Production and reproduction of early and late bred dairy heifers.

A total of 253 heifers bred at first estrus after 350 d of age (350-d breeding age group) and 249 contemporary heifers bred at first estrus after 462 d of age (462-d breeding age group) were used to study the effects of age at first breeding on productive and reproductive performances of first lactation heifers. Heifers of both breeding age groups were subject to similar feeding and management practices. The average age at first calving was 698 d for the 350-d breeding group and 796 d for the 462-d breeding group. Although not statistically significant, heifers of the 462-d breeding group tended to have a higher conception rate at first service (47 vs. 38%) and fewer days between first service and conception (39 vs. 44 d) than those of the 350-d breeding age group. Breeding heifers as early as 350 d of age has no adverse effects on calving ease or retained placenta but does result in calves 1.2 kg lighter at birth. Heifers of the 350-d breeding group had lower milk, protein, and fat yields at both 168 and 308 d of first lactation than those of the 462-d breeding group. A 1-d reduction in age at first calving decreased 308-d milk yield by 2.01 kg for the 350-d breeding group as compared with 4.74 kg for the 462-d breeding group.

Sequential estimation of genetic and phenotypic parameters in multitrait mixed model analysis.

Single-trait and multitrait (2-, 3-, 4-, and 5-trait) restricted maximum likelihood methods were applied to the same set of data with complete information on all traits. Results suggest that parameter estimates from a data set vary depending upon the type of analysis (single- or multitrait model) and upon the other traits included in multitrait analysis. The choice of parameter estimation method for a breeding design should be based on the breeding goal. In parameter estimation or sire evaluation, traits included in a multitrait analysis should correspond to the traits of interest in the breeding goal. Multitrait analysis explores all intercorrelations simultaneously in parameter estimation and thus provides a complete picture of all interrelationships among traits. In contrast, single-trait analysis produces pairwise (simple) correlations and ignores the possible contribution of other related traits under study to the pairwise correlation. The 5-trait model analysis through canonical transformation was about 300% more efficient in terms of computer time than single-trait model analysis of the same 5 traits. In this study, parameter estimates converged faster under multitrait analysis through canonical transformation than under single-trait analysis.

Seroepidemiology of adult T-cell leukemia virus infection and analysis of sero-positive cases in Taiwan.

For a seroepidemiologic study of adult T-cell leukemia virus (ATLV) infection in Taiwan, the gelatin particle agglutination technique and the indirect immunofluorescence method were used for anti-ATLV titration. Sporadic sero-positive cases were found all over the Taiwan districts except among the aborigines (0/947). Sero-positive rates ranged from 0 to 5.6% (except ATL family) and a total of 48 cases were found in 3682 Han-Chinese. Among them 9 cases were newly found in family surveys, and 39 cases were observed in random samples. As an average positive rate was 1.0%, by calculation about 80,000 sero-positive cases are supposed to be present in Taiwan. A most remarkable feature of the sero-positive cases was the high rate in couples. Various patterns of sero-positive cases existed in pedigrees. Anti-ATLV positive sera of Chinese living in Taiwan and Japanese were compared by immunoprecipitation and there was no difference between them. The possible infection route from Japan to Taiwan is discussed.

Studies of natural killer cell activity. I. Normal range of a natural killer cell activity in the healthy Chinese.

This study was undertaken to establish a normal distribution curve for natural killer (NK) cell activity in healthy Chinese. NK cell activity was detected in 123 persons of different age groups, from the newborn to 84 years of age. None had any reported infection. The mononuclear cells were obtained from peripheral blood and cord blood for NK test; K562 cells were used as target cells to assay NK cell activity. The results indicated that cord blood had the lower NK cell activity. Increasing NK cell activity was seen with increasing age. In the 20 year to 49 year range, the NK cell activity reached maximal value. The NK cell activity gradually decreased when the age was greater than 50 years.

Multitrait estimation of relationships of first-lactation yields to body weight changes in Holstein heifers.

An experimental population of 994 Holstein heifers from 56 sires was used to estimate simultaneously heritabilities and genetic and phenotypic correlations between first-lactation yields and prepartum and postpartum weight changes. Variance and covariance components were estimated by the multitrait restricted maximum likelihood method. Heritability estimates were .09, .15, and .32 for first-lactation milk, protein, and fat yields. Heritability estimates ranged from .20 to .34 for prepartum and postpartum body weights and weight changes of first lactation. Weight gain from 350 to 462 d of age was highly correlated, genetically and phenotypically, with first-lactation milk, protein, and fat yields. Genetic and phenotypic correlations between first-lactation yields and body weights at calving and at 56, 112, 168, 224, and 280 d postpartum were positive, suggesting that the larger heifers had higher lactation yields. In contrast, genetic and phenotypic correlations between yields and weight gains during the first lactation were negative, indicating that high-producing heifers gained less weight during lactation than low-producing heifers. Heifers lost an average of 23 kg from calving to 56 d postpartum and gained weight thereafter. On genetic and phenotypic scales, larger heifers at first calving lost more weight from calving to 56 d postpartum and gained less weight from 56 d postpartum onward than smaller heifers.

