Occurrence, transport, and storage of octopamine in human thrombocytes.

3H-octopamine was found to be accumulated in human platelets, achieving a maximum concentration gradient of 30:1. Its accumulation was partially inhibited by reserpine, imipramine, serotonin, ouabain, dinitrophenol, and iodoacetate. When octopamine was added to platelet preparations, it led to a decrease of both endogenous and 14C-serotonin. To determine whether octopamine accumulates in human platelets in vivo, preparations from 6 patients receiving monoamine oxidase inhibitors and 17 control subjects were assayed enzymatically for octopamine. Octopamine was detectable in all of the drug-treated patients, averaging 0.45 +/- 0.06 ng/mg protein, while only 4 of the 17 control subjects had detectable (greater than 0.05 ng/mg protein) platelet octopamine. Although much lower than platelet serotonin levels, these octopamine levels are in the range of those reported for platelet norepinephrine and epinephrine.

Platelet monoamine oxidase activity in acute schizophrenia.

The authors found normal monoamine oxidase (MAO) activity in 40 acute schizophrenic patients, in contrast to previous reports of a genetically linked platelet MAO deficit in chronic schizophrenia. Variations in MAO activity were not significantly associated with the 65 clinical variables analyzed, although there was a tendency for patients in the low-MAO group to have more severely impaired reality testing, more paranoid and grandiose delusions, better prognostic scores, and less restlessness.

Serotonin storage in platelets: estimation of storage-packet size.

Storage-body diameter and volume, and the number of molecules of serotonin contained in a storage body, were estimated for blood platelets. In the human, 5.23 x 10(5) molecules of serotonin are contained in a storage body 198 nanometers in diameter, while in the cat, 31.2 x 10(5) molecules of this amine are contained in a storage body 298 nanometers in diameter.
