[Leukocyte sensitization to tumor and embryonic antigens in gastric cancer and peptic ulcer].

The antigens (AG) of gastric cancer extracts would inhibit the migration of leucocytes in the culture with control serum in 6 among 7 (85%) patients with gastric cancer (GC) of stage I, in 8 of 10 (80%) patients -- stage II, in 24 of 43 (56%) patients-stage III, in 10 of 35 (28.5%) patients -- stage IV. The effect of autoplasma on the inhibition of leucocytes migration (ILM) of GC AG was varied. The mixture of extracts from several tumors suppressed more frequently the migration of leucocytes. The liver extracts antigens of a 10-week human embryo induced ILM in 29 of 49 (59%) patients with GC I--IV. The gastric mucosa antigens induced ILM in 21 of 95 (22%) patients GC I--IV. The migration of leucocytes in 13 of 110 (12%) control patients was suppressed by AG GC in the culture containing control serum. AG of human embryo liver induced ILM in 7 of 55 (13%) examined control patients, 5 of them having ulcerous disease. It is supposed that sensitization to embryonal antigens may procede malignant transformation of gastric ulcers.

[Replacement of the helper function of T cells by an RNA-containing antigen-specific lysis factor].

The lymph node viable cells suspension of immunized mice was centrifugated. The supernatant was chromatographed in Sephadex G-200, and fractions were deproteinized. The deproteinized third fraction (Mol wt 30000) stimulated specifically the plaque-forming cells of intact mice immunized by SRBC. It restored the capacity to antibody production in the lethally irradiated intact mice protected by the syngeneic bone marrow. The activity of this fraction disappeared following treatment with RNA-ase, but not with DNA-ase or trypsin. The first and the second deproteinized fractions of the supernatant inhibited non-specifically the viable lymph node cells of the immunized animals in the intact mice immunized with SRBC.

[Role of T- and B-lymphocytes in the heterogeneity of human cell-mediated reactions to bacterial antigens].

Leucocytes from 30 patients with allergy to tuberculin and bacterial antigens were treated with antithymus (ATS) and anti-immune globulin (AIGS) sera. The leucocyte migration inhibition test (LMIT) was performed with these antigens. ATS abolished the LMIT induced by tuberculin and sometimes by bacterial antigens (staphylococcal, streptococcal etc.). AIGS frequently abolished the LMIT induced by bacterial antigens, but not by tuberculin. In some cases the treatment with any serum abolished the LMIT induced by the antigens, or, on the contrary, it was abolished only by a successive treatment with both sera. The lymphocyte types (T or B) determining the secondary immune response to the same antigen are different in various patients, as well as they differ in the same patients in relation to diverse antigens. Five types of lymphocyte - antigen interrelation in the LMIT have been distinguished.

[Restoration of antineoplastic reactivity of the leukocytes after preincubation].

Specific antitumor sensitization in patients with carcinoma of the stomach was revealed by means of the leukocyte migration inhibition test. After 24-hour preincubation at 4 degrees C in a serum-free medium the areactive leukocytes from patients with carcinoma of the stomach (stages III--IV) acquired the ability to react specifically to allogeneic antigens of the tumor of the same localization. Preincubation did not influence the inhibition of migration of leukocytes in non-tumor patients by stomach carcinoma antigens. The supernatant of preincubated leukocytes of tumor patients contained substances that inhibited migration of indicator leukocytes.

[Suppression of the intradermal hypersensitivity reaction of the delayed type by a serum fraction from patients containing leukocyte migration inhibition factor].

The blood serum of patients with active tuberculosis of the lungs and chronic pneumonia inhibited migration of donor's leukocytes and macrophages of the peritonal exudate of guinea pigs when compared with migration of similar cells in the medium with the serum of cattle or donors. After chromatography these sera were fractionated on the columns with Sephadex G-100. Fractions containing the leukocyte migration inhibition factor (LMIF) suppressed, up to complete abolition, the intradermal reaction to tuberculin in man and guinea pigs sensitized with BCG. The LMIF is supposed to act in the regulation of delayed hypersensitivity reaction.

[Detection of lymphokines in patients' blood serum].

A factor suppressing the migration of donor leukocytes and macrophages of guinea pigs in vitro was revealed in the blood serum of patients suffering from chronic inflammatory diseases (pneumonia, rheumatism, tuberculosis) and carcinoma. A factor stimulating the leukocyte migration was sometimes revealed in the blood sera of the patients. In chromatography of the blood sera on sephadex G-100 the activity of both factors proved to localize in fractions with the mol wt of 15000--45000 dalton. Depression of stimulation of leukocyte migration could be also caused by immunoglobulin fractions (mol wt--150000 dalton) of the blood sera of patients suffering from acute pneumonia, apparently on account of the presence in them of the antigen-antibody complex; however, these sera contained no migration suppression factor. The blood serum fractions with the mol wt of 15000--45000 dalton, including those containing the migration suppression factor inhibited the inhibited the spontaneous and induced by phytohemagglutinin blast transformation lymphocytes, and the immunoglobulin ones--the latter only. Apparently the migration suppression factor of the blood serum served as the product of activated lymphocytes.

