[Family picture studies in patients with paranoid psychoses].

Among the first-degree relatives of 77 patients with delusional functional psychoses the authors found 13 schizophrenics (3.10%), 8 manic-depressives (1.91%) and 14 cases with atypical psychoses (3.34%), following ICD-9 criteria. These figures were compared with the figures in the literature on the genetics of paranoid schizophrenics and nonschizophrenic paranoids, regarding the different composition of the samples. Subdivision of the original 77 patients, using the concept of the axial syndromes, was able to form two homogeneous subgroups of first-degree relatives: one with a schizophrenia prevalence of 6.52% (uncorrected) and without any manic-depressive secondary cases (endogenomorphic-schizophrenic axial syndrome) and another with a manic-depressive illness (MDI) prevalence of 6.58% (uncorrected) without any secondary cases with schizophrenia or atypical psychosis. The first-degree relatives of the patients with an 'organomorphic' axial syndrome (usually excluded in other studies) had an increased rate of manic-depressive secondary cases (3.57%, uncorrected) and the highest rate of atypical psychosis (7.14%) without any schizophrenics, according to the etiopathogenetic heterogeneity of this group. 37 of the 77 patients were not assignable to any of the axial syndromes. In the first-degree relatives of these patients--best comparable to Kendler's criteria for 'delusional disorder'--an increased rate of schizophrenics and atypical psychoses was found (3.59 and 3.08%, uncorrected); the figures for MDI were within normal limits. Our results suggest that the axial syndromes are a useful diagnostic instrument in identifying--from the genetic standpoint--in part very homogeneous subgroups. This allows the conclusion that they are indicators for hypothetical basic disturbances.

Biopotency of alpha-tocopherols as determined by curative myopathy bioassay in the rat.

The decrease in plasma pyruvate kinase activity after administration of various tocopherols to vitamin E-deficient rats provided the basis for a 3-point parallel-slope assay for vitamin E activity. Based on 6 replicate assays, RRR-alpha-tocopheryl acetate (RRR-alpha-TA) was 141% as active as all-rac-alpha-tocopheryl acetate (all-rac-alpha-TA). This confirms many previous reports with other bioassays and is in agreement with the unit-weight relationships assigned by The National Formulary. 2-ambo-alpha-Tocopheryl acetate was 97% as active as all-rac-alpha-TA indicating that only the configuration of the 2-position on the chromanol ring is important in determining the biological activity of alpha-tocopherol.

Kinetics of tissue alpha-tocopherol uptake and depletion following administration of high levels of vitamin E.

Following administration of high levels of vitamin E, plasma tocopherol levels in mature humans and monkeys continued to increase with time. In man plasma tocopherol levels were higher when tocopherol rather than tocopheryl acetate was given. Mature female rats that had been maintained on laboratory chow were put on the diet supplemented with either 1,000 or 10,000 mg/kg of all-rac-alpha-tocopheryl acetate. All tissues analyzed (plasma, platelets, liver, red blood cells, adipose tissue, heart, lung, skeletal muscle, and brain) continued to increase in tocopherol content for the duration of the supplement (20 weeks). It was concluded that it is difficult to saturate tissue with tocopherol and that not only the level, but the duration, of supplementation with vitamin E influences the concentration of vitamin E in all tissues. Liver and adipose tissue both accumulated tocopherol at a very rapid rate compared to other tissues, but once the chow diet was resumed, liver tocopherol decreased very rapidly and adipose tissue decreased very slowly. The studies suggest that at least for a short time period, the liver is the major available storage organ for tocopherol. When animals were put back on the unsupplemented chow diet, platelet and plasma levels returned to baseline values within 1 week, and red blood cells, heart, muscle, lung, and brain within 4 weeks.

Effects of aspirin and related drugs in vitamin E-deficient rats.

Evidence from the literature indicates that in vitamin E-deficient animals prostaglandin (PG) synthesis in platelets is enhanced while it is decreased in the muscle and testis. In the present study the effects of aspirin, a known inhibitor of PG biosynthesis, on vitamin E deficiency signs in the rat were investigated. Administration of aspirin to vitamin E-deficient rats had no protective effect on fetal mortality, incisor depigmentation, body weight gain or red blood cell peroxidative hemolysis. Aspirin prevented the anemia and thrombocythemia observed in vitamin E-deficient rats. Aspirin, salicylic acid and a carbazole prostaglandin inhibitor exacerbated testis degeneration in vitamin E-deficient animals. Addition of aspirin to the diet more than doubled the vitamin E requirement for reversal of necrotizing myopathy.

Influence of age on the vitamin E requirement for resolution of necrotizing myopathy.

