Leukotriene B4 mediates histamine induction of NF-kappaB and IL-8 in human bronchial epithelial cells.

In 16HBE transformed human bronchial epithelial cells, histamine stimulated interleukin (IL)-8 mRNA and protein secretion, and this histamine stimulation was inhibited by the H1-receptor antagonist diphenhydramine (DPH), by the inhibitor of 5-lipoxygenase-activating protein (FLAP) MK-886, by the 5-lipoxygenase inhibitor Zileuton, and by dexamethasone. Histamine stimulated bronchial epithelial cell production of leukotriene B4 (LTB4), and this production was inhibited by FLAP inhibitors MK-886 and L-655,238 and Zileuton. Histamine stimulated IL-8 luciferase reporter gene activity that was inhibited with DPH, dexamethasone, MK-886 and L-655,238, and Zileuton. The inhibition of IL-8 transcription and protein secretion by FLAP inhibitors and Zileuton was reversed with exogenous LTB4. There was increased IL-8 nuclear factor-kappaB (NF-kappaB) DNA-binding activity after histamine stimulation, and this was inhibited by DPH and MK-886. Cytoplasmic phospholipase A2 mRNA levels were also potently induced by histamine. Thus histamine stimulation of bronchial epithelial cells involves binding at H1 receptors, production of LTB4, activation of NF-kappaB and increased expression of IL-8.

CsA-sensitive purine-box transcriptional regulator in bronchial epithelial cells contains NF45, NF90, and Ku.

Human bronchial epithelial (HBE) cells express interleukin (IL)-2 [Y. Aoki, D. Qiu, A. Uyei, and P. N. Kao. Am. J. Physiol. 272 (Lung Cell. Mol. Physiol. 16): L276-L286, 1997]. 16HBE-transformed cells contain constitutive and inducible nuclear DNA-binding activity for the purine-box/nuclear factor (NF) of activated T cell (NFAT) target DNA sequence in the human IL-2 enhancer. Transcriptional activation through the purine-box DNA sequence requires stimulation with phorbol 12-myristate 13-acetate + ionomycin, and this activation is inhibited by cyclosporin A. Immunohistochemical staining of 16HBE cells demonstrates nuclear expression of the purine-box DNA-binding proteins NF45 and NF90 and no expression of NFATp or NFATc. NF90 and NF45 associate with the DNA-dependent protein kinase catalytic subunit and the DNA-targeting subunits Ku80 and Ku70 (N. S. Ting, P. N. Kao, D. W. Chan, L. G. Lintott, and S. P. Lees-Miller. J. Biol. Chem. 273: 2136-2145, 1998). Antibodies to Ku potently inhibit the purine-box DNA-binding complex. The purine-box transcriptional regulator in 16HBE cells likely comprises NF45, NF90, Ku80, Ku70, and the DNA-dependent protein kinase catalytic subunit.

[Observation of antiarrhythmic effects of Cinnamomum migao H. W. Li on experimental arrhythmia].

OBJECTIVE: To investigate the effects of Cinnamomum migao on experimental arrhythmia. METHOD: Arrhythmic models of mice, rabbits, guinea pigs and rats were built using chloroform(Chl), adrenalin(Adr), strophanthin-K (Spt-K) and barium chloride (BaCl2). The affected animals were divided randomly into three groups: control group, Cinnamomum migao (CV-3) group and mexiletine (MXL) group, so as to observe and compare the antiarrhythmic effects. RESULT: CV-3 could reduce the incidence of ventricular fibrillation caused by ch1 in mice and the ventricular tachycardia induced by Adr in rabbits, delay the onset time of this arrhythmia, increase the arrhythmic doses of Spt-K in guinea pigs, reduce the incidence of some arrhythmia caused by BaCl2 in rats and slow down their heart rate. CONCLUSION: CV-3 has obvious antiarrhythmic effects on experimental arrhythmia. The mechanism of these effects is probably related to the arrest of the intraflow of Na+, Ca2+ in the cardiac cells and the depression of their cardiac autoarrhythmicity and conductivity.

[Cell clone technique for Taxus cuspidata Sieb. et Zuicc].

