[An investigation and design of apparatus for measuring fetal lung maturity].

Clinically, the authorized method for determing fetal lung maturity is the measurement of the ratio of surface-active L/S in amniotic fluid. To set up new, simple, rapid and accurate method, an apparatus for measuring fetal lung maturity was made. The paper describes the basic principle and working procedure of the apparatus. 148 amniotic fluid specimens from different pregnancies were measured by means of the apparatus. The result was compared with the L/S. The sensitivity of measurement was 95%, specificity 91%, and accuracy 93%. The operation of the apparatus is simple, convenient and rapid. A small quantity of amniotic fluid is needed. In conclusion, the expected result was attained. The apparatus is of great importance to clinical application.

[Transsphenoidal microsurgical removal of GH-secreting pituitary adenoma in: a report of 200 cases].

From July 1979 to April 1992, 212 cases of GH-secreting pituitary adenoma were treated transsphenoidally by microsurgical technique, of whom two hundred cases monitored with their GH levels at preoperative and postoperative periods. The majority of cases totally 132 cases were of macroadenoma, and 68 cases were of microadenoma. 138 cases were operated on after April 1986. The cure and remission rate averaged to 72.5%, rising from 44.4% (before 1986) to 80.5% for microadenoma and from 31.4% to 69.1% for macroadenoma due to improvement of technique and accumulation of experience. One patient died of angiocardiopathy suddenly during operation. Mortality rate was 0.5%. The follow-up observation of postoperative GH levels in 52 cases revealed that the GH levels within 2 weeks after operation were the same as in 3-6 months. It is thought that the early GH levels may represent the surgical results. The surgical technique is described and factors influencing the surgical results, prevention and treatment of operative complications are discussed in this paper.

Electrophoresis of proteins and protein-protein complexes in native polyacrylamide gels using a horizontal gel apparatus.

Electrophoresis of proteins and protein-protein complexes in polyacrylamide gels under native conditions using a horizontal gel apparatus is described. The advantage of this system is that it permits the detection of both negatively and positively charged proteins as well as protein-protein complexes in the same gel. During electrophoresis, a continuous gel sandwiched between two glass plates is placed horizontally on the platform and submerged in a reservoir buffer. The sample wells are made along the center of the gel, allowing positively and negatively charged proteins to migrate toward the cathode and anode, respectively. Several proteins with varying molecular weights and isoelectric point (pI) values and pairs of proteins capable of forming protein-protein complexes were chosen as model systems to illustrate the methodology. The effects of several parameters on the performance of the gel system including protein molecular weight, pI, and gel concentration were also examined and the results obtained by this method are comparable to those obtained by the vertical system. Following electrophoresis, both negatively and positively charged proteins as well as protein-protein complexes can be transferred by electroblotting onto polyvinylidene difluoride membranes for further analyses.

Sterile ocular inflammatory reactions to monofilament suture material.

OBJECTIVE: To report six cases of sterile inflammatory reactions to fine monofilament suture materials, which is exceedingly rare in ophthalmology. METHODS: We report six patients (four underwent penetrating keratoplasty and two underwent cataract surgery) with unusually severe local inflammatory reactions to suture material. RESULTS: Patients developed multiple focal inflammatory infiltrates and corneal oedema related to 10/0 monofilament nylon sutures. Four patients had evidence of either atopy or raised serum IgE and one had an autoimmune disorder (systemic lupus erythematosis). Onset was four to 11 days in five cases and six weeks in one. Allograft reaction occurred in two of the four corneal grafts, and three grafts failed. One of the cataract patients developed significant against-the-rule astigmatism. Microbial cultures of corneal scrapings, donor corneal rims, and suture material were generally negative. Polymorphonuclear leucocytes were noted on Gram stain in four cases, and cytology of corneal scrapings demonstrated eosinophils in one case. CONCLUSIONS: Sterile inflammatory reactions to monofilament suture material are an uncommon complication of ocular surgery which may lead to corneal graft failure, and which is more common in atopic individuals. Systemic steroid therapy may be required.

Electroblotting proteolytic products from native gel for direct N-terminal sequence analysis: an approach for studying protein-protein interaction.

