[A new zoonosis--investigation of Gardnerella vaginalis disease of fox. V. Studies serotype of Gardnerella vaginalis in fox].

145 strains Gardnerella vaginalis isolated in foxes were isolated from 13 main farms raising foxes in six provinces (regions), China, after antigenicity and immunogenicity of the strains were measured, 1-3 appropriate strains were selected from each farm raising foxes for serotype studies. Cross agglutinin absorption test confirmed that selected 26 strains Gardnerella vaginalis were divided into three serotypes and then the representing strains were used to produce typing serum. Among remaining 119 strain, 108 strains were typable with the typing sera, and 11 strains can't be set. Among three serotypes, serotype I made up 79.1% of the strains. It was shown that serotype I was the principal serotype of Gardnerella vaginalis of fox in China. The test also confirmed that 5 strains of Gardnerella vaginalis isolated from racoon dog, 4 strain Gardnerella vaginalis from mink and 2 strains Gardnerella vaginalis from canine also belonged to serotype I. Supersonic antigben was produced with three serotypes, representative strains. By agar immuno-diffusion test, it confirmed that the antigens of three serotypes formed a obvious blending precipitating line with the homologous or heterologous serotype antiserum. It indicated common antigen existed among all serotypes. The agar immuno-diffusion test results revealed that the precipitating line of the homologous serotype completely blended. It is our opinion that the method of serotyping is reliable.

[The effect of mitomycin C on activities of carbonic anhydrase and ATPase of the ciliary body epithelium in rabbit].

PURPOSE: Enzymohistochemical methods were performed to observe the effect of mitomycin C(MMC) on the activities of carbonic anhydrase (CA) and ATPase of the ciliary body epithelium in rabbits after using 0.2mg/ml mitomycin C during sclerectomy for the further study on the mechanism of the decrease of intraocular pressure postoperatively. METHODS: 0.2mg/ml mitomycin C was topically applicated during the sclerectomy in rabbit eyes. 7 days postoperatively, Hasson's method and plumbum nitrate's method were used to stain the CA and ATPase of the ciliary body epithelium. The grey value of the enzymes was quantitated by Image Analysor. RESULTS: MMC inhibited the activities of CA and ATPase of the ciliary body epithelium underlying the site of MMC application, but the difference of enzymatic activities of the region 90 degree apart from the surgical site and that of surgical site in normal saline control group was not statistically obvious (P < 0.05). CONCLUSIONS: MMC could inhibit the activities of CA and ATPase of ciliary body epithelium, thus leading to the decreased secretion of humor aqueous and the lowerdown of intraocular pressure even persistent hypotony.

Regulatory elements in the promoter region of the renal kallikrein gene in normotensive vs hypertensive rats.

The renal kallikrein-kinin system has been implicated in the pathogenesis of hypertension. The expression level of the renal kallikrein gene in the kidney is significantly lower in spontaneously hypertensive rats (SHR) as compared with that of normotensive (SD and WKY) rats. Deletion analysis showed that the fragment -356/-188 of the promoter contains a transcriptional silencer(s) and the GC rich region located between -77 and -187 is the minimal essential element for directing the expression of the CAT reporter gene in mouse L cells. In the kidney of normotensive vs hypertensive rats, the nuclear protein factors NF1/CTF and SP1 bind differently to the renal kallikrein promoter, but similarly in the salivary gland. The differential transcriptional regulation of the rat renal kallikrein gene in the kidney may be responsible for the genetic difference between normotensive and hypertensive rats.

Intracoronary low-dose beta-irradiation inhibits neointima formation after coronary artery balloon injury in the swine restenosis model.

