Ultrastructural demonstration of T cells in cutaneous tissue sections using specific anti-human T cell antiserum.

The presence of T lymphocytes in the situ in cutaneous infiltrates of contact dermatitis and lichen planus has been demonstrated by an immunoelectronmicroscope technique. A specific anti-human T cell antiserum labelled with peroxidase-conjugated anti-immunoglobulin serum was used to reveal the specific human T lymphocyte antigen (HTLA) which was visualized ultrastructurally. This technique shows that labelling is limited to the surface membrane and allows study of the in situ interrelationship of T cells.

A simple technique for immunoelectronmicroscopic localization of Ig on tissue sections with special reference to cutaneous pathology.

A simple technique using immunoperoxidase-labelled antibodies is reported for the ultrastructural localization of fixed immunoglobulins in tissue lesions. This technique allows the immunolabeling of frozen sections of 15 mu mounted on glass slides. These sections are then closed over for ultrastructural examination, using an Epon-filled gelatin capsule, and then returned to the glass slide. After polymerization, the capsule containing the tissue section is separated from the glass by immersion in liquid nitrogen. This technique is applied to the study of fixed Ig in the skin lesions of lupus erythematosus, pemphigus, bullous pemphigoid and dermatitis herpetiformis. The results obtained show a fine localization of fixed Ig, good tissue conservation and easy orientation of the biological material. It can be applied to the study of tissue antigens.

Porphyria cutanea tarda-like dermatosis by hemodialysis. Ultrastructural study of exposed skin.

The authors studied by electron microscope, the sun-exposed skin of the back of the hand from three heavily hemodialysed patients with a porphyria cutanea tarda-like bullous skin disease. The vascular impairment, like that of PCT, closely resembles that seen during medicamentous phototoxic processes. The connective tissue is infiltrated by large granulo-filamentous masses and the fibroblasts are secretory in appearance. At the dermal-epidermal junction, the abnormalities are important, with a diffuse infiltration of the upper dermis by a hyalin substance, probably resulting in a collagen degeneration and cellular necrosis. The aetiological factors are uncertain, as no common medicamentous factor appeared in our patients, and as the plasticizers used in the hemodialysis tubes probably played no part.

[Influence of environmental conditions on human hematologic constants].

A blood investigation was carried out on 3,030 volunteer blood donors and the results treated on a computer by multivariant analysis. The authors noted a depressor effect of the urban environment on the red and white cells, a rise in small lymphocytes and a reduction in monocytes in patients living in industrial areas. This stimulation of the lymphocyte system may be compared with the experimental results obtained by the same authors in animals.
