An abortive ternary complex of lobster glyceraldehyde-3-phosphate dehydrogenase was produced by the covalent attachment of 3,3,3-trifluoroacetone to Cys- 149 in each subunit. X-ray diffraction analysis of the glyceraldehyde-3-phosphate dehydrogenase-trifluoroacetone-nicotinamide adenine dinucleotide complex showed asymmetry with respect to the active-site conformations of the trifluoroacetone substrate analogue and some catalytic groups. These results are consistent with 19F nuclear magnetic resonance observations of this complex (Bode, J., Blumenstein, M., and Raftery, M. A. (1975), Biochemistry 14, 1153--1160). Different substrate conformations were found on opposite sides of the molecular diad relating subunits whose active centers are in close proximity (the R axis).
The possibility of abolishing pain during operations by needling acupuncture points was detected in China 20 years ago. During the last years the Western World showed great interest in this method, which was tested in a great number of surgical operations. Acupuncture was successful, especially when it was introduced by a short conventional anesthesia. Of special importance seems the possible reduction of anesthetic agents. Though the mode of action of acupuncture cannot yet be explained completely, there exist three different hypotheses: Hypnosis and suggestion, neurophysiological and humoral mechanisms. An actual review on experiments concerning these theories is given.
The applicability of bodyplethysmography in children often fails by the anxiety of sitting alone in the closed plethysmograph cabin. By comparing measurements however it is proved that the results of airway-resistance and thoracic gas volume are not changed in any way, if an adult person accompanies the child inside the cabin. The assumption being essential for a correct measurement are discussed.

The mass media have focused recently on the failure of psychiatric institutions to provide adequate follow-up for its patients released into the inner city. Many expatients deteriorate and become part of the "revolving door": single-room occupancy--city hospitals--state hospital. This paper attempts to demonstrate the feasibility of utilizing existing community resources (for example, a large midtown Manhattan hotel and local service agencies) to cope with this problem. A description of the multiagency organized hotel programs as well as objective and subjective data is provided in order to substantiate our contention that this approach offers a viable alternative to long-term hospitalization.

The history and origin of the science of photobiology are reviewed. Interest in the biologic effects of light gradually increased, beginning with the discovery of ultraviolet and infrared radiation early in the 19th century. The basis of experimental photobiology was laid by the studies of Raab and Tappeiner on photodynamic action and the early uses of phototherapy by Finsen and Dorno. The discovery of the association of porphyrins with some light-related skin diseases and of the capability of chemical agents such as coal tar and bergamot to induce phototoxic contact dermatitis resulted in a flurry of clinical investigations leading to better understanding of the processes of phototoxicity and photoallergy. The early epidemiologic studies of Unna and Dubreuilh relating solar radiation exposure to the formation of actinic keratoses and non-melanoma skin cancer were experimentally confirmed in animals by Findlay, Roffo, and Blum. In the most recent quarter century (1950-1975), cellular and molecular photobiology has been refined. The studies on photochemistry of nucleic acid and of damage and repair mechanisms in DNA have set the stage for understanding the basic processes of biologic effects of light and promise the development of useful applications of specifically directed phototherapy and prevention of such light-induced diseases as skin cancer.

The metabolism and pharmacokinetics of [3H]-labelled 4-(5-isobutylpyrimidinyl-2)-sulphamylphenylacetic acid-3-chloro-6-methoxyanilide sodium ([3H]-glydanile sodium) were investigated in three diabetics and six healthy volunteers after oral and i.v. administration. In the healthy volunteers the level of compound in plasma was correlated to insulin concentration and free fatty acids in plasma or serum, both after oral and i.v. administration of glydanile sodium. After oral administration of 10 mg I13H]-glydanile sodium, maximum [3H] concentration in plasma is found after 2--4 h. The concentration corresponds to 60-90 mug/100 ml when calculated as mug glydanile sodium in plasma. Glydanile is metabolized rapidly. The four metabolites which have been isolated and identified represent more than 95% of total radioactivity in urine. The main metabolite occurring in plasma and urine is the tertiary isobutyl alcohol of glydanile (metabolite I). This metabolite still possesses about 30% of the hypoglycaemic activity of the original compound. The initial half-life of unchanged glydanile after i.v. injection is about 30-40 min. In human plasma more than 99% of the glydanile present is in the protein-bound form. After oral administration, about 20% of radioactivity is eliminated with urine and about 80% with faeces. After i.v. injection, about 70% of radioactivity is eliminated with urine and about 30% with faeces. After oral administration, only about 30% of the compound given was absorbed under these experimental conditions. It may be possible to improve the poor absorption demonstrated in these trials by making the drug available in another, suitable, oral dosage form. Glydanile is eliminated rapidly via urine and faeces after oral and i.v. administration, which means that no accumulation, involving the danger of hypoglycaemic reactions, need be expected. Plasma insulin is increased by a glydanile concentration of 10-20 mug/100 ml plasma.

In 12 patients with bronchial asthma the effects of acupuncture (45 tests altogether) on airway resistance have been investigated. In 9 patients there was a significant decrease of airway resistance 10 minutes, 1 hour and 2 hours after the end of acupuncture. The lowest level for airway resistance (70,1% of control value) was reached during the first hour after acupuncture. The possibility of a merely suggestive effect could be excluded, because "placebo-acupunctures" did not change airway resistance significantly. The comparison with a parasympatholytic acting drug as a metered aerosol (Atrovent) demonstrated that acupuncture had a some-what weaker bronchospasmolytic effect. 3 patients showed after repeated acupunctures no reaction whatever.
