Cadmium metabolism in cdm/cdm mice.

The uptake and metabolism of cadmium by cadmium-susceptible (+/+) and cadmium-resistant (cdm/cdm) strains of mice have been compared. These strains did not differ with respect to the quantitative uptake of cadmium into liver, kidney, or testis. After intraperitoneal administration of a nontoxic dose, more than 80% of the cadmium in liver and testis of both strains is bound to low molecular weight proteins. The chromatographic behavior of these cadmium-binding proteins is not affected by cdm genotype.

Ntau-methylhistidine in human skeletal and smooth muscle proteins.

1. Fifteen pieces of rectus abdominus muscle and fifteen pieces of taenia muscle were removed from patients undergoing various surgical procedures. 2. The muscles were extracted, hydrolysed and the content of Ntau-methylhistidine was measured. 3. The rectus muscles contained 3.13 +/- 0.7 mumols Ntau-methylhistidine/g fat-free dry solid and the taenia muscles 2.4 +/- 0.6 mumols/g fat-free dry solid. There was a statistically significant difference (P less than 0.05) between these values using the Student's unpaired tau test, although this could have been due to contamination of the taenia muscles with connective tissue. 4. The muscle content of Ntau-methylhistidine was at least 40% higher than the only other reported value and therefore the method of calculating muscle protein breakdown based on Ntau-methylhistidine excretion requires revision.

The effect of elective surgery on 3-methylhistidine excretion.

Elective surgical operation in normally nourished patients is associated with an increase of up to 40 per cent in the urinary excretion of 3-methylhistidine during the first three postoperative days. This increase was statistically significant (P less than 0.01) on the second and third postoperative days. This probably represents a minor degree of increased muscle proteolysis, principally due to the operative trauma. The possible effect of postoperative starvation requires further elucidation.

Assessment of the nutritional state of patients with an intestinal fistula.

The arm muscle area, derived fat-free mass, urinary excretion of 3-methylhistidine and creatinine were measured serially in a group of patients with an intestinal fistula receiving prolonged nutritional support. While the two anthropometric tests appeared reasonable indicators of changes in muscle mass in these patients, the marked intra-individual variability in excretion of 3-methylhistidine and creatinine created problems in interpretation. It is suggested that more use be made of serial anthropometry in district general hospitals as an alternative, simple, measure of nutritional status.

Severe chronic intra-abdominal sepsis: a report of 3 cases and a review of patient management.

The management of three cases (one fatal) with extensive intraabdominal sepsis, is described. Management included metabolic and fluid control, intravenous nutrition (where feeding by the oral route is impossible because of multiple intestinal fistuli), maintenance and support of vital organ function, and infection control. Appropriate and timely surgical intervention may be life saving.

[Transplantation of periosteum-coated cortical and cancellous bone grafts for treatment of fractures and pseudarthrosis (author's transl)].

Bone grafts are used in the treatment of fractures just as in the treatment of atrophic pseudarthroses of long tube bones. Only autologous bone graft is a reliable transplantation material. The discussion about different values of cancellous grafts and cortical-cancellous grafts is not completed yet. It will be reported about the own results of 110 transplantations of fresh and ununited fractures of long tube bones between 1968--1975. Only the autologous cortical-cancellous periosteum-coated single graft was used.

Ion channels formed by chemical analogs of gramicidin A.

Channel-forming peptides such as gramicidin A offer the opportunity to study the relationship between chemical structure and transport properties of an ion channel. This article summarizes a number of recent experiments with chemical analogs and derivatives of gramicidin A using artificial lipid bilayer membranes. The introduction of negative charges near the channel mouth leads to an increase in the cation transport rate. Hybrid channels consisting of a neutral and a negatively charged or of a positively and a negatively charged half-channel may be formed. The current-voltage characteristic of these hybrid channels exhibits a pronounced asymmetry. Experiments with charged derivatives of gramicidin A have been used in order to distinguish between different structural models of the dimeric channel; these studies strongly support Urry's model of a single-stranded, head-to-head associated helical dimer. In a further set of experiments gramicidin analogs with modified amino acid sequence were studied. It was found that a single substitution (tryptophan replaced by phenylalanine) leads to marked changes in the conductance of the channel. Analogs with a simplified amino acid sequence such as (L-Trp-D-Leu)7-L-Trp or L-Trp-Gly-(L-Trp-D-Leu)6-L-Trp are able to form cation permeable channels with similar properties as gramicidin A.

Protection of the skin around intestinal fistulas.

The use of two recently introduced skin protective preparations in the treatment of perifistular skin inflammation is described. Karaya gum in paste form (karaya paste) and a compressed wafer of gelatin, pectin and methyl cellulose (Stomahesive) were used, alone or in combination; with drainable collecting appliances. The method protects the inflamed skin from fistula discharge and allows healing to proceed beneath the dressing. A technique to improve the adhesion of the appliance, in spite of large fluid losses, is outlined. Healing of skin excoriation was obtained in nearly all cases. Complete control of leakage was achieved in a group of patients with a high output fistula.
