Effects of exposure of Mya arenaria and Mactromeris polynyma to contaminated marine sediments on phagocytic activity of hemocytes.

Two species of bivalves, Mya arenaria and Mactromeris polynima, were exposed to contaminated marine sediments from Baie des Anglais, Quebec, for a period of 10 and 12 weeks, respectively, in order to determine if there was an effect on the phagocytic activity of hemocytes from each species. These sediments contain elevated levels of both PAHs and PCBs. Uncontaminated beach sand was used as control sediments. After a period of 4 weeks, each species of bivalves were sampled and hemocyte phagocytic activity was monitored by flow cytometry. While phagocytosis by hemocytes from M. polytiyma was significantly suppressed, those from M. arenaria were not different from beach sand-exposed controls. At the end of the exposure period, the phagocytic activity of hemocytes from both species was suppressed. Physiological parameters such as mantle proteins or malondialdehyde levels, total protein and total glycogen levels in the digestive gland were not affected by exposure to contaminated sediments. Moreover, the suppression of phagocytosis was well correlated with the transfer of contaminants from the sediments to the bivalves and their subsequent bioaccumulation, as demonstrated by the PCB body burden. These results support the use of bivalves as good sentinel species to survey sediment contamination and the usefulness of hemocyte phagocytic activity as a sensitive biomarker of exposure to organic contaminants.

Occupational risk factors, ultraviolet radiation, and ocular melanoma: a case-control study in France.

BACKGROUND: Ultraviolet radiation has been suspected as a possible cause of ocular melanoma. Because this association is controversial, we examine the role of occupational exposure to ultraviolet radiation on the occurrence of this rare cancer. MATERIAL AND METHODS: A population-based case-control study was conducted in 10 French administrative areas (départements). Cases were 50 patients with uveal melanoma diagnosed in 1995-1996. Controls were selected at random from electoral rolls, after stratification for age, gender, and area. Among 630 selected persons, 479 (76%) were interviewed. Data on personal characteristics, occupational history, and detailed information on each job held were obtained from face-to-face interviews using a standardized questionnaire. Estimates of occupational exposure to solar and artificial ultraviolet light were made using a job exposure matrix. RESULTS: Results show elevated risks of ocular melanoma for people with light eye color, light skin color, and for subjects with several eye burns. The analysis based on the job exposure matrix showed a significantly increased risk of ocular melanoma in occupational groups exposed to artificial ultraviolet radiation, but not in outdoor occupational groups exposed to sunlight. An elevated risk of ocular melanoma was seen among welders (odds ratio = 7.3; 95% confidence interval = 2.6-20.1 for men), and a dose-response relationship with job duration was observed. The study also showed increased risk of ocular melanoma among male cooks, and among female metal workers and material handling operators. CONCLUSION: Following the present study, the existence of an excess risk of ocular melanoma in welders may now be considered as established. Exposure to ultraviolet light is a likely causal agent, but a possible role of other exposures in the welding processes should not be overlooked.

Claudin-1 is not restricted to tight junctions in the rat epididymis.

The blood-epididymal barrier creates a unique microenvironment critical for sperm maturation. There is little information on proteins comprising epididymal tight and adhering junctions or on factors regulating their expression. Claudins are a family of transmembrane proteins reported to be exclusively localized to tight junctions. In the present study the expression of claudin-l (Cl-1) was examined with respect to the different cell types of the epididymis and its various regions as well as its expression during postnatal development and regulation by testicular factors, using both immunocytochemistry and Northern blot analysis. RT-PCR of adult epididymal and testicular RNA (positive control) indicated that Cl-1 messenger RNA (mRNA) transcripts were present in all regions of the epididymis. In the adult, Cl-1 was localized immunocytochemically along the entire length of the lateral plasma membranes between adjacent principal cells, including apical areas containing tight junctions, as well as at the interface between principal and basal cells and along the basal plasma membrane of the epithelium in relation to the basement membrane. Northern blot analysis of adult epididymis with a rat Cl-1 complementary DNA indicated the presence of two hybridizing bands of 4.0 and 1.5 kb. Postnatally, in the caput-corpus and cauda epididymidis, mRNA levels for both transcripts were lowest on day 7. In the caput-corpus epididymidis, mRNA levels for the 1.5-kb transcript increased significantly between 7 and 14 days, whereas the levels of the 4.0-kb transcript were significantly higher by day 21. Postnatal studies revealed that in the initial segment and caput epididymidis, Cl-1 immunostaining was present along the entire length of the lateral plasma membranes of undifferentiated epididymal epithelial cells as early as day 7, including apical areas containing tight junctions. By day 21, staining was identical to that of adult animals, but as this is an age when androgen levels are not at their peak, the data would suggest that they are not a prominent factor regulating Cl-1 expression. Orchidectomy and orchidectomy plus testosterone replacement experiments revealed differences in Cl-1 immunostaining in the initial segment, suggesting that localization of Cl-1 in epididymal tight junctions is androgen dependant. Thus, Cl-1 expression in the initial segment appears to be only partially under the control of androgens. However, in all other epididymal regions, orchidectomy with or without testosterone replacement, revealed no changes to the normal staining pattern, suggesting that androgens do not regulate Cl-1 expression in these regions. Taken together, these studies demonstrate that Cl-1 expression in the epididymis is not localized exclusively to tight junctions, but appears along the entire interfaces of adjacent epithelial cells as well as along the basal plasma membrane, suggesting a role for Cl-1 as an adhesion molecule. The data also suggest that the regulation of Cl-1 in the epididymis is complex and multifactorial.

