Noncoding 3' sequences of the transferrin receptor gene are required for mRNA regulation by iron.

The cell-surface receptor for transferrin mediates cellular uptake of iron from serum. Transferrin receptor protein and mRNA levels are increased in cells treated with iron chelating agents, and are decreased by treatment with iron salts or hemin. Here we report that expression of human transferrin receptor cDNA constructions in stably transfected mouse fibroblasts is regulated both by the iron chelator, desferrioxamine, and by hemin. We found that sequences within the 3' noncoding region are required for the iron-dependent feed-back regulation of receptor expression, whereas the presence of the transferrin receptor promoter region is not necessary. Regulation by iron is observed when transcription is initiated at either the SV-40 early promoter or the transferrin receptor promoter, but deletion of a 2.3 kb fragment within the 2.6 kb 3' noncoding region of the cDNA abolishes regulation and increases the constitutive level of receptor expression. Furthermore, the 3' deletion does not affect the decrease in receptors which is observed in response to growth arrest, indicating that transferrin receptor expression is controlled by at least two distinct mechanisms.

Recent advances in pathology as applied to orbital biopsy. Practical considerations.

Advances in pathology allow for more specific diagnoses of orbital disease. The authors discuss the value of awareness of advances in cytology, histochemistry, immunohistochemistry and electron microscopy as applied to orbital disease. Modern cytologic technique can aid in clearer visualization of cellular detail with improved diagnosis of thin needle aspiration biopsies. Histochemistry offers an increasing range of methods for identification of cellular and extracellular substances such as amyloid, fibrin, neuroglia, and collagen. Immunohistochemistry allows for identification of an ever-increasing number of component antigens including immunoglobulins, myoglobins, keratin, glial fibre protein, etc. Electronmicroscopic technique including plastic embedding allow for specific identification of lesions based on subcellular components and characteristic nuclear, cytoplasmic, membrane, basement membrane and stromal components. The value of these methods has been demonstrated with case presentations of "small round cell tumors" of the adult and child. In addition, the pathologic diagnosis of several rare lesions of the orbit including neuroendocrine carcinoma, histiocytosis X, simultaneously occurring poorly differentiated mucoepidermoid carcinoma of lacrimal gland and adenocarcinoma of the prostate are demonstrated to underline the advances in technology. Emphasis is placed on the management of biopsy material to maximize diagnostic potential.

Cloned mouse mammary tumor virus DNA exhibits glucocorticoid-dependent expression in simian virus 40-transformed mink cells.

Mink lung epithelial cells were transfected with two cloned mouse mammary tumor virus (MMTV) DNAs, a 9-kilobase clone derived from an unintegrated exogenous viral genome and a 14-kilobase clone containing an integrated endogenous provirus along with cellular flanking sequences. Mink lung cells were chosen because they do not contain endogenous MMTV sequences. On the basis of our observation that simian virus 40 DNA efficiently transforms these cells, we isolated cell clones containing MMTV DNA by using transformation with simian virus 40 DNA as a selective marker in cotransfection experiments. Levels of the 9-kilobase MMTV mRNA representing the entire viral genome and of the spliced 4.4-kilobase mRNA which codes for the viral envelope proteins were glucocorticoid dependent in transformed cells. Expression of low levels of Pr77gag, the precursor of the group-specific viral core proteins, and of gPr73env, the precursor of the viral envelope proteins, was also hormone dependent. We conclude that these cloned MMTV DNAs contain all the information necessary for the synthesis of normal viral RNAs and proteins. These findings also provide further evidence that the DNA sequences involved in the hormone responsiveness of MMTV expression are contained within the viral genome.

Perinatal factors in hand and eye preference: data from the Collaborative Perinatal Project.

Data from 5,899 girls and boys who participated in the NIH Collaborative Perinatal Project were analyzed for relationships between birth order and birth stress and hand and eye preference as measured at age seven. None of the birth variables were significantly related to hand preference at the 0.05 level for either boys or girls. However, left eye preference was related to birth stress in boys. When both sexes were combined, left eye preference was also associated with low and high birthweight, and also with right lateral position of the head in the maternal pelvis.

Survival after necrotizing enterocolitis of leukemia treated with oral vancomycin.

A 56-yr-old female with chronic lymphocytic leukemia developed hemolytic anemia and thrombocytopenia. Prednisone therapy was instituted, but her disease was further complicated by the development of necrotizing enterocolitis. No specific enteropathogens were identified, and the stools were consistently negative for Clostridium difficile toxin. Treatment with vancomycin was instituted and resulted in complete recovery. In view of the high mortality and poor results of treatment in necrotizing enterocolitis, it is postulated that a further trial of this drug is justified.

A framework for local health service control.

In a major speech recently, Dr. David Owen, former minister of health, argued the case for a genuine localist approach to NHS organisation. He outlines here the advantages of separately elected health authorities financed by local income tax. Under these new arrangements the service, he believes, would be better able to meet the challenge of the cinderella areas and central government could revert to its rightful role of upholding standards and broadly planning resources.
