[Autonomic alcoholic polyneuropathy].

Measurement of heart rate variability was performed in a group of 106 healthy volunteers and 31 alcoholics. The mean momentary arrhythmia of normal individuals shows an exponential decrease with growing age. In 5 of 31 alcoholics a cardiac autonomic neuropathy was found with this parameter. The withdrawing alcoholics had a significant increase of heart rate and decrease of heart rate variability compared to later measurements, what is in agreement with the concept of hyperexcitability in early withdrawal.

Integration of transfected LTR sequences into the c-raf proto-oncogene: activation by promoter insertion.

A malignant cell line (clone S1) isolated after co-transfection of normal NIH3T3 DNA and Moloney leukemia virus long terminal repeat (Mo-LTR) sequences has previously been described to contain an activated c-raf oncogene. Here, we report the isolation by molecular cloning and the structural analysis of the LTR-activated c-raf gene. As shown by Southern blot and nucleotide sequence analyses, the transfected Mo-LTR sequences integrated into the 5th intron of the endogenous c-raf proto-oncogene. This intragenic LTR insertion led to the expression of high levels of LTR-U5-c-raf hybrid transcripts indicating an initiation of transcription from the Mo-LTR promoter. Transcriptional activation of c-raf is accompanied by the synthesis of large amounts of cytoplasmic c-raf protein. Immunoblot analysis suggests that the proteins encoded by the LTR-activated c-raf gene are truncated compared with the normal c-raf gene product(s). Our results indicate a promoter insertion mechanism of c-raf activation.

Automated modification of the Ames test with COBAS Bact.

The automatic analyser for microbiology "COBAS Bact" from Roche, supplemented by a Hewlett-Packard 9816S is used to automate the Salmonella mutagenicity test, developed by Ames. More than 30 compounds were tested in this system and the results were shown to be in good agreement with those obtained with the standard Ames test.

Heart rate variability of chronic alcoholics in withdrawal and abstinence.

Measurement of heart rate variability was performed in 106 healthy volunteers and 31 alcoholics. The mean momentary arrhythmia (MMA) of normal individuals showed an exponential decrease with increasing age. Cardiac autonomic neuropathy was found in 5 of 31 alcoholics. In agreement with the concept of hyperexcitability the withdrawing alcoholics had a significant increase in heart rate and decrease in MMA compared to later measurements. The result suggests that the diagnosis of autonomic alcoholic neuropathy may be reliable only after at least a one week period of abstinence from alcohol.

Extended life span and tumorigenicity of nonestablished mouse connective tissue cells transformed by the fos oncogene of FBR-MuSV.

We have analyzed the transforming potential of two fos oncogene products in nonestablished cultures of mouse connective tissue cells: p55fos of FBJ-MuSV and p75gag-fos of FBR-MuSV. Although both proteins induced morphological transformation and colony formation at low cell density in a G418 resistance selection assay, p75gag-fos exhibited more pronounced transforming potential than p55fos. In addition, p75gag-fos-transformed cells overcame crisis with a high probability and were tumorigenic in syngenic mice. These properties of the FBR-MuSV appear to be linked to structural alterations in the p75gag-fos oncogene product. Polyoma virus large T protein complemented the transforming potential of fos, in that it not only increased the probability of establishment of fos-transformed cells but also enhanced fos-induced morphological transformation. Our results suggest that different oncogenes affect morphological transformation, low cell density growth, establishment, and tumorigenicity to various degrees.

Isolation and propagation of hepatitis A virus in hepatoma cell cultures.

Hepatitis A virus (HAV) was isolated directly from human faeces in PLC/PRF/5 cells. In the first passage cell-bound and supernatant viruses were found by immune electron microscopy and by enzymeimmunoassay. Serial passaging of HAV in PLC/PRF/5 cells resulted in its adaptation to the cell line and in reduction of the incubation time. HAV was still detectable after 10 cell passages. Cell-bound as well as supernatant HAV were employed as antigens in anti HAV IgM-enzymeimmunoassay.