[Effect of serum fractions from cancer patients on leukocyte migration out of capillary tubes].

Blood serum of patients suffering from cancer of the stomach and urinary bladder inhibited in vitro migration of autologous leukocytes, leukocytes of donors and control patients, and also guinea pig macrophages in over half of cases. In chromatography of these sera on Sephadex G-100 the activity inhibiting the leukocyte migration was revealed in fraction I (Mol. wt. over 100000) and in fractions IV and V (Mol. wt under 30 000). The blood serum and its fractions from cancer patients failed to eliminate the leukocyte migration inhibition caused by the tumour antigens in comparison with the leukocyte migration in the medium with control serum without any antigens. As suggested, the activity of fraction I inhibiting the leukocyte migration was due to the antigen-antibody complex, and of fraction IV and V--to a factor similar by its properties to the factor produced in vitro by lymphocytes stimulated by the antigens or mitogens.

[Suppression of leukocyte migration by autologous and allogeneic tumor extracts in kidney cancer patients].

The reaction of leucocyte migration inhibition (RLMI), using antigens of autochthonous and allogenic tumors, was utilized to examine 21 patients with renal cancer and in 37 control patients. The antigens of renal cancer (AG RC) would suppress specifically leucocyte migration in all patients with cancer of the kidney, as compared with the migration without antigens or in the presence of normal tissue antigens of the tumor involved kidney. In 5 of 37 cases AG RC as well as those of normal renal tissue inhibited leucocyte migration in patients with urolithiasis and pyelonephritis. Autologous blood plasma in patients with renal cancer would contribute to inhibition of leucocyte migration by cancer antigens. There are some common specific antigens in renal cancer, recognized by lymphocytes from different patients with the tumor in question. RLMI may be used to establish the immune diagnosis of cancer of the kidney.

[Detection of a factor suppressing leukocyte migration in the sera of allergy patients following antigen administration].

The effect on donor leukocyte migration of serum obtained from the patients with tuberculosis of the lungs, chronic pneumonia and healthy persons was studied after subcutaneous or intradermal injection of the microbial antigen (PPD, streptococcus and staphylococcus antigen). A factor inhibiting donor leukocyte migration appeared in the blood serum of sensitized individuals after the antigen injection. This factor proved to be localized in the serum fraction III obtained after the gel-filtration of sephadex G-200, and is sorbed by leukocytes.

[Antigen-specific lysis in vitro of lymph node cells of intact mice injected with RNA from viable lymph node cells of immunized animals].

Supernatant fluid obtained after centrifugation of the suspension of viable lymph node cells of immunized animals proved to induce in vivo in the lymph node cells of intact mice sensitivity to lysis with a specific antigen in vitro. This property was possessed after chromatography of the supernatant fluid on Sephadex G-200 by the 3rd fraction (MW about 30000 dalton). DNA-ase, trypsin or deproteinization failed to influence whereas RNA-ase inactivated this fraction in respect to the inducing properties.

[Immunochemical characteristics of a factor causing antigen-specific lysis of thymus-dependent lymphocytes].

A factor capable of lysing in vitro, in the presence of a specific antigen, the cells of the lymph nodes and the thymus of intact mice was revealed in the supernatant obtained after the centrifugation of a suspension of viable cells of the lymph nodes and the thymus of the immunized mice. It was found by immunochemical methods that this factor had a mol. wt. of about 30000 dalton and an electrophoretic mobility in polyacrylamide gel exceeding that of mouse blood serum albumin. Besides, it was revealed by the precipitation reaction in agar that it was not an immunoglobulin or its chains.

[Study of bacterial allergy in acute appendicitis by the method of leukocyte migration inhibition in vitro].

Under study was the effect of various bacterial allergens on the migration of blood leucocytes from capillaries in vitro in 23 patients with acute appendicitis and in 18 healthy individuals. Bacterial allergens wound suppress leucocytes migration in all patients. Most frequently, sensibilization to allergens of hemolytic staphylococcus and hemolytic streptococcus was found. In 4 healthy persons this methods indicated sensibilization to some allergens. The interaction of activity sensibilized lymphocytes and passively sensibilized polynuclears of the appendix with bacterial antigens is assumed to tigger the hyperergic reaction in acute appendicitis.