Flow cytometry as a tool to monitor the disturbance of phagocytosis in the clam Mya arenaria hemocytes following in vitro exposure to heavy metals.

The effectiveness of toxicology biomonitoring programs could be improved by the addition of sensitive biomarkers. In this study the cell viability and sensitivity of phagocytic function of phagocytes from bivalves (Mya arenaria) to selected heavy metals were measured by flow cytometry, a novel approach. Hemocytes (phagocytes) collected from bivalves by puncture of the posterior adductor muscle were incubated in vitro for 18 h in hemolymph containing 10(-9)-10(-3)M of cadmium chloride, zinc chloride, mercuric chloride, methylmercury chloride or silver nitrate, before determining their capacity to phagocytose fluorescent latex beads by flow cytometry. Heterogeneity of the hemocyte cell population was determined by forward scatter (FSC) and side scatter (SSC) cytometric profile which showed two distinct cell populations. At low doses (10(-9), 10(-8) M), all the metal compounds studied stimulated phagocytic activity except silver nitrate. At higher levels of exposure (10(-6), 10(7) M), all metals caused a significant concentration-related decrease in hemocyte phagocytosis activity. From the concentration of each metal inducing 50% suppression (IC50) of the phagocytic activity, the immunotoxic potential of metals with respect to phagocytic function can be ranked in the following increasing order: ZnCl2 < CdCl2 < AgNO3 < HgCl2 < CH3HgCl. Parallel analysis of hemocyte viability showed that suppression of phagocytosis by heavy metals was not solely related to a decreased cell viability. These results reveal the high but different degree of sensitivity of the phagocytosis activity of bivalves with respect to heavy metals, as measured by flow cytometry, and demonstrate that flow cytometry is a potentially useful tool in ecotoxicological monitoring.

Purification, crystallization and preliminary X-ray crystallographic studies of S. cerevisiae Hsp40 Sis1.

Heat-shock protein 70 (Hsp70), one of the major molecular chaperones, has been shown to play a central role in many cellular processes. Heat-shock protein 40 (Hsp40) works as a co-chaperone for Hsp70. Hsp40, bound by unfolded polypeptide, can interact directly with Hsp70 to stimulate the ATPase activity of Hsp70. Hsp40 can also bind to unfolded polypeptides and prevent them from aggregating in vitro, thus acting as an independent molecular chaperone. The S. cerevisiae Hsp40 Sis1 C-terminal peptide-binding domain has been crystallized. The crystals diffract to 2.7 A and belong to space group P41212 or P43212 with a = 73.63, c = 80.16 A. The structure determination by the MAD method is under way.

Advanced-practice sonography in obstetrics and gynecology: a pilot study investigating the efficacy of the ultrasound practitioner.

Advanced-practice (AP) ultrasonography by an AP sonographer or ultrasound practitioner is an emerging allied health profession. The efficacy of this professional in sonographic diagnosis has not been evaluated. This report describes a comparison of the efficacies of diagnosis using a traditional approach and diagnosis by the AP sonographer. Between 1991 and 1995, the authors performed a retrospective, case-controlled study. Patients were evaluated with either (1) a sonographer-based method (limited supervision with perinatal consultation as necessary) or 2) a sonologist-dominant method (direct supervision by a radiologist). Demographic information, ultrasonographic findings, delivery information, and data outcomes were compared. Multiple statistical methods were used and p < 0.05 was considered significant. Of 840 patients studied, 420 were evaluated with the sonographer-based method and 420 with a sonologist-dominant method. There was no significant difference in any potentially predisposing maternal risk factor (except maternal age and education), medical history, variable relevant risk factors associated with the current pregnancy, characteristics associated with delivery, or prenatal ultrasonographic variable. The evaluation of diagnostic efficacy for birth defects showed no difference. The sensitivity and specificity of diagnosis using the sonographer-based method were 100% and 94.7% for the detection of fetal abnormalities, respectively, and 100% and 91.4%, for the sonologist-dominant method. The diagnostic efficacy of the AP sonographer or ultrasound practitioner was found to be similar to that of the traditional sonographer-sonologist model. While, intuitively, there are significant advantages to multiple observers, the experienced, well-trained AP sonographer can function independently, with only discretionary consultation and assistance.