BACKGROUND: Neointima formation contributing to recurrent stenosis remains a major limitation of percutaneous transluminal angioplasty. Endovascular low-dose gamma-irradiation has been shown to reduce intimal thickening (hyperplasia) after balloon overstretch injury in pig coronary arteries, a model of restenosis. The objective of this study was to determine whether the use of a beta-emitting radioisotope for this application would have similar effects and to examine the dose-response relations with this approach. METHODS AND RESULTS: Normal domestic pigs underwent balloon overstretch injury in the left anterior descending and left circumflex and coronary arteries. A flexible catheter was introduced by random assignment into one of these arteries and was afterloaded with a 2.5-cm ribbon of encapsulated 90Strontium/90Yttrium sources (90Sr/Y, a pure beta-emitter). It was left in place for a period of time sufficient to deliver one of four doses: 7, 14, 28, or 56 Gy, to a depth of 2 mm. Animals were killed 14 days after balloon injury, the coronary vasculature was pressure-perfusion fixed, and histomorphometric analysis of arterial cross sections was performed. All arteries treated with radiation demonstrated significantly decreased neointima formation compared with control arteries. The ratio of intimal area to medial fracture length was inversely correlated with increasing radiation dose: control (no radiation), 0.47; 7 Gy, 0.34; 14 Gy, 0.20; 28 Gy, 0.08; and 56 Gy, 0.02 (r = -.78, P < .000001). Scanning electron microscopy demonstrated a confluent layer of endothelium-like cells both in control and in 14 Gy-irradiated arteries. There was neither evidence of significant necrosis nor excess fibrosis in the media, adventitia, or perivascular space of the coronary arteries or adjacent myocardium in the irradiated groups. Furthermore, the exposure to the staff and the total body exposure to the pig with the beta source was a small fraction of the dose previously measured and calculated with 192Ir, a gamma-emitting radioisotope. CONCLUSIONS: Administration of endovascular beta-radiation to the site of coronary arterial overstretch balloon injury in pigs with 90Sr/Y is technically feasible and safe. Radiation doses between 7 and 56 Gy showed evidence of inhibition of neointima formation. A dose-response relation was demonstrated, but no further inhibitory effect was seen beyond 28 Gy. These data suggest that intracoronary beta-irradiation is practical and feasible and may aid in preventing clinical restenosis.

Expression and signaling specificity of the IFNAR chain of the type I interferon receptor complex.

The IFNAR chain of the type I interferon (IFN) receptor (IFNIR) undergoes rapid ligand-dependent tyrosine phosphorylation and acts as a species-specific transducer for type I IFN action. Using the vaccinia/T7 expression system to amplify IFNAR expression, we found that human HeLa-S3 cells transiently express high levels of cell surface IFNAR chains (approximately 250,000 chains per cell). Metabolic labeling and immunoblot analysis of transfected HeLa cells show that the IFNAR chain is initially detected as 65-kDa and 98-kDa precursors, and then as the 130-kDa mature protein. Due to variation in N-glycosylation, the apparent molecular mass of the mature IFNAR chain varies from 105 to 135 kDa in different cells. IFNIR structure was characterized in various human cell lines by analyzing 125I-labeled IFN cross-linked complexes recognized by various antibodies against IFNIR subunits and JAK protein-tyrosine kinases. Precipitation of cross-linked material from Daudi cells with anti-IFNAR antibodies showed that IFNAR was present in a 240-kDa complex. Precipitation of cross-linked material from U937 cells with anti-TYK2 sera revealed a 240-kDa complex, which apparently did not contain IFNAR and was not present in IFN-resistant HEC1B cells. The tyrosine phosphorylation and down-regulation of the IFNAR chain were induced by type I IFN in several human cell lines of diverse origins but not in HEC1B cells. However, of type I IFNs, IFN-beta uniquely induced the tyrosine phosphorylation of a 105-kDa protein associated with the IFNAR chain in two lymphoblastoid cell lines (Daudi and U266), demonstrating the specificity of transmembrane signaling for IFN-beta and IFN-alpha through the IFNAR chain.

The human and mouse receptors for hyaluronan-mediated motility, RHAMM, genes (HMMR) map to human chromosome 5q33.2-qter and mouse chromosome 11.

The gene for the receptor for hyaluronan-mediated motility, RHAMM (designated hyaluronan-mediated motility receptor, HMMR (human) and Hmmr (mouse), for mapping purposes), was localized to human chromosome 5q33.2-qter by somatic cell and radiation hybrid analyses. Investigation of two interspecific backcrosses localized the mouse RHAMM (Hmmr) locus 18 cM from the centromere of mouse chromosome 11 within a region of synteny homology with human chromosome 5q23-q35 genes. The map position of the human RHAMM gene places it in a region comparatively rich in disease-associated genes, including those for low-frequency hearing loss, dominant limb-girdle muscular dystrophy, diastrophic dysplasia, Treacher Collins syndrome, and myeloid disorders associated with the 5q- syndrome. The RHAMM gene location and its ability to transform cells when overexpressed implicate RHAMM as a possible candidate gene in the pathogenesis of the recently described t(5;14)(q33-q34;q11) acute lymphoblastic leukemias.

Intracoronary radiation before stent implantation inhibits neointima formation in stented porcine coronary arteries.

