[Anti-implantation action of nylestriol and its effect on uterine cytosol estrogen and progesterone receptor in rabbits].

The anti-implantation action of nylestriol in rabbit was studied. The changes of uterine cytosol estrogen and progesterone receptor (ER and PR) were analysed with the dextran-coated charcoal method. Plasma progesterone level of early pregnant rabbits was measured by radioimmunoassay. The results showed that oral nylestriol could obviously prevent the early pregnancy of rabbit by anti-implantation of blastocyst. The content of ER decreased in the nylestriol group (P < 0.01). The content of PR in nylestriol group was lower than that of the control group (P < 0.05). No significant difference of plasma progesterone level was noted between the control and treatment groups. These results suggest that there is close relationship between the anti-implantation effect of nylestriol and the decreased content of cytosol ER and PR during the pre-implantation period.

Western blot analysis of antibody responses to influenza virion proteins.

An immunoblotting procedure was developed to detect antibody responses in mice and humans to influenza virion proteins. The technique was capable of detecting 1.5 micrograms of haemagglutinin (HA) on nitrocellulose strips at a 1:5000 dilution of a mouse serum with an initial haemagglutination inhibition titre of 20. The effects of the use of the blocking agent Tween-20 on virion proteins were also studied. The commonly used concentration of 0.05% (v/v) Tween-20, when included in blocking and incubation buffers, greatly reduced the amount of detectable matrix protein but caused no detectable loss of HA and neuraminidase/nucleoprotein proteins. If virion proteins were separated by polyacrylamide gel electrophoresis under reducing conditions, antibody bound to HA2 more strongly than to HA1. Under non-reducing conditions, more antibody bound to the uncleaved HA protein than to other proteins. IgG1 and IgG2a antibody responses in mice to each protein were stronger than IgG2b and IgG3 responses.

[Effects of the spleen on immune state in patients with gastric cancer].

For analysing the effects of the spleen on immune state of gastric cancer patients, T-lymphocyte subsets and serum immunosuppressive factors of splenic venous and peripheral blood were assayed. The results showed that the proportion of CD4+ cells in peripheral blood, especially in splenic venous blood of early cancer patients was higher than that in advanced cancer patients. The proportion of CD8+ cells in splenic venous blood of the advanced cancer patients was higher not only than that in peripheral blood of advanced cancer patients, but also than that in splenic venous blood of early cancer patients. The ratio of CD4+/CD8+ cells in peripheral blood, especially in splenic venous blood showed a decrease with the increase of cancer stage. The immunosuppressive activity of sera of peripheral blood, especially of splenic venous blood was increased significantly in keeping with the cancer stage. It seems that the spleen takes part in regulation of tumor immunity. In early cancer patients it shows some anticancer effects on forming and releasing supplementary T cells (CD4+). However, in advanced cancer patients it effects on the development of immunosuppression through forming and releasing suppressive T cells and serum immunosuppressive factors.

[The relationship between hepatitis B and renin-angiotensin-aldosterone system].

The angiotensin II (AT II) levels in plasma were measured 68 times by using radio-immunoassay in 30 patients with viral hepatitis B (HB) and in 35 healthy persons as the control group. The results showed that the AT II levels of patients with HB were much higher than those of the control group (P less than 0.001). They were 219.25 +/- 91.31 ng/L and 60.70 +/- 10.73 ng/L, respectively. These indicated that the renin-angiotensin-aldosterone system (RAAS) of the patients was in exciting state. The levels of 8 cases of severe chronic active hepatitis (SCAH) (AT II = 270.40 +/- 106.55 ng/L, 6 cases of subacute fulminant hepatitis (SFH) (AT II = 332.80 +/- 140.12 ng/L), and 4 cases of hepatocirrhosis (HC) (AT II = 218.50 +/- 97.64 pg/ml) were all higher than those of 8 cases of chronic active hepatitis (CAH) (100.50 +/- 83.81 ng/L) and those of 4 cases of acute icteric hepatitis (A1H) (123.33 +/- 64.97 ng/L). These findings showed that the levels of AT II were directly related with the severity of the illness. The AT II levels of 8 cases of HRS (270.50 +/- 66.31 ng/L) were higher that those without HRS (174.50 +/- 78.48 ng/L). After treatment with captopril (CPT), the renal function of the patients returned to normal and the patients got better, while the AT II levels decreased greatly. The results suggested that high AT II levels in plasma may be one of the causes aggressing the HRS. The CPT may inhibit the produce of AT II and there are some therapeutic effects for the HRS.

