6-thioguanine: antimitotic effect on human lymphocytes in vitro prevented by adenine.

The action of 6-thioguanine on the division of PHA-stimulated human lymphocytes in vitro has been investigated and shown to have a definite antimitotic effect which is prevented by adenine but not by guanine or hypoxanthine. It is suggested that in this test system 6-TG exerts its main inhibitory action on phosphoribosylpyrophosphate amidotransferase.

Effects of large doses of colistin sulphomethate sodium on renal function.

High doses of colistin were used in the treatment of severely ill patients with refractory klebsiella chest and urinary tract infections. At the same time renal function was monitored to determine possible nephrotoxicity. In all patients it produced acute renal failure and in some acute tubular necrosis. Though renal failure contributed to the final cause of death in some cases, in the majority death was due to the primary neurological illness.

Trial of phenoxymethylpenicillin, phenethicillin, and lincomycin in treatment of staphylococcal sepsis in a casualty department.

A comparative trial of phenoxymethylpenicillin (penicillin V), phenethicillin (Broxil), and lincomycin (Lincocin) against superficial staphylococcal infections seen in a casualty department showed no difference in the efficacy of the three agents, though half the staphylococci isolated were resistant to penicillin. Possible reasons include the fact that antibiotic treatment may not affect superficial staphylococcal infections, or that the organisms concerned may have been weak formers of penicillinase.Half the patients treated with lincomycin complained of diarrhoea and 5% of those treated with phenethicillin suffered from nausea.

Regressive events in the postnatal development of association projections in the visual cortex.

In newborn kittens, neurones in area 17 of the visual cortex projecting to area 18 are distributed in bands of uniform density across the superficial layers (laminae II, III and the upper part of IV) and the deep layers (V and VI). During weeks 2 and 3 postnatal, the cells of origin of this association pathway become mainly restricted to discrete, dense clusters, approximately 600 microns from centre to centre, in the upper layers, with intervening zones free of association cells, as in the adult cat. We have used retrogradely transported, long-lasting neuronal markers to investigate this developmental refinement of the pattern of cortico-cortical connections. The results, reported here, indicate that axonal retraction plays a significant part in the maturation of the clustered organization of superficial layer neurones projecting to area 18, but that cell death may also be a factor in the elimination of the inappropriate projection from the deep laminae.

Predicting the potential for recovery within 24 hours of a head injury.

A linear discriminant model is applied to clinical and radiological signs from 492 consecutive head injuries transferred to a neurosurgical unit over a period of four years. Unstable discriminators may be eliminated and those remaining transformed to interval scales in order to release their full prognostic potential. Patient selection criteria included being in coma, judged in the accident and emergency (A & E) department to be susceptible to developing a complicating secondary event, having a depressed fracture and presenting evidence of leakage of cerebrospinal fluid. These criteria are considerably less restrictive than in most other head injury series. The study is also ambitious in attempting prognosis after only 24 hours. The virtues of early and accurate prognosis include counselling of relatives, patient selection for new forms of treatment and to provide an aid towards medical audit. Neither the relaxed attitude towards patient selection nor the speed of prognosis is found to jeopardize the overall efficiency of allocation which is as high as 91.5%. In fact misallocation is only slightly increased if prognosis is attempted within 60 minutes of admission to the A & E department.
