[Multicystic renal dysplasia in the adult (author's transl)].

In the literature 59 cases of multicystic dysplasia of the kidney in adults have been reported. Many authors use different descriptions to characterize the same aspect of identical disease causing a confusing nomenclature. The disease is caused by a malformation in development. On the basis of three clinical case reports diagnosis, differential diagnosis and prognosis is discussed. One patient had an additional malformation (M. Klippel-Feil).

[Prevention of urinary calculi by means of a benzbromarone-citrate combination].

The action of a benzbromarone citrate combination (Harolan) in the prevention of uric acid and oxalate stones was studied in 21 patients suffering from calcium oxalate stones. The investigation was carried out under standard diet to eliminate possible variabilities in the excretion of lithogenous and inhibitory substances due to individual eating habits. Under Harolan a significant reduction in the serum uric acid level as well as simultaneous elevation of the urine pH-value could be achieved, the latter resulting in improved solubility of uric acid, the excretion of which is increased during the first days of treatment. Neither in 24 hour urine nor in the circadian rhythm was the solubility curve for uric acid exceeded during the test period. Side-effects were not observed.

Calorimetric investigation of the complex phase behavior of glucocerebroside dispersions.

The thermotropic behavior of aqueous dispersions of glucocerebroside from Gaucher's spleen has been investigated by different scanning calorimetry. These results indicate that glucocerebroside undergoes two distinct phase transitions centered at 47 and 83 degrees C, respectively. The high-temperature transition is associated with the main gel-liquid crystalline transition and has an enthalpy change of 13.6 kcal/mol of lipid; this transition is not rapidly reversible and the liquid crystalline phase supercools to a metastable gel phase. The low-temperature transition is exothermic with an enthalpy change of -6.3 kcal/mol and involves a transformation of the metastable gel phase to a more highly ordered gel conformation, without involving changes in the conformational state of the hydrocarbon chains. The behavior of these transitions as a function of the amount of water suggests that the origin of the metastability is related to a hydration-dehydration process of the cerebroside molecule. Experiments with synthetic-D-erythro-N-palmitoylglucocerebroside revealed the same thermotropic behavior. These glucocerebroside transitions are irreversible and together define a unidirectional cycle in which each state of the molecule can only be reached by completing the entire cycle.

Protein-lipid interaction. Biophysical studies of (Ca2+ + Mg2+)-ATPase reconstituted systems.

Differential scanning calorimetry, fluorescence spectroscopy and freeze-fracture electron microscopy have been applied to a study of the reconstituted Ca2+-ATPase proteins from sarcoplasmic reticulum when they are incorporated into pure lipid/water systems. The results obtained with these techniques have been used to examine the effects of this intrinsic protein upon the surrounding lipid at temperatures above and below the main lipid solid-fluid phase transition temperature (Tc). 1. Above this Tc value, the freeze-fracture data show that the proteins are randomly distributed within the plane of the bilayer. The fluorescence data show that as the protein content in the bilayer increases, so does the 'microviscosity'. 2. Below Tc the proteins occur in high protein to lipid patches, separate from the remaining crystalline lipid. The fluorescence data indicate that at these temperatures the presence of the protein causes a decrease in microviscosity, whilst the calorimetric data indicate a decrease in enthalpy of the main lipid transition. 3. A premelting of the high protein to lipid patches formed by phase separation within the lipid bilayers is indicated by the calorimetric and fluorescence data. This observation is used to rationalise the 'anomalous' properties of the dipalmitoyl phosphatidylcholine-ATPase of exhibiting activity at temperatures well below the lipid phase transition at 41 degrees C.

Perturbations of membrane structure by optical probes: II. Differential scanning calorimetry of dipalmitoyllecithin and its analogs interacting with Merocyanine 540.

Differential scanning calorimetry of multilamellar liposomes, interacting with the optical probe Merocyanine 540, yields quantitative information about perturbances of the bilayer structure induced by this dye. At low dye: lipid ratios, the dye perturbs primarily its own microenvironment, which is laterally separated from the unmodified lipid domain and exhibits modified thermotropic properties. A further increase in the dye concentration results in a perturbance of the whole lipid bilayer. The degree of perturbance is sensitive to structural modifications in the head-group region of the lipids. It is concluded that Merocyanine 540 reports in every case, even at infinite dilution, on localized events originating from a perturbed microenvironment.

