Immunoadsorbents for the isolation of high-affinity anti-hapten antibodies in high yield.

Anti-hapten antibodies with affinities close to 10(8)/M can be isolated in high yield by elution under mild conditions from immunoadsorbents prepared by coupling haptens directly to an aminoethylamino (AEA) derivative of beaded agarose. In experiments with rabbit anti-2,4-dinitrophenyl (DNP) antibody with an average association constant (Ko) of 1.2 X 10(9)/M, elution of antibody bound to a Sepharose-AEA-DNP column produced a yield of 57%. With rabbit antibody to the 5-dimethylaminonaphthalene-1-sulfonyl (DANS) hapten with Ko=2.7 X 10(8)/M and a Sips heterogeneity index a=0.52, a 20% yield of the antibody found in the starting material by quantitative precipitin analysis was obtained by specific precipitation and dissolution of the precipitate with 0.18 M Na DANSulfonate; this antibody had Ko=6.6 X 10(8)/M and a=0.66. Purification of another aliquot of the same antibody preparation by immunoadsorption on DANS-AEA-Sepharose and elution with the same hapten concentration produced a yield of 75% of antibody with Ko=5.7 X 10(7)/M and a=0.29. In 3 production runs with the new method, apparent yields of 181, 101 and 94% (based on quantitative precipitin analysis of the starting material) of anti-DANS antibody were obtained. In 2 of these runs, the immunoadsorbent was further eluted with 0.1 M acetic acid and then with 0.1 M acetic acid in 10% dioxane; additonal yields of 9 and 4% were obtained with acetic acid and further yields of 15 and 11% with acetic acid/dioxane; the antibody eluted with acetic acid was damaged as shown by its failure to mediate passive immune lysis of DANS-coupled sheep erythrocytes, but the antibody eluted with acetic acid/dioxane was recovered intact.

In vitro inhibition of tritiated thymidine uptake in Morris hepatoma cells by normal rat liver extract: a possible liver chalone.

A phosphate-buffered saline extract of normal rat liver tissue almost completely inhibited DNA synthesis in Buffalo rat Morris hepatoma 7777 cells in vitro. This effect was tissue-specific because it did not occur with extracts of kidney, spleen, heart, lung, muscle, and hepatoma. The liver extracts did not inhibit in vitro human prostate carcinoma or phytohemagglutinin-stimulated human and rat peripheral blood lymphocyte cultures. The addition of 10% fetal calf serum in the incubation medium had no effect on this inhibition. We determined that doses of liver extract that inhibit thymidine incorporation were not toxic inasmuch as treated cells remained viable, as indicated by trypan blue exclusion. The liver extract may thus contain a cell-specific mitotic inhibitor, a chalone for hepatoma cells.

A study of dust-induced lymphocyte blastogenesis.

The in vitro measurement of dust allergen-stimulated lymphocyte blastogenesis by the incorporation of H3-thymidine can be a useful measurement for the evaluation of the cellular immune function in dust allergy patients. Dust allergy patients receiving immunotherapy had lower lymphocyte responses than those not receiving immunotherapy. Additionally, dust allergy patients as a whole showed higher lymphocyte stimulation indices than normal control.

A new allergen from the perienteric fluid of Ascaris suum with respect to charges.

A NEW ALLERGEN WITH PI in the 6.0 region was obtained from the perienteric fluid of Ascaris suum through isoelectric focusing techniques. This new allergen shows strong allergenicity, at a concentration of 10(-9) microgram/ml, in rats passively sensitized with the reaginic antiserum produced against Ascaris perienteric fluid.

Reaginic immune response to tree pollen antigen in LBN rats.

The immunogenicity and cross-antigenicity of ten common tree pollen allergens were studied in LBN rats. The rats produced IgE antibodies to each of the allergens used (maple, willow, poplar, ash, oak, sycamore, hickory, walnut, birch, and elm), yet the allergens had extremely limited cross-reactivity. The possible explanation for the limited cross-antigenicity is discussed.