Nephrotic syndrome associated with varicella infection.

A 4-year-old boy developed nephrotic syndrome following varicella infection. Serologic studies during the early phase of the disease demonstrated a decrease in serum C3, C4, and properdin factor B. Renal biopsy revealed an acute proliferative glomerulonephritis with deposition of immunoglobulins A (IgA) and M, C3, C1q, and varicella virus antigen in the glomerulus, suggesting an immune complex deposition. Ultrastructurally, this suggested a postinfectious immune complex glomerulonephritis. These phenomena suggested that varicella virus antigen antibody complexes were deposited in the glomerulus and activated the classic and alternative pathway of complements, leading to an immune complex glomerulonephritis. During the nephrotic phase, an increase in OKT8 cells and decrease of the OKT4 cells were demonstrated. Two months later, this alteration returned to normal as the renal disease was in remission. This change of lymphocyte subsets during varicella infection may play a role in the pathogenesis of nephrotic syndrome.

T-cell subsets and response to mitogens in patients with steroid responsive nephrotic syndrome.

T-cell subsets, lymphocyte proliferative response, circulating immune complexes (CIC) and serum C3, C4 and properdin factor B were studied in 22 children suffering from steroid-responsive nephrotic syndrome (SRNS) during the acute, remission and relapse phases. Renal biopsies were performed on seven children, including three patients who relapsed after remission and all showed minimal change in their nephrotic syndrome. The lymphocyte subsets were normal in SRNS patients, but the three relapse patients had significant increase in OKT8 cells and decrease in OKT4/OKT8 ratio during both the acute and relapse phases. Use of autologous plasma will lower the lympho-proliferative response in SRNS patients during the onset of the disease and in relapse. The circulating immune complexes were detectable in seven cases during acute onset, but the serum levels of complements were normal. A review of the total data suggests that suppressor T-cell dysfunction may play a certain role in the immunopathogenesis of SRNS.

In vitro effect of a bovine thymic extract (thymostimulin) on T-cell differentiation in cord blood lymphocytes.

Cord blood lymphocytes were isolated from 40 normal newborns. The initial 20 samples were used to determine the dose-response curve of bovine thymic extract (Thymostimulin) by the measurement of active T cells. Results showed that the active T cells increased significantly when the Thymostimulin concentration was increased to 50 ng/ml and 100 ng/ml. The remaining 20 samples were divided into three portions and preincubated either with 50 ng and 100 ng of Thymostimulin or without Thymostimulin. The total T cells, active T cells, B cells, T-cell subsets, and lymphoproliferative responses to phytohemagglutinin, concanavalin A, and pokeweed mitogen were determined. The results showed that the active T cells, total T cells, B cells, OKT4 cells, and OKT8 cells were significantly increased after Thymostimulin treatment. The lymphoproliferative responses were also significantly increased. These data strongly support our conclusion that Thymostimulin has a marked stimulating effect on human lymphocyte proliferation and differentiation.

Studies on hyperimmunoglobulinemia E syndrome in five children.

T cell subpopulations, lymphoproliferative response, autologous mixed lymphocyte reaction (AMLR), antibody-dependent cellular cytotoxicity (ADCC), natural killer (NK) activity and chemotaxis were studied in five hyperimmunoglobulinemia E (hyper IgE) syndrome patients. Both absolute OKT8 (suppressor T) cells and OKIa1 cells were decreased in number. The OKT4/OKT8 ratio increased. The lymphoproliferative response showed that the patients' lymphocytes cultured in medium containing autologous plasma proliferated much less vigorously, as expressed by transformation index, than those cultured in AB healthy control plasma. The peak proliferative response in AMLR in patients was significantly lower than that in the control group. The proliferative response of the fresh autologous mononuclear cells (MNC) exerted suppressor activity in the patients, while those from the normal controls exerted helper activity. Supernatant from AMLR and an isolated T cell culture of patients exerted suppressor activity, while those from the normal controls had none. In the ADCC reaction, patient's polymorphonuclears were significantly impaired in target cell lysis. The NK activity and Leu-11 cells in the patients also showed a significant decrease. In vitro neutrophil chemotaxis was consistently impaired in two out of five patients. Therefore, there is evidence of abnormal regulatory T cell functions and defect in PMN-ADCC activity and NK lysis assay in patients with hyper IgE syndrome.