The cell clone technique was studied for Taxus cuspidata with needle-derived calli cells. The plating efficiency(PE) of the third generation of suspension culture cells was found to be the highest one and the PE of the medium solidified with phytagel was found to be almost the same as that of the medium solidified with low melting agarose gel. Solid-solid double layer culture was found to promote the PE of Taxus cuspidata cells moderately. A large number of stable cell lines with high growth rate were obtained and the browning problem of cells was completely solved via plate culture of the cell clones of T. cuspidata. The growth rate of the best cell line reached 0.4 g.FW/g.FW.d.

[Effects of Curcuma phaeocaulis on learning and memory and lipid peroxide in mice].

Nine months mice were inoculated and the method of water maze test and lipid peroxide determination to study the effects of extract of Curcuma phaeocaulis Valeton on learning and memory ability and retarding of senescence. Results showed that in water maze test the time for the group with Curcuma phaeocaulis Valeton needed from initial position to the end was significantly shortened compared with old control. The error numbers for the group of Curcuma phaeocaulis Valeton were significantly decreased compared with old control after starting experiment for 18 days and 24 days and for 35 days no significance difference was showed between groups. The levels of brain, liver and red blood cells lipid peroxide in the group with Curcuma phaeocaulis Valeton were considerably lower than old control, hemoglobin content was considerably increased than old control.

Specificities of anti-sialyl-Tn and anti-Tn monoclonal antibodies generated using novel clustered synthetic glycopeptide epitopes.

The fine specificities of MAbs generated using novel synthetic clustered STn and Tn glycopeptides as immunogens were compared with the anti-TAG-72 antibodies B72.3 and CC49. Hapten inhibition experiments demonstrated the specificity of several of the MAbs for STn and Tn expressed on ovine submaxillary mucin and tumor derived MUC-1 mucin. Amongst the STn specific MAbs only the B195.3 MAb shows absolute dependence on the presence of sialic acid and specificity to the simple disaccharide NANAA alpha2-6-GalNAc. Identification of tumor associated carbohydrate epitopes in cluster and monomer configurations are possible using MAbs detecting the defined structure specificities described herein.

Complete genome sequence of Methanobacterium thermoautotrophicum deltaH: functional analysis and comparative genomics.

The complete 1,751,377-bp sequence of the genome of the thermophilic archaeon Methanobacterium thermoautotrophicum deltaH has been determined by a whole-genome shotgun sequencing approach. A total of 1,855 open reading frames (ORFs) have been identified that appear to encode polypeptides, 844 (46%) of which have been assigned putative functions based on their similarities to database sequences with assigned functions. A total of 514 (28%) of the ORF-encoded polypeptides are related to sequences with unknown functions, and 496 (27%) have little or no homology to sequences in public databases. Comparisons with Eucarya-, Bacteria-, and Archaea-specific databases reveal that 1,013 of the putative gene products (54%) are most similar to polypeptide sequences described previously for other organisms in the domain Archaea. Comparisons with the Methanococcus jannaschii genome data underline the extensive divergence that has occurred between these two methanogens; only 352 (19%) of M. thermoautotrophicum ORFs encode sequences that are >50% identical to M. jannaschii polypeptides, and there is little conservation in the relative locations of orthologous genes. When the M. thermoautotrophicum ORFs are compared to sequences from only the eucaryal and bacterial domains, 786 (42%) are more similar to bacterial sequences and 241 (13%) are more similar to eucaryal sequences. The bacterial domain-like gene products include the majority of those predicted to be involved in cofactor and small molecule biosyntheses, intermediary metabolism, transport, nitrogen fixation, regulatory functions, and interactions with the environment. Most proteins predicted to be involved in DNA metabolism, transcription, and translation are more similar to eucaryal sequences. Gene structure and organization have features that are typical of the Bacteria, including genes that encode polypeptides closely related to eucaryal proteins. There are 24 polypeptides that could form two-component sensor kinase-response regulator systems and homologs of the bacterial Hsp70-response proteins DnaK and DnaJ, which are notably absent in M. jannaschii. DNA replication initiation and chromosome packaging in M. thermoautotrophicum are predicted to have eucaryal features, based on the presence of two Cdc6 homologs and three histones; however, the presence of an ftsZ gene indicates a bacterial type of cell division initiation. The DNA polymerases include an X-family repair type and an unusual archaeal B type formed by two separate polypeptides. The DNA-dependent RNA polymerase (RNAP) subunits A', A", B', B" and H are encoded in a typical archaeal RNAP operon, although a second A' subunit-encoding gene is present at a remote location. There are two rRNA operons, and 39 tRNA genes are dispersed around the genome, although most of these occur in clusters. Three of the tRNA genes have introns, including the tRNAPro (GGG) gene, which contains a second intron at an unprecedented location. There is no selenocysteinyl-tRNA gene nor evidence for classically organized IS elements, prophages, or plasmids. The genome contains one intein and two extended repeats (3.6 and 8.6 kb) that are members of a family with 18 representatives in the M. jannaschii genome.