Stability of urinary HVA and VMA on filter paper.

The study examined the stability of HVA and VMA in 1-ml aliquots of a single urine sample stored on filter paper at different temperatures for 2 years. The results showed that HVA and VMA were stable in dried filter paper when stored at 4 degrees C or lower temperature. Storage at room temperature resulted in degradation of the sample.

Diamond-Blackfan anemia as an unusual cause of nonimmune hydrops fetalis: a case report.

BACKGROUND: Diamond-Blackfan anemia is an unusual cause of childhood anemia that results from an absence of bone marrow red cell precursors. To our knowledge, the in utero development of Diamond-Blackfan anemia has been recognized and reported in only two prior instances. CASE: We report the case of a patient who presented with hydrops fetalis during the second trimester of pregnancy. After prenatal evaluation revealed anemia of undetermined etiology, the fetus received intrauterine transfusion therapy. Despite successful treatment of the anemia and hydrops fetalis, in utero renal failure ensued. The infant died neonatally. Postmortem examination of a bone marrow aspirate indicated Diamond-Blackfan anemia as the cause of the hydrops fetalis. CONCLUSION: Diamond-Blackfan anemia may be an unrecognized cause of hydrops fetalis. The suboptimal outcome in this case underscores the larger issue of apparently successful treatment of in utero anemia with little or no impact on postnatal survival.

Cancer risks associated with occupational exposure to magnetic fields among electric utility workers in Ontario and Quebec, Canada, and France: 1970-1989.

To determine whether occupational exposure to magnetic fields of 50-60 Hz was associated with cancer among electric utility workers, the authors used a case-control design nested within three cohorts of workers at electric utilities: Electricité de France--Gaz de France, 170,000 men; Ontario Hydro, 31,543 men; and Hydro-Québec, 21,749 men. During the observation period, 1970-1989, 4,151 new cases of cancer occurred. Each participant's cumulative exposure to magnetic fields was estimated based on measurements of current exposure of 2,066 workers performing tasks similar to those in the cohorts using personal dosimetry. Estimates were also made of past exposure based on knowledge of current loading, work practices, and usage. Workers who had more than the median cumulative exposure to magnetic fields (3.1 microtesla (microT)-years) had a higher risk for acute nonlymphoid leukemia (odds ratio (OR) = 2.41, 95% confidence interval (CI) 1.07-5.44). The same observation holds for acute myeloid leukemia (OR = 3.15, 95% CI 1.20-8.27). There was also an elevated risk for mean exposure above 0.2 microT (acute nonlymphoid leukemia, OR = 2.36, 95% CI 1.00-5.58; acute myeloid leukemia, OR = 2.25, 95% CI 0.79-6.46). However, there were no clear dose-response trends with increasing exposure and no consistency among the three utilities. Men whose cumulative exposure to magnetic fields was above the 90th percentile (15.7 microT-years) had an elevated risk for brain cancer (OR = 1.95, 95% CI 0.76-5.00) that was not statistically significant. No association with magnetic fields was observed for any of the other 29 types of cancer studied, including skin melanoma, male breast cancer, and prostate cancer. Controlling for potential confounding factors did not change the results.

Breeding experiments to combine the X-linked sparse-fur (spf) mutation with the autosomal recessive BALB/cByJ strain: testing the biochemical phenotype of double-mutant mice as a model for ammonia: fatty acyl CoA synergism.

Breeding experiments were conducted to combine the X-linked sparse-fur (spf) mutation with ornithine transcarbamylase deficiency and the autosomal recessive deficiency of short-chain acyl CoA dehydrogenase (SCAD) in BALB/cByJ mice. We obtained spf/Y (scad/scad), spf/+ (scad/scad) and spf/spf (scad/scad) double mutants amongst the F2 progeny, which were tested and separated on the basis of urinary orotate and the GC/MS analysis of urinary butyrylglycine, methylsuccinate and ethylmalonate. The testing of the biochemical type was feasible both on the basis of a 24-h urine collection form adult mice kept in metabolic cages and on the basis of urine spots collected on filter paper from younger progeny. It is postulated that the spf/Y (scad/scad) double-mutant may serve as a useful animal model to study the ammonia: fatty acyl CoA synergism.

