Preliminary investigations of the metabolism and pharmacological activity of beta-hydroxytryptamines in mammals.

beta-Hydroxytryptamine and beta-hydroxy-5-hydroxytryptamine were incubated with rat liver slices and oxidative deamination was established as the main route of metabolism: in both instances the corresponding indole-3-glycollic acids and indole-3-ethane diols were the major metabolites. However, the rates of deamination of beta-hydroxylated tryptamines, as measured manometrically, were found to be much slower than those of tryptamines nonhydroxylated in the side chain. The pharmacological activities of beta-hydroxylated tryptamines were tested in guinea-pigs on resistance of respiratory pathways, spontaneous respiration, electrocardiogram, blood pressure and isolated ileum, using tryptamine and 5-HT as reference substances. The effects of tryptamines hydroxylated in the side chain were in general similar to those of corresponding tryptamines but of much lower intensities; only in increasing the blood pressure was beta-hydroxytryptamine as active as tryptamine. The different reactions of these two groups of substances in the presence of some antagonists indicate that the receptors are probably not the same.

Enzymatic properties of prostaglandin synthetase from bovine seminal vesicles.

The relative quantities of PGE2 and PGF2alpha biosynthesized are strictly dependent on the concentration of the precursor, arachidonate, on the nature and on the quantity of sulfhydrylated cofactor present. The sigmoid aspect of the curve of PGF2alpha biosynthesis as a function of arachidonate concentration is directly dependent on the GSH concentration. Treatment of the microsomes by low concentrations of HgCl (5 muM) modifies the kinetics of PGE2 and PGF2alpha biosynthesis. Similarly, the presence of L-8027, a competitive inhibitor, alters qualitatively and quantitatively the production of PGE2 and PGF2alpha as a function of arachidonate concentration.

[Double aortic arch in infancy and childhood].

The diagnosis of double aortic arch was suspected by oesophagography and subsequently proven by angiography in six children. The patients' age ranged from 14 days to 13 years. The diagnostic value of angiography in establishing the existence of two aortic arches is emphasized, which can easily be performed by right heart catheterization. In five patients the aorta descended to the right of the spine; the left arch was hypoplastic and became stenotic or atretic distal to the subclavian artery. In only one case the aorta descended on the left side. In this case both arches had approximately the same size. Four patients were symptomatic immediately after birth and presented with severe inspiratory stridor. The clinical course was so dramatic that surgery was inevitable at the age of four weeks, at 4, 6 and 7 months respectively. The youngest patient died postoperatively. In the other infants the stridor disappeared only months after surgery. One girl had a stridorous respiration occasionally during infancy, later on she complained of dysphagia. She was operated on at the age of 13 years. The double aortic arch was an incidental finding in a girl of 8 years; surgery was not performed.