Human airway epithelial cells express interleukin-2 in vitro.

Human airway epithelial cells (AEC) produce the T cell growth factor interleukin (IL)-2 that likely modulates the T cell lung inflammatory response. IL-2 mRNA from cultured AEC and from Jurkat T cells was analyzed by reverse transcription-polymerase chain reaction and Northern hybridization. IL-2 mRNA is present constitutively in AEC and is enhanced twofold after stimulation with phorbol 12-myristate 13-acetate (PMA; 20 ng/ml) + histamine (2 mM). Normal human AEC secrete IL-2 at rest (7 pg/ml), and IL-2 secretion is increased threefold after stimulation with PMA + histamine; this increase is inhibited by dexamethasone and diphenhydramine. Transcriptional regulation of IL-2 was investigated with a transgenic human AEC line, 16HBE/IL-2 luciferase; there is constitutive IL-2 transcription at rest, and IL-2 transcription is enhanced 8-fold by PMA and 25-fold by PMA + histamine. IL-2 regulation differs fundamentally between AEC and Jurkat T cells. AEC IL-2 likely promotes local proliferation of T cells and may contribute to pathological airway inflammation in asthma.

[The relationship between human chorionic gonadotrophin and lipoprotein metabolism].

We compared human chorionic gonadotrophin (hCG-beta) with lipids, lipoproteins and apolipoproteins in serum between pre- and postmenopausal women, and observed the changes of levels of hCG-beta in animals suffering from hypertriglyceridemia. The results have shown that the concentration of hCG-beta in postmenopausal women is much higher than that in premenopausal women (P < 0.001). Correspondingly, triglyceride (TG) is elevated (P < 0.05). High density lipoprotein cholesterol (HDL-C) and HDL-C/LDL-C ratio are decreased (P < 0.001, P < 0.01). On the other hand, the tendency of highing hCG-beta level has been found in animal study. The concentration of hCG-beta in male rats suffering form hypertriglyceridemia is higher than that in normal animals (P < 0.05).

A methylnickel intermediate in a bimetallic mechanism of acetyl-coenzyme A synthesis by anaerobic bacteria.

Resonance Raman (RR) spectroscopy was used to identify a methylnickel adduct (upsilon Ni-C = 422 wave numbers) of carbon monoxide dehydrogenase (CODH) from Clostridium thermoaceticum. Formed at a nickel/iron-sulfur cluster on CODH called center A, the methylnickel species is the precursor of the methyl group of acetyl-coenzyme A in an anaerobic pathway of carbon monoxide or carbon dioxide fixation. Rapid kinetic and RR studies demonstrated that methylation of nickel occurs by heterolysis of the methyl-cobalt bond (upsilon Co-C = 429 wave numbers) of a methylated corrinoid/iron-sulfur protein. In combination with the earlier finding of an iron-carbonyl adduct at center A, detection of the methylnickel intermediate establishes a bimetallic mechanism for acetyl-coenzyme A synthesis.

[A preliminary report on diagnostic complementarity of gut-associated and egg-associated antigenemia in schistosomiasis japonica].