Family study HLA antigen and Ir gene linkage in ragweed and dust allergy.

A family was studied for HLA antigen Ir-gene linkage in ragweed and dust allergy by clinical history, skin test, mixed lymphocyte cultures, lymphocyte culture for cellular immunity to antigen E and dust allergens, RAST assay for ragweed and dust antigens and HLA typing. These results confirm previous observations that ragweed Ir genes are linked to the inheritance of the HLA complex. The Ir genes may map at some distance from the closely linked HLA and MLC genes. It is also of interest that Ir genes for dust allergy in this family appear closely associated with ragweed genes. The set of twins with identical HLA complexes but different expression of presumably identical Ir genes will bear watching.

Separation of six bovine complement components and one inactivator (1, 2).

Six components (C1, C5, C6, C7, C8 and C9) of bovine complement and one inactivator (C3 in) could be separated from bovine serum. Bovine C1 was separated by precipitation at low molarity (0.03 M of relative salt concentration) other components by DEAE-cellulose chromatography using 0.005 M sodium phosphate buffer, pH 7.5, as a base for solvents having the relative salt concentration adjusted by addition of NaCl from 0.03 to 0.3 M. The separated bovine complement components could be tested using intermediates formed from sheep erythrocytes, rabbit hemolysin, guinea pig C1 and remaining human complement components. C2, C3 and C4 of bovine origin remained undetected either because of incompatibility with the intermediates used or interference of inhibitors or inactivators.

Cooperative binding of a complement component to antigen-antibody complexes. III. Complexes containing IgM antibodies.

The first component of guinea pig complement (C1)2) is bound in a cooperative manner to antigen-antibody complexes containing rabbit IgM antibodies, as was previously shown to be the case for IgG antibodies. The shape of the binding curves is consistent with an allosteric mechanism involving clusters of 10 interacting C1 binding sites. Similar results were obtained with IgM antibodies against an artificial hapten and against a natural constituent of the erythrocyte membrane.

Effect of hypocapnia in the first three days of life on the subsequent development of periventricular leukomalacia in premature infants.

Several etiologies for causing periventricular leukomalacia (PVL) in preterm infants have been investigated worldwidely. The aim of this study is to investigate whether hypocapnia plays a role in the pathogenesis of the subsequent development of PVL. From Jan. 1997 to June 1998, 91 premature infants (mean gestational age 28.8 +/- 2.5 weeks and birth body weight 1169 +/- 385 g) born in Chung Gung Children's Hospital requiring mechanical ventilation in the first 72 hrs were enrolled in our study. Serial neuorosonogram were followed on day 1, 3, 7 and then at least every 2 to 3 weeks until discharge. These infants were divided into two groups defined as the PVL group and non-PVL group. Variables comprising perinatal data were analyzed and compared between these two groups to screen out possible risk factors related to the development of PVL. There was no difference noted. As for the effect of PaCO2, we found the infants with PVL to have mean lower PaCO2 during the first 72 hours of life. (P = 0.004). Logistic regression model revealed that the incidence for PVL increased significantly when mean PaCO2 is lowered to less than 25 mmHg. Hypocapnia during the first three days of life seems to be an important etiological factor associated with the development of periventricular leukomalacia. Therefore, the ventilatory strategy in the care of the premature neonates should not only provide adequate ventilation but also careful avoidance of hypocapnia.

Hepatoblastoma in infancy and childhood: a clinical and pathological study of 32 cases.

During the twenty-nine-year period from June 1959 to October 1988, 32 infants and children with histology-proved hepatoblastoma were seen at the Department of Pediatrics, National Taiwan University Hospital (NTUH). The age at diagnosis ranged from four days to six years, with a mean of one year and eight months. Most of the patients (87.5%) were under the age of three. There was no sex predilection (16 males, 16 females). The most common presenting signs were an upper abdominal mass (71.9%) and/or abdominal distention (62.5%). Physically, all the patients revealed hepatomegaly, with or without mass. Laboratory abnormalities included primarily thrombocytosis (64.3%), elevated serum aspartate aminotransferase (75.0%), serum alanine aminotransferase (62.5%), cholesterol (78.3%) and alphafetoprotein (92.3%). According to the classification of Ishak and Glunz, 56.3% of these belonged to the epithelial type; the others belonged to the mixed type. During the follow-up period, 6 patients with complete resection of the tumor remained alive from 2 years to 17 years and 4 months. Twenty-two patients died from 8 days to 16 months after diagnosis. Four patients were lost to follow-up. The two-year survival rate was 21.4% (6/28). Complete resection of the tumor was the key treatment for achieving survival.