BACKGROUND: Stent implantation has been shown to reduce restenosis by establishing a larger lumen but not by reducing neointima formation. We have previously shown that ionizing radiation reduced neointima formation after balloon injury in a swine model of restenosis. The purpose of this study was to determine whether endovascular irradiation of the coronary artery before stent implantation would affect neointima formation. METHODS AND RESULTS: Nine normolipemic pigs underwent coronary angiography, and segments of the left anterior descending and left circumflex arteries were chosen as targets for stenting. A high-activity 192Ir source was used to deliver 14 Gy by random assignment to one of the vessels. After this, 3.5-mm tantalum stents were implanted in both arteries. Three additional pigs were treated with a 90Sr/Y source (a pure beta-emitter) delivering 14 Gy to five segments of coronary vessels that were stented immediately after irradiation. Stent-to-artery ratio was similar in the radiated and the control arteries. Animals received aspirin 325 mg daily and were killed at 28 days. The intimal area was significantly reduced in the irradiated stented arteries compared with control arteries treated with stent only (1.98 mm2 with 192Ir and 2.53 mm2 with 90Sr/Y versus 3.82 mm2 in the control stented arteries, P < .005). CONCLUSIONS: Endovascular radiation before coronary stenting reduces neointima formation and may further reduce the restenosis rate after stent implantation.

Induction of apoptosis by beta-lapachone in human prostate cancer cells.

beta-Lapachone, a plant product, has been shown to be a novel inhibitor of DNA topoisomerase I, with a mode of action different from camptothecin and a chemical structure distinct from those of current anti-cancer drugs. We observed that beta-lapachone, at concentrations of less than 8 microM, induces cell death with characteristics of apoptosis in human prostate cancer cell lines. This effect of beta-lapachone was also observed in a human promyelocytic leukemia cell line (HL-60). beta-Lapachone-induced apoptosis is independent of p53 expression, and ectopic overexpression of bcl-2 did not confer significant resistance to beta-lapachone. Among other human carcinoma and adenoma cell lines tested, human breast and ovary carcinoma showed sensitivity to the cytotoxic effect of beta-lapachone without manifesting signs of apoptosis. These results suggest that beta-lapachone is a potential compound to be added to cancer chemotherapy, particularly for prostate cancer.

Overexpression of the hyaluronan receptor RHAMM is transforming and is also required for H-ras transformation.

Overexpression of the RHAMM gene by transfection into fibroblasts is transforming and causes spontaneous metastases in the lung. H-ras-transformed fibrosarcomas transfected with a dominant suppressor mutant of RHAMM exhibit a so-called revertant phenotype and are completely nontumorigenic and nonmetastatic. Conversely, fibroblasts stably expressing low levels of RHAMM as a result of antisense transfection are resistant to ras transformation. Collectively, these results indicate that RHAMM acts downstream of ras. The loss of functional RHAMM ablates signaling within focal adhesions, in particular changes in focal adhesion kinase phosphorylation, and as a result these focal adhesions are unable to turn over in response to hyaluronan. These results provide evidence of the oncogenic potential of a novel extracellular matrix receptor and establish a functional link between transformation by ras and signaling within focal adhesions that are required for transformation by this oncogene.

Identification of a domain in the angiotensin II type 1 receptor determining Gq coupling by the use of receptor chimeras.

The angiotensin II type 1 (AT1R) and type 2 (AT2R) receptors belong to the seven transmembrane receptor superfamily. Previous studies have suggested that the AT1R couples to a Gq signaling pathway, whereas the AT2R does not associate with Gq. To identify the role that individual intracellular domains play in AT1R function, AT1R/AT2R chimeric receptors were prepared by substitution of intracellular loops. CHO cells expressing these chimeras were used to test angiotensin II-induced c-fos expression and Ca2+ mobilization which are involved in the AT1R signaling pathway through Gq coupling. Substitution of the second intracellular loop (IC2) and the cytoplasmic tail between the two receptors did not affect AT1R function. However, exchange of the third intracellular loop (IC3) resulted in the loss of function in the AT1R and conferred to the AT2R the ability to constitutively activate the fos promoter. These findings suggest that the third intracellular loop of the AT1R is critical for Gq coupling. Substitution of discrete amino acid sequences of the third intracellular loop indicate that its N-terminal and C-terminal portions, especially the seven amino acids 219-225 in the N-terminal portion, are important for AT1R function, and that the intermediate portion of this loop is not required for Gq coupling.

Reciprocal modulations between p53 and Tat of human immunodeficiency virus type 1.

Infection by human immunodeficiency virus type 1 (HIV-1) causes acquired immunodeficiency syndrome (AIDS) after a long clinical latency. This disease is associated with a spectrum of cancers. Here we report that wild-type p53 is a potent suppressor of Tat, a major transactivator of HIV-1. Reciprocally, Tat inhibits the transcription of p53. Downregulation of p53 by upregulated tat may be important for the establishment of productive viral infection in a cell and also may be involved in the development of AIDS-related malignancies.

Genomic instability of microsatellite repeats in prostate cancer: relationship to clinicopathological variables.