With Hyroxylapatite purified preparations and BACH (biotin aminocapryl hydrazide) biotinylated McAbs, 274-2H10 and 273-2H1, recognizing different egg-associated epitopes, biotin-avidin (BA) involved alkaline phosphatase (AP) ELISA with detecting sensitivities reaching nanogram levels (10(-9), were set up. The detectable limit for crude preparations of Schistosoma japonicum SEAJ-TCA in 2H10-ELISA achieved 1. 0.3. 2 ng/ml, in which only S. japonicum specific egg antigens were efficiently detected, whereas with 2H1-ELISA, which could detect SEA-TCA of both S. japonicum and S. mansoni species, an end point of detecting 3.2 ng/ml was obtained. Repeated tests with human serum groups revealed very significant differences of extinction OD readings between patients and normal individuals. For detection combinations, a previously established anti-CAA homologous AP-ELISA system was parallelly used for gut-associated antigenemia determinations. Taking the mean extinction OD reading of a parallel normal serum group plus 3 SD as corresponding cut off values, 3 patient groups (n = 82, 52, 39) from different areas of transmission intensity were subjected to accumulating determinations for egg- and gut-associated antigenemia. Improved detectabilities to variable extent were achieved in either of the 2 or 3 combinations. The study thus demonstrated that the diagnostic efficiency for human schistosomiasis could be improved by multi-epitope detections for more than one target molecule using corresponding McAbs, especially in areas where the transmission intensity of the disease is comparatively lower.

[Postoperative management of crisis of myasthenia gravis: analysis of 20 patients].

From June 1983 to March 1992, 95 patients with of myasthenia gravis were treated with thyrmoma-thymectomy. Among them 90 patients were taken midsternotomy, and others standard lateral thoracotomy. All the patients had postoperative crisis of myathenia except one who had a cholinergic crisis. 12 patients (60%) belonged to type III and IV According to the Osserman's classification. The pathological findings showed that 17 patients had combined thyrmoma. The management for crisis was satisfactory with a success rate of 95%. The mortality rate was 5% (1/20) in early postoperative stage and 35% (7/20) in later stage, respectively. 12 patients survived for 2.5 to 10 years. Tracheotomy, mechanical ventilation adjustment, control of infection of the respiratory tract, and nourishment supports are the key measures for successful postoperative therapy and decrease of mortality.

Moving a hospital information system towards a client server architecture.

This paper is intended to give you an overview about the migration process of the Hospital Information System (HIS) at the University Hospital of Giessen towards an open client server based architecture. It describes the software architecture of the existing mainframe applications and the network interfaces involved in opening the centralized software architecture to a wide variety of remote applications.

[Influence of Cinnamomum migao H.W.Li oil on hemodynamic action in anesthetized cats].

Cinnamomum migao oil (CV-3) 0.2ml.kg-1 was intraduodenum given to anesthetized open-chest cats. Ninety minutes after treatment with the oil, the SAP and DAP were slowly reduced to 15.2% and 14.2%, respectively. At the sametime, the HR was slowed down and CO decreased. The left ventricular pressure (LVP), the left ventricular end diastolic pressure (LVEDP), dp/dtmax and Vmax were remarkably reduced respectively. The LVP-(dp/dtmax)P-1 loop was narrowed. Isosorbide dinitrate showed similar effects to CV-3.

Nature's carbonylation catalyst: Raman spectroscopic evidence that carbon monoxide binds to iron, not nickel, in CO dehydrogenase.

Carbon monoxide dehydrogenase catalyzes the synthesis of acetyl-coenzyme A from coenzyme A, a methyl group, and carbon monoxide. The carbon monoxide binds to a mixed metal center of the enzyme, which contains nickel bridged to an iron-sulfur cluster. Resonance Raman spectroscopy has been used to identify both C-O stretching and metal-CO stretching vibrations of the carbon monoxide adduct of the enzyme. This adduct was shown by isotopic exchange to be on the pathway for acetyl-coenzyme A synthesis. The metal to which carbon monoxide is bound was established to be iron, not nickel, by preparation of enzyme from bacteria grown on iron-54 and nickel-64. The Fe-CO frequency is low, 360 wave numbers, implying a weak bond, probably because of electron donation from sulfide and thiolate ligands of the iron. A bimetallic mechanism is proposed, in which carbon monoxide binds to an iron atom and is subsequently attacked by a methyl group on a nearby nickel atom, forming an acetyl ligand, which is then transferred to coenzyme A.
