary bone tumor. However, difficulties may arise to distinguish malignant histiocytoma of bone from other malignant bone tumors, such as osteosarcoma. In the present study, the ultrastructure of five cases of malignant fibrous histiocytoma of bone is compared with that of osteosarcoma. The results show that malignant fibrous histiocytoma is composed mainly of histiocytic cells and fibroblastic cells. In addition, xanthomatous cells, undifferentiated cells, and giant cells may be observed. By contrast, the predominant cell type in osteosarcoma is the neoplastic osteoblast, characterized by abundant rough endoplasmic reticulum. Signs of matrix calcification in the intercellular matrix between the collagen fibrils are regularly observed in osteosarcoma, but not in malignant histiocytoma. From these results it is concluded that the ultrastructure of malignant fibrous histiocytoma arising in bone is morphologically identical with the soft tissue counterpart of this tumor. The components of the tumor are derived from neoplastic histiocytes. This cytogenesis differs from that of osteosarcoma, which is derived from neoplastic osteoblasts. Therefore, from the ultrastructural point of view, malignant fibrous histiocytoma of bone should be accepted as a distinct histologic entity among bone tumors.