Weanling rats were fed either a vitamin E-deficient diet or the deficient diet supplemented with vitamin E or the antioxidant ethoxyquin. Ethoxyquin was not effective in preventing the elevation in platelet number or percent aggregation. However, ethoxyquin was as effective as vitamin E in maintaining body weight, testes weight, pigmentation of the incisors and in preventing myopathy, observed either histologically or by an elevation in plasma glutamic oxalacetic transaminase (GOT) activity. Removal of ethoxyquin from the diet after 19 weeks of feeding resulted in a rapid onset of myopathy. These observations permitted us to study the requirement for vitamin E at various ages without the complication of any cumulative pathological effects of vitamin E deficiency. Ethoxyquin was deleted from the diet at 8, 20, 44 and 64 weeks of age. The requirement for vitamin E to reduce plasma GOT and pyruvate kinase (PK) activity was measured at 12, 24, 48 and 68 weeks of age. The requirement for vitamin E did not change significantly during this time period.

Nonschizophrenic paranoid syndromes.

After presenting their model for the description of delusions, the authors report some results of two catamnestic studies in patients with delusional syndromes. There is evidence for the hypothesis that the usual classification of such patients as paranoid schizophrenics is frequently not justified. A reasonable number of the cases might belong to cyclothymia or at least approach it. Disappearance of delusions and complete restitution frequently occur in this group, while chronicity seems connected with persistent psychopathological signs in the accompanying symptomatology supposed to represent--in part--organic brain syndromes or congenital or acquired psychopathic personalities.

[Psychiatric hospitals in motion].

Papers in respect to the topic "On the possibility of influencing the course of psychiatric illness" which were presented at the 5th Steinhof Symposium, October 1979 in Vienna, are summarized with the aim to focus on and to support necessary efforts for an improvement of psychiatric care. Investigations in inpatient populations (Strömgren, E.: Past and future changes in inpatient population; Gabriel, E.: Modifications in inpatient populations of big psychiatric hospitals) showed the importance of institutional differentiation and the similarity of problems in different countries, e.g. Denmark and Austria is mentioned. The papers of Ciompi (Prognosis in rehabilitation of chronic psychiatric patients) and Pfolz et al. (Factors preventing discharge of long-term inpatients from hospital) summarize concepts of inpatient care. In the papers of Schöny et. al. (Ward group therapy--its importance for the opening of psychiatric wards) and Lyon (Supervision of community--auxiliary lay workers) modern concepts of a pragmatic psychiatry are described. Trends in psychiatric inpatient care are becoming apparent which raise the hope that psychiatric hospitals will provide--through the aforementioned differentiation--more adequate care for the individual patient.

[Statistical data analysis of physiologic parameters in patients with endomorphic-depressive phases. An attempt to explore the changeable effects between physiologic parameters, examination times during the phase and various experimental conditions].

Range-corrected (according to Lykken) physiological data from the parameters of heart rate (HF), respiratory rate (AF), skin potential (skin potential level; SPL; skin potential response, SPR) and skin resistance level, SRL; skin resistance response, SRR) were obtained under an acoustic activation paradigm in the course of a longitudinal investigation on 7 women with endomorph depression treated with thymoleptic medication. These data form the matrix for two- and threefold analyses of variance as well as for concordance analyses according to Kendall. During six measurements (three each taken in the morning and evening) one finds differing levels of activation: relatively low ones at the beginning and at the end, high ones during the 'labilization phase' of the treatment. The varying 'reactivity' to the diverse examination sequences of an acoustic activation program is little affected by the degree of these changes; patient-specific 'characteristics' of the course of the activation are just as recognizable throughout the course of the investigation as are 'stimulus-specific' reactions. Each of the parameters is appropriate for the estimation of different physiological variables; SPR seems rather to be a situation-specific variable, HF and AF are probably suitable for establishing the level of activation, while changes in SPL seem to represent patient-specific characteristics.

Fluorinated retinoic acids and their analogues. 1. Synthesis and Biological activity of (4-methoxy-2,3,6-trimethylphenyl)nonatetraenoic acid analogues.

(4-Methoyx-2,3,6-trimethylphenyl)nonatetraenoic acids, esters, and amides (analogues of retinoic acid) bearing a fluorine atom(s) or a trifluoromethyl group on the polyene side chain were synthesized. The biological activities of these compounds and of 10-, 12-, and 14-fluororetinoic acid esters were evaluated in vivo in a chemically induced mouse papilloma test; the toxicities were assessed in an in vivo mouse hypervitaminosis A test. Antipapilloma activity greater than the parent nonfluorinated ester was found for 1c (ethyl 12-fluororetinoate) and 23 and 39 (aromatic 4- and 6-fluororetinoid esters, respectively). A similar increase in antipapilloma activity was observed for 71 and 72, the aromatic 4- and 6-fluororetinoic acids, respectively, relative to 2 and for 73 (aromatic 4-fluororetinoid amide) relative to 4.