Sixty-six patients with prostatic adenocarcinoma were screened for somatic instability at 8 microsatellite marker loci on 5 chromosomes. Differences in unrelated microsatellites for tumor and normal DNA were detected in 13 (19.7%) patients. Only extraglandular spread (nodal involvement and distant metastasis) was found to show significant association with somatic instability after controlling for other clinicopathological variables (P < 0.05). Microsatellite instability may possibly occur during the early stages of neoplastic transformation in a subset of prostate cancer rather than as a late event. This may be related to a phenotype with growth advantage. The frequency of this mutator phenotype is much higher in the United States than Japan, reflecting racial differences in the molecular tumorigenesis of this malignancy.

Hemolytically inactive C5b67 complex: an agonist of polymorphonuclear leukocytes.

The activity of hemolytically inactive C5b67, designated iC5b67, was evaluated as an agonist for functional responses of human polymorphonuclear leukocytes (PMN). C5b67 was formed from purified human complement components and decayed in phosphate-buffered saline (PBS) until it had no lytic activity for sheep erythrocytes in a standard assay. iC5b67, at nanomolar concentrations, stimulated PMN chemotaxis and Ca2+ fluxes, but inhibited superoxide production and failed to upregulate CR1 and CR3. There was no significant contamination of the iC5b67 with C5a to explain these results. Neither isolated C5b6 nor C7 alone exhibited the activities of iC5b67, while insolubilized anti-C7 could remove the PMN agonist activity from the iC5b67 preparation. Binding studies to define a specific receptor for iC5b67 on PMN were hampered by the very hydrophobic nature of the ligand. 125I-iC5b67, by contrast to hemolytically active 125I-C5b67, was unable to insert in erythrocytes, suggesting that iC5b67 need not insert in the PMN membrane to induce signaling. Two lines of evidence suggest that iC5b67 and C5a and FMLP share common steps in intracellular signaling (1) pretreatment of PMN with iC5b67 deactivates PMN for C5a- and FMLP-induced chemotaxis; and (2) pretreatment of PMN with pertussis toxin inhibits iC5b67-induced chemotaxis. Thus, iC5b67 has important effects on the activity of PMN and G-proteins and Ca2+ are involved in the signaling.

Induction of apoptosis in uninfected lymphocytes by HIV-1 Tat protein.

Infection by human immunodeficiency virus-type 1 (HIV-1) is typified by the progressive depletion of CD4 T lymphocytes and deterioration of immune function in most patients. A central unresolved issue in acquired immunodeficiency syndrome (AIDS) pathogenesis is the mechanism underlying this T cell depletion. HIV-1 Tat protein was shown to induce cell death by apoptosis in a T cell line and in cultured peripheral blood mononuclear cells from uninfected donors. This Tat-induced apoptosis was inhibitable by growth factors and was associated with enhanced activation of cyclin-dependent kinases.

Intracellular maturation of the Newcastle disease virus fusion protein is affected by strain differences in the predicted amphipathic alpha-helix adjacent to the fusion domain.

The fusion (F) glycoprotein of Newcastle disease virus (NDV) contains a predicted amphipathic alpha-helix C-terminal to its fusion domain. Of the 13 available NDV F protein sequences, only the Australia-Victoria (AV) strain alpha-helix is weakened, by the replacement of Ala159 with Thr. In this report, we demonstrate that the efficiency of cleavage and virion incorporation of the AV F protein, unlike that of other strains, is temperature sensitive. Pulse/chase experiments at 42 degrees revealed disulfide-linked aggregates containing both the F and hemagglutinin-neuraminidase glycoproteins in strain AV, but not in strain Beaudette C. Furthermore, a revertant derived from AV, whose helix-weakening Thr159 has been replaced with the consensus Ala, produced fewer F protein aggregates, confirming the structural importance of this region in maturation. In addition, a novel disulfide-defined folding intermediate of the F protein was detected.

Early detection of apoptosis in defined lymphocyte populations in vivo.

Recently many methods have been developed for the detection of apoptosis. However, all of them have some limitations in determining whether specific subsets of cells are undergoing apoptosis. In this paper we describe a technique in which one simultaneously stains for cell surface markers with fluorescent monoclonal antibodies and for nuclear DNA breaks using in situ DNA nick translation detectable by fluorescence. The method has been evaluated using radiation-induced programmed cell death of lymphocytes and compared with some other techniques. It was found that the method is very specific and sensitive. It enabled us to enumerate apoptotic cells at the single cell level and simultaneously determine their subset-specific surface antigen profile both in vivo and in vitro. It is also insensitive to nicks present in replicating cells. Our data suggest that this method may be useful for the study of programmed cell death of antigen specific T cells in vivo.