Acquisition of Thermotolerance in Soybean Seedlings : Synthesis and Accumulation of Heat Shock Proteins and their Cellular Localization.

When soybean Glycine max var Wayne seedlings are shifted from a normal growth temperature of 28 degrees C up to 40 degrees C (heat shock or HS), there is a dramatic change in protein synthesis. A new set of proteins known as heat shock proteins (HSPs) is produced and normal protein synthesis is greatly reduced. A brief 10-minute exposure to 45 degrees C followed by incubation at 28 degrees C also results in the synthesis of HSPs. Prolonged incubation (e.g. 1-2 hours) at 45 degrees C results in greatly impaired protein synthesis and seedling death. However, a pretreatment at 40 degrees C or a brief (10-minute) pulse treatment at 45 degrees C followed by a 28 degrees C incubation provide protection (thermal tolerance) to a subsequent exposure at 45 degrees C. Maximum thermoprotection is achieved by a 2-hour 40 degrees C pretreatment or after 2 hours at 28 degrees C with a prior 10-minute 45 degrees C exposure. Arsenite treatment (50 micromolar for 3 hours) also induces the synthesis of HSP-like proteins, and also provides thermoprotection to a 45 degrees C HS; thus, there is a strong positive correlation between the accumulation of HSPs and the acquisition of thermal tolerance under a range of conditions.During 40 degrees C HS, some HSPs become localized and stably associated with purified organelle fractions (e.g. nuclei, mitochondria, and ribosomes) while others do not. A chase at 28 degrees C results in the gradual loss over a 4-hour period of the HSPs from the organelle fractions, but the HSPs remain selectively localized during a 40 degrees C chase period. If the seedlings are subjected to a second HS after a 28 degrees C chase, the HSPs rapidly (complete within 15 minute) relocalize in the organelle fractions. The relative amount of the HSPs which relocalize during a second HS increases with higher temperatures from 40 degrees C to 45 degrees C. Proteins induced by arsenite treatment are not selectively localized with organelle fractions at 28 degrees C but become organelle-associated during a subsequent HS at 40 degrees C.

Clinical and immunopathologic study of mesangial IgM nephropathy: report of 41 cases.

Forty-one out of 408 cases (or 10%) of primary glomerular disease had diffuse fine granular to arc-like short linear mesangial deposits of IgM by direct immunofluorescence. The IgM deposition was accompanied by C1q and/or C4 in the same locality in 29 cases, by C3 in 10, and by trace amounts of IgA in 6. Properdin-factor B was not detected. Fine granular electron dense deposits of low density were detected in the mesangium in all 41 cases by electron microscopy, usually as a discrete granular or arc-like pattern beneath the mesangial glomerular basement membrane and correlated well with the immunofluorescence findings. An immune complex disease with complement activation via the classical pathway is suggested. The ages of the patients varied from 2 to 58 years (average 23.8 years). A male predominance of 2.2:1 was identified. Serum IgM level was elevated in 46.7% of the cases. The majority (87.8%) of the cases manifested a nephrotic syndrome or relapse at time of biopsy, and the remaining cases experienced persistent or intermittent proteinuria. Among the 36 nephrotic patients, 22 cases (61.1%) demonstrated complete remission with steroid therapy, 9 cases (25%) were resistant, and 5 cases (13.9%) had partial remission. Complete and partial remissions were later achieved with cytotoxic drugs or methylprednisolone pulse therapy in 3 and 4 cases respectively in the steroid resistant patients. Frequent relapses occurred during the course in 22 out of 32 cases (68.8%) who had experienced complete or partial remission. Follow-up study after biopsy demonstrated that sustained complete remission was achieved with prednisolone with or without cytotoxic drugs and pulse therapy in only 14 (42.4%) of the 33 nephrotic cases who had been followed up for longer than 6 months, and six of them had had previous relapses. Pathologically, 56.1% of the patients showed mild to moderate increase in mesangial matrix and cellularity. Focal and segmental sclerosis was demonstrated in four cases (9.8%). However, minimal glomerular change was also common (34.1%). The patients with minimal change seemed to have a higher complete remission rate than patients with more evident glomerular alterations, although the difference was not statistically significant. This clinical and immunopathological study suggests that mesangial IgM nephropathy is an important disease in Taiwan, with a variable response to treatment and frequent relapses.(ABSTRACT TRUNCATED AT 400 WORDS)

Reproductive performance of crossline and pureline dairy heifers.

