Treatment of herpes simplex virus infections.

Herpes simplex virus infections cause considerable morbidity in immunocompromised and immunocompetent individuals. The development of immunomodulatory agents and nucleoside analog drugs has been responsible for important advances in the treatment of HSV infections. Acyclovir, an acyclic guanosine derivative, has played a major role in the treatment and suppression of HSV episodes in immunocompromised and immunocompetent patients.

Cognitive functioning of older people in relation to social and personality variables.

Age differences in performance on memory measures and in subjective ratings of memory adequacy were examined in the context of 12 social, personality, adjustment, and lifestyle measures. Participants were 285 men and women, aged 65 to 93, of middle- and working-class backgrounds. A series of multivariate and univariate analyses revealed that a large proportion of the age differences and virtually all of the social-class differences on memory measures could be accounted for by contextual variables, with education, intellectual activity, extroversion, neuroticism, and lie scores (on the Eysenck Personality Inventory) all accounting for more of the variance in memory performance than did age. Self-rated memory adequacy was not correlated with performance, and although the expected finding of lower ratings by older participants was obtained with the working-class group, the opposite was true for the middle-class group. Implications of these results for understanding age differences in memory are discussed.

Personality test reliability: correlates for older subjects.

This study reports the findings of an investigation of psychological and social factors related to test-retest stability and change in scores on the Eysenck Personality Inventory given to 167 older men and women. Higher reliability coefficients on the Eysenck were associated with better scores on a measure of well being and higher levels of education.

Microbial fermentative preparation of L-[15N2]lysine and its tracer: application to serum amino acid kinetic studies.

The microorganism Brevibacterium flavum 21129 has been used to produce multigram batches of L-[15N2]lysine of high purity and isotopic enrichment by supplementation of the growth medium with (15NH4)2SO4 of 98.0 atom% excess. The doubly 15N-labeled lysine can be detected at dilutions 10 times greater than singly labeled lysine when isotope dilution curves are analyzed by gas chromatography-mass spectrometry. This enhanced sensitivity permits kinetic measurements of plasma free-lysine isotope content over a 300-fold dilution during 6 h following a single oral bolus of 5 mg/kg body wt. This inexpensive preparation method lends itself to the production of highly useful biochemical compounds for kinetic studies of human nutrition.

Interaction between Entamoeba histolytica and the immune system. I. Mitogenicity of Entamoeba histolytica extracts for human peripheral T lymphocytes.

The effect of Entamoeba histolytica extracts (E.h.e.) on proliferation in vitro of human peripheral blood lymphocytes, cord blood lymphocytes, and lymphocytes enriched for T cells or non-T cells was investigated. As tested by 3H-thymidine uptake and by morphologic criteria, E.h.e. stimulate T lymphocyte proliferation. Non-T lymphocytes do not respond to E.h.e. The response of lymphocytes to E.h.e. differed from their response to Con A in several respects, indicating either that E.h.e. and Con A act on different subsets of T cells or that E.h.e. act by different mechanism on the Con A reactive cells.

Mitogenicity of Entamoeba histolytica extracts for murine lymphocytes.

Aqueous extracts or aqueous extracts of delipidated Entamoeba histolytica (E.h.e.) contain a mitogenic principle for murine lymphocytes. As detected by [3H]-thymidine incorporation and blast transformation, E.h.e. acted predominantly on T cells of splenic origin, but not on thymocytes or bone marrow cells. Furthermore, E.h.e. induced proliferation of a subset of non-T-cells which is present in the spleen of athymic nude mice, adhered to nylon wool, but could not be activated to produce antibody. It seems possible that polyclonal activation of lymphocytes by E. histolytica might play a role in the disturbance of the immune system as manifested in the impaired cell mediated immune response of E. histolytica infected hosts.

Asymptomatic transient uveitis in children with inflammatory bowel disease.

Although acute anterior uveitis has been noted in children with inflammatory bowel disease, it has not been appreciated in the absence of ocular symptoms. To determine the presence of asymptomatic uveitis, slit-lamp examinations were performed in 19 children with granulomatous bowel disease and seven with ulcerative colitis. In the former group, six had uveitis, while no abnormalities were noted in those with ulcerative colitis. Abnormalities consisted of cells and flare in the anterior chamber. In the group with asymptomatic uveitis, all were male, three were black, and all had colonic involvement. No positive correlations were noted between the presence of uveitis and bowel symptoms, duration of illness, extraintestinal manifestations, or specific treatment regimens. None of the six children with uveitis had evidence of spondylitis, and five were HLA-B27-negative. Repeated eye examinations six to 12 months later disclosed no evidence of uveitis in four of five children and improvement in the remaining child. These data suggest that asymptomatic transient uveitis is common in children with granulomatous bowel disease, but progression to severe adult uveal disease remains unclear.

Immunization against Schistosoma mansoni in rhesus monkeys and the requirement of activation of both cell-mediated and humoral mechanisms.

When groups of rhesus monkeys were pretreated with BCG plus hyperimmune serum from monkeys with chronic schistosomiasis or with dialyzable transfer factor from uninfected monkeys plus hyperimmune serum and were challenged with 1,500 cercariae of Schistosoma mansoni, the mean worm burdens were significantly lower than that of untreated controls. Pretreatment with neither BCG alone nor Corynebacterium parvum plus a membrane antigen of adult worms of S. mansoni affected susceptibility. Neither lymphocyte proliferation in the presence of mitogens or schistosome antigen nor serological responsiveness (as measured by gel diffusion, Cercarienhüllenreaktion, circumoval precipitation, or enzyme-linked immunoabsorbent assay) correlated with the degree of resistance of the animals to S. mansoni. The pretreatment procedures used did not cause any abnormal histopathological responses and did not alter the characteristic host response to schistosome eggs in the lungs, liver, mesenteric lymph nodes, and colon.