A profile of cerebral and hepatic carnitine, ammonia, and energy metabolism in a model of organic aciduria: BALB/cByJ mouse with short-chain acyl-CoA dehydrogenase deficiency.

Spontaneous animal models of inborn errors of metabolism are valuable tools for defining the pathogenesis of these disorders and also the mechanism of various therapeutic approaches. In the present study, we have employed BALB/cByJ mice with an autosomal recessive deficiency of short-chain acyl-CoA dehydrogenase (SCAD). These animals were characterized by a marked urinary excretion of ethylmalonic and methylsuccinic acids along with butyrylglycine. Using adult homozygous mice we have studied the basic cerebral and hepatic profile of carnitine, ammonia, and energy metabolism. The effects of fasting and a short-term supplement of L-carnitine have been evaluated in comparison with control BALB/cJ mice. The mutant mice had low levels of acetyl-CoA and high levels of lactate compared to control mice. Fasting aggravated this condition by further decreasing acetyl-CoA and increasing lactate levels in the mutant mice. Free carnitine levels were significantly decreased in liver with fasting. Long-chain acylcarnitines were significantly lower in the brain of mutant mice. A short-term supplementation of L-carnitine resulted in general increases of carnitine levels in liver and muscle, but they still remained lower in mutant BALB/cByJ mice as compared to control BALB/cJ mice. L-Carnitine treatment increased cerebral CoA-SH levels and both hepatic and cerebral acetyl-CoA levels in mutant mice. Hyperammonemia which has been described frequently in acyl-CoA dehydrogenase deficiencies was not observed in adult BALB/cByJ mice. This could be due to a rapid conjugation of butyryl-CoA with glycine by an increased activity of glycine N-acyltransferase.

Ubiquitin and ubiquitin conjugates in human lens.

Ubiquitin, an 8.5 kDa polypeptide found almost universally in plants and animals, is a normal component in the lens. The best documented function for ubiquitin involves its conjugation to proteins as a signal to initiate degradation. Conjugates for ubiquitin-dependent degradation tend to be of very high molecular mass and are rapidly degraded. Another role of ubiquitin conjugation may be as a stabilizer during stress for protection of constituent proteins, resulting in ubiquitin conjugates that are long-lived. Examination of clear and cataractous human lenses of < 1 to > 50 years revealed the dramatic accumulation of ubiquitin and ubiquitin conjugates with age, beginning at approximately 10 years. Epithelial tissue contained predominantly conjugates of > 250 kDa, although ubiquitin conjugates were found at 98 and 40-60 kDa in tissues from older donors. The water-soluble, urea-soluble and urea-insoluble fractions of lens cortex and core also contain ubiquitin conjugates that accrue with age. High molecular mass conjugates (> 250 kDa) are particularly prominent in older lens tissue. Cataractous lenses, as compared with normal lenses of the same age, show more of these high molecular mass conjugates in the urea-soluble and urea-insoluble fractions of cortex and core. Heterogeneous conjugates in the 20-85 kDa range accumulate in an age-related fashion in all lens cortex and core fractions. While levels of free ubiquitin are significant in the epithelium and the water-soluble cortex and core for all ages, there is no detectable free ubiquitin in the urea-soluble and urea-insoluble fraction under conditions used in this study.(ABSTRACT TRUNCATED AT 250 WORDS)

Malignant melanoma of the skin among workers in a telecommunications industry: mortality study 1976-83.

An incidence study of malignant melanoma of the skin (MMS), conducted previously among the workers of four plants of a large telecommunications industry located in Montreal, Canada, showed a standardised incidence ratio of 2.7 (95% confidence interval (95% CI) 1.3-5.02) for the years 1976 to 1983. To describe more precisely the magnitude of the problem a mortality study was started among the same population (n = 9590) for the same period (1976-83). At the end of 1983, 9180 workers were alive, 261 were dead, and 149 (1.5%) were not traced. Standardised mortality ratios (SMRs) for all causes of death were surprisingly low for men (SMR = 0.57; 95% CI 0.50-0.64) and women (SMR = 0.56; 95% CI 0.37-0.82). The SMRs for major causes of death were also less than expected. These results may be explained by a pronounced selection bias (healthy worker effect) and by the short duration of follow up (eight years). For MMS, two deaths occurred among men (SMR = 2.00; 95% CI 0.24-7.22) and one among women (SMR = 4.81; 95% CI 0.12-26.78). A third man who died of MMS was miscoded as having a primary pulmonary melanoma. Including this case increased the SMR for MMS to 3.00 (95% CI 0.62-8.77; p = 0.08). Polyvinyl chloride and polychlorinated biphenyls were used in the plants and some of the workers did soldering. A planned case-control study will investigate other possible exposures at work.

