Restriction endonuclease (EcoR1) analysis of Brucella ovis DNA.

Seven Brucella ovis isolates from diverse sources (including two used in the manufacture of a Br. ovis vaccine, four field strains and a reference strain from the National Culture Type Collection in London) were subjected to bacterial restriction endonuclease DNA analysis using the endonuclease EcoR1. No genetic differences were detected. It was concluded that strain variation is unlikely to be a problem in Br. ovis control schemes, whether these are based on vaccination or on the eradication of the disease using serological tests.

A strain of Haemonchus contortus resistant to thiophanate.

The occurrence of a field strain of Haemonchus contortus resistant to thiophanate is reported for the first time in New Zealand. In a controlled anthelmintic trial with experimentally infected animals, thiophanate at 50-100 mg/kg had no appreciable effect on the Haemonchus burden. Albendazole reduced faecal strongylate egg counts by 95% in animals with naturally acquired 59 infections.

Ovine dermatophilosis in young sheep associated with the grazing of Brassica spp. crops.

The occurrence of dermatophilosis in five month old lambs associated with the grazing of Brassica spp. crops is reported on four farms. The prevalence of the disease varied from 3% to 45% within affected flocks. An additional finding was the occurrence of nervous signs in a small number of more severely affected lambs, characterised by a sudden lowering of the hindquarters. The association between dermatophilosis and the grazing of Brassica spp. crops is discussed.

Anatomical considerations of the distal interphalangeal joint of sheep.

The ligamentous attachments and the extent of the capsule of the distal interphalangeal joint of sheep was studied using both dissection and radiographic techniques. The distal interphalangeal joint was considered to be most vulnerable to infection and trauma on the axial, or interdigital, aspect where the joint capsule protrudes above the coronary border. In sheep the interdigital ligament and each proximal collateral sesamoidean ligament help to unite the two digits: a role performed in cattle by the cruciate ligament.

Observations on an outbreak of foot abscess in sheep.

An outbreak of ovine interdigital dermatitis and foot abscess in which approximately a third of 300 Perendale ram hoggets had acute lameness was investigated. Both foot infections occurred simultaneously in the flock, and ovine interdigital dermatitis preceded the occurrence of foot abscess in seven instances. It was suggested that the behaviour pattern and sexual activity of the young rams predisposed them to ovine interdigital dermatitis and foot abscess. Although no controlled trials were conducted, the lame sheep responded well following the combined therapeutic measures of dry surroundings, formalin footbathing and selected antibiotic therapy.

Electromyographic assessment of neuromuscular blockade induced by atracurium.

The effects of atracurium besylate 0.3 mg kg-1 or 0.6 mg kg-1 on neuromuscular function were assessed by electromyography in 11 normal subjects, using successive trains of four supramaximal stimuli. After the induction of anaesthesia, the relation between the reduction in the amplitude of the compound muscle action potential and decrement was studied during the onset and recovery of nondepolarizing blockade. During induction, the decrease in the amplitude of the initial compound muscle action potential was usually greater than decrement, and resembled the pattern observed with pancuronium. In contrast, during recovery from myoneural blockade, decrement was invariably greater than the reduction in the amplitude of the action potential. It was considered that these effects were consistent with the action of atracurium on more than one group of receptors at the neuromuscular junction. The effects of atracurium were rapidly antagonized by neostigmine, and supplementary doses of the drug showed no evidence of cumulation.

An inherited cataract in New Zealand Romney sheep.

A bilateral cataract was noted to occur in sheep on a New Zealand Romney stud. Extensive breeding trials showed that this defect was inherited as an autosomal dominant. As such this form of cataract is of minimal importance to the sheep industry as control is merely by culling affected individuals.

Isolation, physicochemical properties, and the macromolecular composition of the vitelline and fertilization envelopes from Xenopus laevis eggs.

As a step toward defining in molecular terms the sperm-triggered block to polyspermy reaction established by the egg at fertilization, vitelline (VE) and fertilization (FE) envelopes were isolated from eggs of the Sounth African clawed toad Xenopus laevis and some of their physicochemical properties determined. Envelopes were isolated after lysis of the fertilized or unfertilized eggs by sieving techniques; isolated envelopes retained their in situ morphology as determined by electron microscopy. The isolated envelopes had different solubility properties and, in general, VE was more readily dissolved by aqueous solvents than FE, although both could be completely dissolved by detergents or chaotropic agents. Changes in envelope solubility correlated with the progression of the cortical reaction implicating a role for cortical granule material in modifying the solubility properties of the envelope. The VE and FE were composed of protein and carbohydrate with no lipid components detected. As determined by immunodiffusion experiments, the FE contained the same antigens as the VE plus components derived from the cortical granules and the innermost jelly layer, J. The macromolecular composition of the envelopes was determined by sodium dodecyl sulfate gel electrophoresis. The VE contained at least 11 glycoproteins with molecular weights ranging from 125 000 to less than 16 000 with two components (40 000 and 33 000) accounting for almost two-thirds of the total stainable material. The FE contained ten glycoproteins that had the same molecular weights as those in the VE. One glycoprotein component underwent a reduction in molecular weight from 77 000 to 67 500 when the VE was converted to the FE. This molecular weight change was interpreted as the probable result of limited proteolysis. In addition, the FE gel electrophoresis patterns contained macromolecular components derived from the cortical granules and jelly layer, J, consistent with the immunodiffusion experiments. These components were absent when the FE was prepared in the absence of Ca2+, suggesting a role for Ca2+ in binding the VE, cortical granules, and J components together. We concluded that the conversion of the glycoproteinaceous VE to FE at fertilization is caused by interaction of the VE with components from the cortical granules and jelly layer J. These interactions are of both a chemical and physical nature.
