Effects of acute cocaine treatment on the turnover of 5-hydroxytryptamine in the rat brain.

1. The effects of cocaine (20 mg/kg s.c.) on 5-hydroxytryptamine (5-HT) turnover were examined in rats. 2. In vivo cocaine administration resulted in decreased turnover of 5-HT, as indicated by the decreased accumulation of 5-HT after pargyline administration and the decreased accumulation of 5-hydroxyindoleacetic acid (5-HIAA) following probenecid injection. 3. A time-related decrease in 5-HIAA concentrations and a small fall in 5-HT concentrations in the whole brain were observed following the acute administration of cocaine hydrochloride (20 mg/kg). Tryptophan levels were found to be slightly decreased in the brain. 4. Enhanced reactivity, but neither stereotypy nor hyperthermia, was observed following cocaine injection (20 mg/kg). 5. It is concluded that cocaine inhibits the turnover of brain 5-HT and that this action of cocaine may be responsible for the differences in a number of pharmacological effects between cocaine and amphetamine.

Blood serotonin in schizophrenic children. A preliminary study.

Blood serotonin levels were measured in schizophrenic children, all of whom showed manifestations of illness in the first 2 years of life, and controls. Serotonin levels were higher in patients (mean = 0.267 mug/ml) than in controls (mean = 0.218 mug/ml), although the difference did not reach statistical significance. Serotonin levels were significantly higher in patients with florid psychosis and those with lower IQs than in patients in remission or partial remission or higher IQs.

Immunogenicity of inactivated influenza vaccine, purified by adsorption chromatography on porous glas.

A chromatographic process has been developed for a mass purification and concentration of influenza virus. The final virus suspension, purified some 400 times, is characterized by its antigenic properties and protein content. The virus was killed by UV-light and the resulting vaccine tried on 345 people. The immunity level is 8 times higher than the control. In mice which were infected after immunization with a homologous and heterologous influenza virus strains a high protection index was found (10(6) EID50 for homologous and 10(4) EID50 for heterologous strains).

Use of synthesis inhibitors in defining a role for biogenic amines during imipramine treatment in depressed patients.

Endogenously depressed patients who showed an antidepressant response to the tricyclic drug imipramine continued to show sustained well being after alpha-MPT was added whereas depression returned when small doses of PCPA were added for brief periods. In one patient the antidepressant response to imipramine occurred after pre- and continued treatment with alpha-MPT. Urinary excretion levels of MHPG in one of the patients studied longitudinally did not correspond to the direction of clinical affective state but did reflect anticipated changes during alpha-MPT treatment. Implications are that serotonergic mechanisms are likely involved in the anti-depressant effects of imipramine in man.

Photographic measurements of retinal blood oxygen saturation: falling saturation rabbit experiments.

A noninvasive photographic method of measuring retinal blood oxygen saturation and arteriovenous oxygen difference was developed and calibrated on rabbits. The experiments reported show that the accuracy of the measurements made using the method is better than 1.0 per cent. The method is applicable to vessels located anywhere in the fundus and represents a significant improvement over previous methods.

The search for the dopamine receptor: tribulations.

In an attempt to identify specific binding to dopamine receptors the binding of 3H-pimozide to preparations of brain and to an artificial cellulose membrane was examined. Binding occurred rapidly, was pH and temperature dependent, and was displaceable by other neuroleptics. Displaceable binding was saturable at 5 X 10(-8) M pimozide. A weak correlation between IC50's for displacement of binding by drugs and their clinical potencies was observed. Displaceable binding of 3H-pimozide in different brain areas did not correlate with dopamine levels. The similarity of binding properties to brain and to artificial membrane suggests that these effects are a function of the physical-chemical properties of these drugs, and that these properties may be related to their clinical effect.

Dopamine synthesis: stimulation by a hypothalamic factor.

The effect of treatment with the factor that inhibits the release of melanocyte stimulating hormone (MSH) identified as 1-prolyl-1-leucylglycinamide (MIF) on brain catecholamine synthesis was examined in normal and hypophysectomized rats. The tripeptide induced a dose-related increase in striatal dopamine synthesis in slices obtained from treated normal animals but not in hypophysectomized animals. Hypothalamic norepinephrine synthesis was unaltered by MIF treatment in normal as well as in hypophysectomized rats. In addition, dopamine and norepinephrine syntheses were depressed in untreated hypophysectomized animals, as compared to normal controls. These results constitute the first direct demonstration of a central neurochemical effect of a hypothalamic factor.