[Neurophysiologic findings in chronic alcohol abuse].

Neurophysiological examinations of 100 long term alcohol dependent patients who were having neuropsychiatric treatment, showed symptoms of polytopic damage of the peripheral and central nervous system. The results show that for recognition of the damage an extensive diagnostic programme must be used.

[Electrophysiologic studies of alcohol dependent patients].

Electrophysiological methods were used to examine 52 ward patients with a long history of alcohol abuse. Alertness fluctuations were much more common in the EEG than among the healthy control subjects. 86.5% of the ENG findings were abnormal and indicated demyelinisation in most cases. The latence times of the brain stem potentials were longer than normal in about half of the patients and were definitely pathological in 13.5%. There was no statistical correlation between drinking habits and the results of the examination. Unknown pathogenetic factors are therefore suspected. The polytopic pattern of damage necessitates the use of a broad range of signostic techniques.

Nutrition of newborns small for gestational age with human milk lyophilisate enriched human milk during the first week of life.

In 24 very low birth weight infants appropriate or small for gestational age, the metabolic response to nutrition with human milk lyophilisate enriched human milk was estimated during the first week of life. In contrast to newborns appropriate for gestational age, in newborns small for gestational age signs of metabolic overloading could be observed: increased urinary amino acid excretion and neonatal cholestasis. Both depended on the degree of fetal growth retardation. It was concluded that nutrition with human milk lyophilisate enriched human milk cannot be recommended for very low birth weight infants during the first week of life. In newborns small for gestational age the metabolic situation has to be estimated before starting a high protein diet. If the level of total bile acids amounts to more than 30 mumol/l, protein intake should be increased carefully during the first week of life.

Influence of temperature on basal and induced hepatic monooxygenase activity in rats of different ages.

The effect of temperature on the rates of ethylmorphine N-demethylation and ethoxycoumarin O-deethylation was studied with liver homogenates prepared from 1- to 120-day-old rats. Breaks at temperatures of 21.5-28.0 degrees C were observed in the Arrhenius plots of these reactions, differences being dependent mainly on age of the animals rather than on reaction type. No breaks could be demonstrated for ethylmorphine N-demethylation with liver homogenates of 10- and 120-day-old rats and for ethoxycoumarin O-deethylation in newborn and 120-day-old rats. In poikilothermic 3-, 10- and homothermic 30-day-old rats ethylmorphine N-demethylation rate was dependent on incubation temperature (7, 20 and 37 degrees C). The relative increase in N-demethylation rate after phenobarbital treatment was most marked in 30-day-old rats at an incubation temperature of 37 degrees C. In infantile rats the relative induction was highest at 7 degrees C.

[Electromyography diagnosis of laryngeal paralysis].

The differential diagnosis of laryngeal muscle paralysis can be improved by electromyographical and neurographical analysis. The findings of 30 patients with recurrent paralysis were put forward and compared with the results among 30 test people.

The influence of triiodothyronine (T3) and inducers on hepatic drug metabolism in rats of different ages.

After a 3-day treatment with T3 the ethylmorphine N-demethylation rate was diminished in 10- to 60-day-old rats, whereas the ethoxycoumarin O-deethylation rate was distinctly enhanced in some age-groups, particularly in 33-day-old rats. P-450 concentration was decreased in young rats and not changed in 60-day-old ones. Phenobarbital administration enhanced the rate of both reactions and P-450 concentration in all age-groups. This treatment significantly attenuated the T3 effects on P-450 concentration and ethylmorphine N-demethylation. beta-Naphthoflavone induced ethoxycoumarin O-deethylation and P-450 concentration in all age-groups. Compared with non-induced animals, the T3 effects on ethoxycoumarin O-deethylation (increase) and ethylmorphine N-demethylation (decrease) were more distinct, whereas P-450 concentration was scarcely influenced by T3. Age-differences in T3 actions were generally diminished by inducers.