Mis-swallowing of cleaning naphtha: report of three cases.

Cleaning naphtha, a product of Chinese Petroleum Corporation, is a complex hydrocarbon mixture which contains mainly aliphatic hydrocarbons. It is used as a cleaning fluid and solvent. Three toddlers mis-swallowed a mouth of cleaning naphtha accidentally. One developed multiple organ failure before death, another completely recovered without sequelae, and the other died very soon after mis-swallowing. Two fatal cases were both induced vomiting with aspiration immediately after mis-swallowing by their parents. Therefore, inducing vomiting should not be encouraged especially at home, and prevention of aspiration is very important in the management of such cases.

A fatal case of viral pneumonia in a child infected with adenovirus type 3.

This report describes the clinical, roentgenologic, pathologic, and virologic findings in a 2 years and ten months old girl who died from a severe pneumonia. Initially, the patient presented with fever and cough for 2 days. Physically, the patient appeared lethargic, and breathing sounds revealed diffuse rales and wheezing. Hemogram showed mild leukocytosis and lymphocyte predominant. Chest X ray revealed diffuse interstitial infiltration of the right upper lung, left upper and left lower lung field. Bacteria infection was first impressed. Although treated with several antibiotics (ampicillin, cefuroxime, amikacin, penicillin, cephazolin, imipenem and vancomycin) in three different hospitals, the patient's condition went downhill and the patient died 2 weeks later. Finally, adenoviurs type 3 was isolated from sputum specimen taken before death and necropsy lung tissue. The lung pathology showed diffuse necrotizing inflammation with fibrinopurulent exudate, and eosinophilic intranuclear inclusion bodies were also noted in the alveolar cells. These data proved the diagnosis of adenovirus pneumonia. This case adds to the evidence that adenovirus type 3 infection during childhood may cause fatal disease.

Dyskeratosis congenita preceded by severe aplastic anemia: report of one case.

Dyskeratosis congenita is a rare hereditary disease which usually manifests with skin hyperpigmentation, nail dystrophy, and leukoplakia of the mucous membrane (triad). This report describes a six-year-old boy with severe aplastic anemia who was later diagnosed to have dyskeratosis congenita. His unusual presentation was pancytopenia followed by leukoplakia of the tongue, hyperpigmentation of the skin and dystrophy of the nails. Treatment with horse anti-human lymphocyte immunoglobulin (ALG) for his aplastic anemia was not effective.

Gastric perforation in the neonate: clinical analysis of 12 cases.

Gastric perforation is a rare abdominal catastrophe which associated with high mortality in newborn infants. From June 1978 to July 1991, twelve cases of neonatal gastric perforation presented at Chang Gung Memorial Hospital. Male to female ratio was 9:3. The most common presenting signs were abdominal distension (100%), feeding intolerance (92%), respiratory distress (67%) and poor activity (58%). All cases had significant symptoms between two and five days of age. All of the abdominal plain film showed pneumoperitonium. The most common site of perforation was the great curvature of the stomach (83%). Among the 10 pathological reports available, 8 cases had ischemic change and 2 cases had hemorrhage and inflammatory cell infiltration. There was a high mortality rate of 58% in this series. Male, hyponatremia (serum sodium < 130 meq/l) and metabolic acidosis (pH < 7.3) were poor prognostic factors. This report suggests that early diagnosis and early management before clinical deterioration of the metabolic status may improve prognosis for neonatal gastric perforation patients.
