[Obstructive diseases of the respiratory tract caused by industrial allergens (author's transl)].

Obstructive diseases of the respiratory tract caused by allergising substances at the place of work, must be classified and compensated as occupational diseases - provided certain conditions are fulfilled - according to item No. 4301 of the currently valid Industrial Health Insurance legislation. The article discusses the aetiopathogenesis as well as the special features of clinic and diagnosis of occupationally conditioned allergic obstruction of the respiratory tract, previously known as asthma bronchiale. In contrast to "spontaneous" allergy, the occupational cause of the disease is due to the often massive exposition to allergens at the place of work, as well as the frequently aggressive potency of several industrial allergens, these being considered as the actual "shaping" factors. The article stresses the need for early diagnosis, as far as possible already in the is allergic conjunctivitis and rhinitis. Treatment directed at removing the causes includes, fundamentally, prophylaxis against exposure (considering the time during which the allergens remain dormant without triggering an allergy); obviously, this involves change of occupation or place of work, whereas specific treatment aimed at producing hyposensibilisation must remain an exception. The article concludes with a description of the possibilities of prevention mainly from the medical point of view.

The expression of keratin genes in epidermis and cultured epidermal cells.

Cultured human epidermal cells and human stratum corneum (callus) contain a number of keratins of different molecular size, but the size distribution is not the same in the two cases. To characterize these keratins in more detail, we compared them by amino acid analysis, immunological reactivity and one-dimensional peptide mapping (Cleveland et al., 1977). No differences in amino acid compositon could be detected among keratins of stratum corneum differing in molecular size by as much as 50%, suggesting that some repeating structure may be present in these molecules. Examination of polypeptide fragments produced by partial enzymatic hydrolysis showed strong similarities among all the keratins of stratum corneum and of cultured epidermal cells, even extending to the keratins of rodents; but the keratins of similar size, whether of stratum corneum or cultured cells, were more closely related than keratins of different size. This conclusion was supported by studies of the immunological reactivity of the keratins. How the epidermal cell generates a family of keratins is a problem of considerable interest. The differences in size and structure between the keratins of stratum corneum and cultured epidermal cells suggest that the epidermal cell can modify the expression its keratin genes.

Short RNA chains synthesized at low pH are initiated at promoter sites.

Under non optimal conditions- either with limiting substrate concentrations (1) or at low pH (2)- RNA polymerase of Escherichia coli synthesizes very short RNA chains. By sequencing one RNA species synthesized at pH 5.8 upon T7 DNA we were able to demonstrate that under these conditions transcription is initiated at a normal promoter site (here A1) but however is terminated soon afterwards at specific artificial sites not used in vivo.

Propantheline as a diagnostic tool--a serious complication.

Propantheline bromide is a synthetic quaternary ammonium compound widely used in urologic patients to block the action of acetylcholine at the postganglionic nerve endings of the bladder's parasympathetic innervation. It is used clinically in a wide range of doses in ambulatory outpatients as well as hospitalized patients. To document its effects on the bladder, test doses are given with a cystometrogram documenting the dynamic changes. Herein we report a case of serious cardiac toxicity from a diagnostic test dose.

[Studies on influence of intensity factors on the population dynamic of migratory root nematodes with special consideration of the genus Pratylenchus Filipjev. III. Influence of three years irrigation (author's transl)].

Results of a field experiment on the effect of three years irrigation on the population dynamic of migratory nematodes of the cereals are described. In case of constant high moisture content the Pratylenchus spec. were inhibited. A cumulation of this effect during three years was not observed. The influence was only low on the species of the genus Tylenchorhychus, Paratylenchus, on the species of the families Aphelenchoididae/Aphelenchidae, Dorylaimidae, and on the saprophytic nematodes of the order Rhabditida.

The interdependence of magnesium with spermidine and phosphoenolpyruvate in an enzyme-synthesizing system in vitro.

The DNA-dependent syntheses of different enzymes of the bacteriophages T3 and T7 were studied in an Escherichia coli system in vitro with respect to the optimal Mg2+ concentration and its interdependence with substituting (e.g. spermidine) and complexing agents (e.g. phosphoenolpyruvate). The following results were obtained. 1. The optimal conditions for the syntheses of the different enzymes were not identical. The optima for RNA polymerase synthesis were 8 mM Mg2+, 10 mM P-pyruvate and 3 mM spermidine; for S-adenosyl-L-methionine cleaving enzyme synthesis, 6 mM Mg2+, 6 mM P-pyruvate and 3 mM spermidine; and for lysozyme synthesis, 13-18 mM Mg2+, 28 mM P-pyruvate and 3-0 mM spermidine. 2. The optimal conditions for the synthesis of analog enzymes (RNA polymerases and lysozymes) from the two templates were identical with experimental error. 3. Mg2+ and spermidine substituted for each other in relation to the number of their charges. 4. The apparent complexing of one Mg2+ molecule required the addition of 3-5 P pyruvate molecules. 5. Under the optimal conditions the enzyme-synthesizing activity was higher by more than a factor of 10 compared to previously described systems.