A differential scanning calorimetry study of the interaction of gangliosides with peanut lectin, serotonin and daunomycin.

Thermotropic behaviour of human Tay-Sachs ganglioside and of mixed bovine brain gangliosides, before and after interaction with peanut lectin, serotonin and daunomycin, was investigated. Interaction of mixed brain gangliosides with peanut lectin or serotonin causes a decrease in the enthalpy of melting, whereas interaction of this lectin with Tay-Sachs ganglioside does not influence the enthalpy of melting. Serotonin causes a small increase in the enthalpy of melting of the Tay-Sachs ganglioside.

[Effect of azathioprine and prednisolone on recipients of xenografts presensitized with donor species-specific antigen (author's transl)].

Presensitization induced by pretreatment of the recipients with donor species-specific spleen antigen cannot be completely reversed by immunosuppressive drugs. However, there is a suppressive influence of the combined administration of azathioprine and prednisolone on hemagglutinating antibody production after transplantation as well as an effect on function and survival time of renal xenografts.

[Effect of anti-donor lymphocyte globulin on renal xenograft survived (author's transl)].

This paper reports about experimental xenografting of kidneys in closely related species (fox-dog) modifying primary rejection by treatment of the recipients with anti-donor lymphocyte globulin (ADLG). In respect of the phylogenetic constellations, ADLG contains lymphocytotoxic and hemagglutinating antibodies reactive in vitro with lymphocytes and erythrocytes of foxes as well as of dogs. Furthermore, donor species-specific papain-separated lymphocyte antigen is capable of reducing cytotoxicity of ADLG by adsorption of its lymphocytotoxic antibodies. In vivo ADLG, used as single therapy, has an immunosuppressive effect on kidney graft survival, which results in significant prolongation of mean survival time (9.6 +/- 1.5 days, n = 5).

[Percutaneous puncture of renal cysts for diagnostic and therapeutic purposes (author's transl)].

Percutaneous needle puncture of space-occupying lesions is an innocuous procedure in the differential diagnosis of renal masses. In conjunction with intravenous urography, ultrasonography and CT-scan it is a method of considerable diagnostic value, which will lead to a nearly complete accurate diagnosis. In this study we will report our experiences with diagnostic and therapeutic puncture of renal cysts. The instillation of Lipiodol has been given up, since a follow-up study became impossible due to radiological problems. The diagnostic procedure to differentiate expanding lesions of the kidney and the technique of percutaneous needle puncture are described as well as indications for surgical exploration.

[Induction of partial immune tolerance in a closely-related species system by a single injection of papain-extracted, donor species specific, lymphocyte membrane antigens].

This paper reports of experimental xenografting of kidneys in closely related species (fox-dog) modifying primary graft rejection by single i.v. injection of donorspecies-specific lymphocyte antigen in soluble form. Papain treatment was used to separate histocompatibility antigens from lymphocytes of donors. This antigen, in vitro, is capable of reducing cytotoxicity of xeno-antisera by adsorption of their lymphocytotoxic antibodies. A single intraoperative i.v. injection of this antigen to the transplant recipients results in the subsequent failure of humoral antibody production, thus this antigen-mediated effect is thought to be a state of B-cell tolerance. Survival times are more than doubled in comparison to untreated controls and last in average 14.6 +/- 1.74 days (n = 5). Histological studies demonstrate rather well conserved renal parenchym with sporadic infiltrations of round cells.

[Citric acid excretion in stone-formers and healthy controls on normal and standard diet (author's transl)].

The citric acid excretion and concentration in 24 hours urine was measured in 70 stone-formers and 16 normal subjects (controls) on normal food intake followed by 9 days on a standard diet. On days 9 and 10 on the standard diet urine was collected in 3 hours portions and citric acid and calcium was measured. From day 7 to day 10 on standard diet we found only a slight change in the 24-hour excretion of citric acid ("steady state"). The citric acid excretion and concentration is significantly lower in stone-formers than in healthy controls. In both groups there is a sinusoid rhythm in citric acid excretion and concentration throughout the 24 hour period. The calcium concentration is at its maximum when the citric acid concentration is at its minimum.

Differential scanning calorimetry of chromaffin granule membranes.

Differential scanning calorimetry thermograms of native chromaffin granule membranes exhibit several peaks in the 15-35 degrees C region. Extraction of cholesterol increases the size of the melting peaks. Addition of Ca2+ ions does not seem to influence the lipid transitions.