Diagnosis of polyhydramnios in early gestation: indication for prenatal diagnosis?

Previously published reports have indicated that idiopathic polyhydramnios may be associated with trisomies 18 and 21 and that chromosomal analysis is indicated. Furthermore, the natural history and fetal outcome of polyhydramnios diagnosed in early gestation have not been well delineated. We identified 138 pregnancies with polyhydramnios prior to 26 weeks' gestation. Of 131 complete cases, 21 were diagnosed as severe, 18 as moderate, and 92 as mild polyhydramnios. Congenital abnormalities were noted in 18 of 21 severe cases (86 per cent). Two of the remaining three cases were twin-to-twin transfusion. Thirteen of 18 cases with moderate polyhydramnios (72 per cent) were associated with anomalies; six of the remaining cases were twin-to-twin transfusion. Sixteen of 92 cases of mild polyhydramnios (17 per cent) were associated with congenital abnormalities. In 69 of 76 cases of mild hydramnios not associated with anomalies (91 per cent), the hydramnios resolved prior to delivery. Only 2 of 16 (13 per cent) associated with anomalies resolved. In 4 of 5 cases (80 per cent) with moderate hydramnios and no anomalies, the amniotic fluid volume was normal on subsequent ultrasound. No case of moderate polyhydramnios associated with anomalies or maternal conditions nor any case of severe polyhydramnios resolved. There were seven cases of chromosomal abnormalities in this series; all were associated with sonographic findings in addition to the presence of polyhydramnios. On the basis of these data, we doubt the benefit of amniocentesis following the early diagnosis of idiopathic polyhydramnios in the absence of other ultrasound findings.

Occupational and residential 60-Hz electromagnetic fields and high-frequency electric transients: exposure assessment using a new dosimeter.

One problem that has limited past epidemiologic studies of cancer and exposure to extremely low-frequency (0-100 Hz) electric and magnetic fields has been the lack of adequate methods for assessing personal exposure to these fields. A new 60-Hz electromagnetic field dosimeter was tested to assess occupational and residential exposures of a group of electrical utility workers and a comparison background group over a 7-day period. Comparing work periods only, utility workers' exposures were significantly higher than background levels by a factor of about 10 for electric (E) and magnetic (B) fields and by a factor of 171 for high-frequency transient electric (HFTE) fields. When overall weekly time-weighted averages combining work and nonwork exposures were compared, ratios of the exposed to background groups were lower. B and HFTE exposure ratios remained statistically significant, with values of 3.5 and 58, respectively, whereas the electric field exposure ratio was no longer significant, with a value of 1.7. E-field exposures of the background group were the highest during the nonwork period, probably reflecting the use of electrical appliances at home. Residential E- and B-field exposures were in the same range as published results from other surveys, whereas occupational E-field exposures tended to be lower than exposures reported in other studies. The high variability associated with occupational exposures probably accounts for the latter discrepancy. Worker acceptance of wearing the dosimeter was good: 95% of participants found it to be of little or no inconvenience while at work. At home, 37% found the device to be inconvenient in its present form but would not object to wearing a slightly smaller and lighter dosimeter.

Increased incidence of malignant melanoma of the skin in workers in a telecommunications industry.

In 1982 physicians at a hospital melanoma clinic in Montreal noticed that among their patients there had been seven men working in a single telecommunications company. This raised suspicions that working in that industry might be associated with development of malignant melanoma of the skin (MMS). A preliminary gross comparison with general population rates indicated that there was an increased risk in this working group. To estimate the risk of MMS more accurately, a standardised incidence ratio (SIR) was calculated based on the rates of MMS in the local population of the Greater Metropolitan Montreal Area for the years 1976-83. During that period, among workers in all plants for the company, 10 male cases of MMS were observed for an expected number of 3.7 (SIR = 2.7; 95% CI = 1.31-5.02). No cases were observed among female workers (expected = 1.3). The excess was significant among cases with a short latency (less than 20 years since beginning of employment). There was no apparent pattern of exposure based on job titles or departments.