Data of 2779 purebred and crossbred heifers collected from five research stations of Agriculture Canada were used to study additive and nonadditive genetic effects on ages at first heat and at first breeding and conception rate at first service. Of these heifers, 2378 heifers had information on ages at first conception and at first freshening, days from first service to conception, and gestation length. The model included station, year of birth, sire, breed additive, maternal, and heterosis effects where sire effects were treated as random. Station differences were a significant source of variation for all reproductive traits. Year of birth had significant effects on four of seven reproductive traits. Breed additive effects for all genetic groups were not significant except for Finnish Ayrshire and American Holstein. No significant maternal effects were detected. Of 21 combinations of heterosis effects, six combinations showed significance. Partial regression coefficients ranged from negative to positive, suggesting that breed additive, maternal, and heterosis effects could increase or decrease for each percent increase of genetic contribution, depending upon the trait, breed group, and type of inheritance.

Acute immune complex mediated glomerulonephritis in a Chinese girl with Wiskott-Aldrich syndrome variant.

This study presents a 12-year-old girl with Wiskott-Aldrich syndrome variant, who developed acute glomerulonephritis without history of transfer factor therapy and the efficacy of splenectomy for the control of the patient's thrombocytopenia. The patient presented with eczema, severe thrombocytopenia and immunodeficiency. The impaired immunity was featured by impaired delayed hypersensitivity and lymphoproliferative response to nonspecific mitogen, low serum IgM, low isohaemagglutinins, recurrent infections and high IgE. She developed hematuria about one month prior to admission. For her severe thrombocytopenia splenectomy was performed and proved to be effective. At the time of splenectomy, renal biopsy was done and showed proliferative glomerulonephritis with coarse granular deposition of immunoglobulins (IgA and IgM) and C3. Electron microscopy demonstrated granular electron-dense deposits in the glomerulus, indicating an immune complex glomerulonephritis.

Use of the pulmonary artery for left ventricular venting during cardiac operations.

Data relating to the hemodynamic efficaciousness and mechanism of action of a pulmonary artery catheter or vent used for left ventricular venting during cardiac operations are presented. The pulmonary artery vent is a plastic sump catheter that is introduced into the main pulmonary artery through a purse-string suture and connected via a roller pump to the venous reservoir of the heart-lung perfusion machine. Placement and removal require only a few minutes. The pulmonary artery vent retrieved 85% of a 99mtechnetium-labeled solution placed in the left atrium during aortic cross-clamping, and there was no detectable radioactivity in peripheral or aortic root blood samples. Pulmonary artery vent return during cardiopulmonary bypass in 10 patients undergoing coronary artery bypass averaged 12.5 L. The effectiveness of left ventricular decompression was evaluated in 20 patients also undergoing bypass grafting. Use of the pulmonary artery vent consistently and significantly decreased left heart pressures, compared to the control situation with the vent off, with the aortic cross-clamp applied, and in both the fibrillating and beating heart in the early postischemic reperfusion period. We reached the following conclusions: (1) The pulmonary artery vent withdraws left heart blood via the pulmonary vasculature, in addition to returning right heart spillover and retrieving bronchial flow. (2) Left heart pressures are reduced to levels which reduce oxygen demands and preserve endocardial perfusion, therefore protecting myocardium, during fibrillation and during coronary reperfusion of the beating heart. (3) Because of its effectiveness and safety, especially the impossibility of introducing air into the left ventricle, the pulmonary artery vent is recommended for routine left ventricular venting.

Acyl specificity in triacylglycerol synthesis by mammary adenocarcinoma R3230AC in Fischer rats.

Activities and acyl specificities of both sn-glycero-3-phosphate and diacylglycerol acyltransferases in microsomal fractions isolated from homogenates of the mammary adenocarcinoma R3230AC carried by Fischer rats were compared to those from normal mammary glands of lactating Fischer rats. Although the neoplasm exhibited lower activities for these two enzyme reactions, the specificities for acyl-CoAs as donors were quite similar to those found in the normal tissue counterpart. Long-chain acyl-CoAs were preferred substrates for the sn-glycero-3-phosphate acyltransferase reaction while acyltransferase with diacylglycerol as acceptor showed much less preference. With both normal and neoplastic tissues, the products of each reaction were the same, i.e., phosphatides with sn-glycero-3-phosphate and triacylglycerol with diacylglycerol as acyl acceptors, respectively. All results support the concept of a non-random distribution of fatty acids in the triacylglycerol of this mammary adenocarcinoma in virgin rats which is the same as that from the normal tissue in lactating animals.
