Lactate dehydrogenase of trout: hybridization in vivo and in vitro.

Speckled trout and lake trout contain five forms of lactate dehydrogenase, but a diflerent electrophoretic distribution of isozymes characterizes each species. The hybrid splake, which is produced artificially by fertilizing lake trout eggs with speckled trout sperm, contains nine isozymes. This comnplement of isozymeg In vivo could be produced in vitro by recombination of subunits from tissues of the parent species. In the splake trout, this complement is the result of heterozygosity at the gene locus responsible for synthesis of LDH-5. Extracts of trout eyes contain at least two additional forms of LDH which could not be demonstrated in other tissues.

Thermal unfolding of bacteriophage T4 short tail fibers.

The short tail fibers of bacteriophage T4 are composed of a homotrimer of the product of gene 12 (P12) with a molecular weight of 165,000. P12 is capable of reconstituting defective phage particles lacking gene 12. After heating to 75 degrees C, P12 was able to retain 90% of its ability to reconstitute T412- particles. When heated above 75 degrees C, P12 was no longer capable of fully reconstituting defective phage particles. By 95 degrees C, the reconstitution efficiency of the P12 preparation was reduced by 4 orders of magnitude. Thermal unfolding was also monitored by heating the protein in the presence of SDS to freeze partially unfolded states; by protease hydrolysis; and by intrinsic fluorescence changes. Exposure to SDS had little effect for temperatures up to 55 degrees C, but by 65 degrees C, the reconstitution efficiency of P12 treated with 0.01% SDS dropped to less than 1% of the original titer. Thermolysin digestion of P12 heated to various temperatures showed that treated P12 started to inactivate before 45 degrees C and inactivation was essentially complete by 55 degrees C. Intrinsic fluorescence data of heated P12 indicated that the protein begins to unfold by 45 degrees C and exhibits distinct peak shifts at 60 degrees C and above 80 degrees C. We conclude that, in the absence of SDS or proteases, P12 heated to 75 degrees C can refold back to an active conformation. Trimeric P12 undergoes some irreversible denaturation between 75 and 85 degrees C, and heating between 85 and 95 degrees C results in dissociation of P12 into monomers.

Diagnosis and successful management of septal myocardial abscess: a complication of bacterial endocarditis.

A case of acute enterococcal aortic valve endocarditis is presented in which the complication of a septal myocardial abscess was diagnosed clinically and successfully treated surgically. This represents the first instant, to our knowledge, in which the preoperative diagnosis of a myocardial abscess served as the indication for emergency cardiac surgical intervention in active endocarditis with successful outcome. The diagnostic parameters permitting clinical recognition of a myocardial abscess include the development of advancing degrees of atrioventricular and bundle branch block, and the finding of pericarditis or pericardial effusion in aortic valvular infections. Two additional findings were noted in the present case: echocardiographic evidence of septal thickening, and loss of septal Q waves on the electrocardiogram. Since myocardial abscesses do not respond to medical therapy, continuous electrocardiographic monitoring and frequent echocardiographic determinations are recommended in cases of active aortic valve endocarditis to permit early diagnosis and surgical management of this complication.

Response of tissue-equivalent ionization chamber to 15-MeV neutrons.

The calibration (in rad/nC) of an air-filled tissue-equivalent ionization chamber for neutrons of 15-MeV average energy was determined by measuring absolute fluences and calculating the kerma per unit fluence. The neutron calibration determined from the gamma-ray calibration and application of the Bragg-Gray relation was found to be 4% higher than that based on the fluence measurement. Additional data were taken to obtain calibration factors with tissue-equivalent gas; the same difference betwen the two methods was observed.

Electrocardiographic changes in coronary care unit patients during defecation.

Fifty-one consecutive patients admitted to a coronary care unit were monitored for ECG changes while having a total of 63 bowel movements (BM's). Twenty-three patients had proven myocardial infarction while 28 had no acute cardiac disease. ECG changes in patients using either commode or bedpan facilities were compared during defecation. Patients using the commode were found to have a greater increase in heart rate and other nonspecific ECG abnormalities compared to those using the bedpan.

Calpain and calpastatin are located between the plasma membrane and outer acrosomal membrane of cynomolgus macaque spermatozoa.

Mammalian sperm must undergo an acrosome reaction prior to penetration of the zona pellucida and subsequent fusion with an oocyte. Sperm gain the capability to acrosome react after a period of capacitation, which primarily involves biochemical changes in the sperm membranes. The morphological events of the acrosome reaction have been well-documented, but the underlying cellular mechanisms that regulate capacitation and the acrosome reaction remain unclear. Antibodies to the 2 ubiquitous calpains, mu and m, as well as the small subunit, which associates with both calpains, were localized at the ultrastructural level to the region between the plasma membrane and the outer acrosomal membrane of cynomolgus macaque sperm. After the acrosome reaction, all of the anti-calpain antibodies labeled the acrosomal shroud, suggesting that calpains are located throughout the cytoplasmic area between the 2 outer sperm membranes. Calpastatin is an endogenous modulator of calpain activity and is also localized within the same cytoplasmic region as calpains. The antibodies used for ultrastructural localization were also used to probe Western blots of sperm extracts. Antibodies to either the mu- or m-calpain recognized an 80-kd protein, which is similar to the molecular weights of other ubiquitous calpains described. The small subunit (30 kd) was also recognized with a specific monoclonal antibody. An antibody to calpastatin recognized a major band at 78 kd and a lighter band at 45 kd, while the antibody to the testis-specific isoform of calpastatin (TCAST) recognized a 110-kd protein. We hypothesize that this cysteine protease system may be functional in cynomolgus macaque sperm during capacitation, the acrosome reaction, or both.

Measles during the Gulf War: a public health threat in Israel, the West Bank, and Gaza.

A measles outbreak began in Ramallah District in the West Bank in the period just before the outbreak of the Gulf War in 1991. After a local control program, the outbreak was contained to a total of 236 reported cases, including one death, but sporadic cases appeared over the ensuing months. An epidemic of measles followed 2-3 weeks later in Beer Sheva district in Israel, initially among the Bedouin population just prior to the outbreak of the war, then spreading to the Jewish population. The epidemic reached its peak during the war in February, continuing into March. There were a total of over 433 reported cases, with 203 hospitalizations and 7 deaths. The epidemic was slowed by curtailment of school and other civilian activities, and a mass vaccination campaign reaching 60,000 children, but was spread by crowded conditions in hospital, and was spread to other parts of the country by internal population migration. An epidemic of a preventable disease occurring during a national emergency adds to the burdens of the health system. The public health system must be prepared to contain epidemics and other public health threats in wartime, employing standard control methods developed and practiced during peacetime. Routine immunization policies should take into account the possibility of a civilian epidemic during national emergencies, and the need to assure protection of both the civilian and military population in preparation for such contingencies.