Temporal propositions in a HIS-oriented expert system shell.

This paper describes one of the most important aspects in our recent research on building a HIS-oriented expert system shell: representation and processing of temporal propositions about time-series. With respect to time attribute, medical objects have been classified into three categories: time-independent objects, pointwise time-dependent objects and durationwise time-dependent objects. Temporal propositions can be applied only to time-dependent objects. Through implementation of fuzzy relational operators and their uses, the representation and processing of such propositions become more reasonable and more natural.

Discovering objects in a blood recipient information system.

Application of object-oriented (OO) methodologies has been generally considered as a solution to the problem of improving the software development process and managing the so-called software crisis. Among them, object-oriented analysis (OOA) is the most essential and is a vital prerequisite for the successful use of other OO methodologies. Though there are already a good deal of OOA methods published, the most important aspect common to all these methods: discovering objects classes truly relevant to the given problem domain, has remained a subject to be intensively researched. In this paper, using the successful development of a blood recipient information system as an example, we present our approach which is based on the conceptual framework of responsibility-driven OOA. In the discussion, we also suggest that it may be inadequate to simply attribute the software crisis to the waterfall model of the software development life-cycle. We are convinced that the real causes for the failure of some software and information systems should be sought in the methodologies used in some crucial phases of the software development process. Furthermore, a software system can also fail if object classes essential to the problem domain are not discovered, implemented and visualized, so that the real-world situation cannot be faithfully traced by it.

A protocol for geographically randomized snail surveys in schistosomiasis fieldwork using the global positioning system.

A protocol was created for performing geographically randomized snail surveys for schistosomiasis research using the global positioning system (GPS). This protocol differs from traditional surveys in its ability to accurately map and measure the spatial distribution of snail habitat. The protocol was used to map irrigation ditches, the primary habitat for Oncomelania hupensis, in two residence areas in Sichuan Province, China. From the 7,450 meters of mapped ditches, snail surveys were performed at 203 random sites along the ditch network. Of these, 116 (57.1%) sites had snails. The total number of living snails captured was 2,014, resulting in an average snail density of 0.27 snails per linear meter of potential habitat.

Correlation between DNA methylation and pathological changes in human hepatocellular carcinoma.

BACKGROUND/AIMS: This study was undertaken to obtain information at the molecular level on the possible mechanism of human hepatocarcinogenesis and also to provide a clue for further study. METHODOLOGY: We examined the methylation patterns of c-myc and c-N-ras oncogenes, which are most closely related to HCC, by using the Southern blot technique and HpaII/MspI restriction enzymes. In addition, we measured the level of global DNA methylation in cancerous and paracancerous tissues of HCC by incubating DNA with 3H-S-adenosylmethionine (3H-SAM) in the presence of methylase. The measured global level of DNA methylation was compared with pathological changes of the liver. The methylation patterns of oncogenes as well as the levels of global DNA methylation were compared with pathological changes. RESULTS: The results showed that the hypomethylation rates of c-myc and c-N-ras oncogenes were 30% and 61% respectively in human hepatocellular carcinoma, coinciding with a decreased level of global DNA methylation. DNA hypomethylation was highly significant in cases with a tendency of tumor infiltration or metastasis. CONCLUSIONS: It is concluded that abnormal DNA methylation plays an important role in the process of hepatocellular carcinogenesis. DNA methylation level is closely correlated with the biological characteristic of liver cancer, the lower the level of DNA methylation, the stronger the infiltration and metastatic capacity.