Proteins which are electroblotted from native gels onto polyvinylidene difluoride (PVDF) membranes are suitable for detailed structural analysis. This method, in conjunction with limited proteolysis and N-terminal sequencing, has been used to study the molecular interactions between native protein molecules. The interaction between recombinant interleukin-2 (rIL-2) and its receptor (rIL-2R alpha) was examined as a model system. The working strategy consists of (i) proteolysis of rIL-2R alpha and rIL-2R alpha/rIL-2 complex, (ii) separation of the major proteolytic products by native polyacrylamide gel electrophoresis followed by electroblotting onto PVDF membrane, and (iii) sequence analysis of the blotted protein bands for the identification of peptide regions sensitive to proteolysis. Results have indicated that the exon 3 encoded region in rIL-2R alpha is sensitive to proteolysis regardless whether it is complexed with rIL-2 or not. This suggests that no major conformational changes occur in rIL-2R alpha during interaction with rIL-2. This electroblotting approach is, therefore, useful for studying protein-protein interaction in solution.

Treatment and control of the intestinal fluke, Skrjabinotrema ovis.

The small intestinal fluke, Skrjabinotrema ovis, is common among sheep in the Xinjiang region of western China. Up to 65,000 flukes per sheep were found during September when sheep were grazing summer pastures at high altitudes. Efficacy studies of some common flukicides revealed that oxyclosanide and triclabendazole were not effective in reducing the egg output of S. ovis, whereas closantel and praziquantel reduced egg counts by up to 98%. Praziquantel at 25 and 50 mg kg-1 reduced worm counts by 99%. Albendazole at 15 mg kg-1 reduced egg counts by 83% and worm counts by 91% but at lower dose rates it was ineffective.

Enhancement of cellular immune function during cold adaptation of BALB/c inbred mice.

The cell-mediated immune function of cold-adapted BALB/c inbred mice was studied in experiments of splenic lymphocyte blastogenesis, indicated by tritium-labeled deoxythymidine incorporation and SDS-PAGE autoradiography of synthetic proteins in lymphocytes. Male BALB/c inbred mice were randomly divided into two groups: control (living at 25 degrees C) and cold-exposed (living at 2 degrees C). Results are as follows: in contrast with the control group, there was an obvious fluctuation of cell-mediated immune function in the cold-exposed group at initial cold exposure because of transient stress to cold; then cell-mediated immune function gradually recovered to control level. From Day 15, the cell-mediated immune function of the cold-exposed group was remarkably enhanced. On Day 15, the lymphocyte blastogenesis rate was increased by 20.66% (P less than 0.05), which implies the onset of cold adaptation; on Days 21 and 31, the rates increased by 80.15% (P less than 0.05) and 40.36% (P less than 0.05), respectively. Two to six months later, with continuing cold exposure, the murine lymphocyte blastogenesis rate in the cold-exposed group remained higher than that in the control group. The lymphocyte protein synthesis of the cold-exposed group, indicated by tritium-labeled leucine incorporation, apparently increased on Day 15 and the stimulated rate was 101.47% (P less than 0.05). SDS-PAGE autoradiography of synthetic proteins in lymphocytes demonstrated that after 2 weeks of cold exposure, protein bands were enriched in both quantity and quality. [Effects of four adrenergic drugs on electroacupuncture analgesia].

The role of central nor-epinephrine (NE) in electroacupuncture (EA) analgesia is a controversial question., it is probably due to the complication of adrenergic receptors. The present results show: (1) Clonidine 30 micrograms/2ml/kg ip had no significant effect on the pain threshold, but decreased the analgesic effect of EA. Clonidine 1.5 and 3 micrograms were injected into the lateral cerebral ventricles. After 45 minutes, the analgesic effect of EA was lowered as compared with the saline controls respectively. (2) Yohimbine had no significant effect on the basal pain threshold, but (icv Yoh 50 micrograms) elevated the analgesic effect of EA. (3) 2-adrenoceptor agonist methoxamine decreased the analgesic effect of EA. (4) Another 2-adrenoceptor antagonist prazosin (icv 16 micrograms) enhanced the analgesic effect of EA. These results suggest that an activation of alpha 1- or alpha 2-adrenoceptors would decrease the analgesic effect of EA.

Significance of the 56th tyrosine on cytotoxic activity of tumor necrosis factor.

After using site-specific mutagenesis a conserved amino acid residue, the 56th tyrosine in human tumor necrosis was substituted by glutamine. The expression level of the mutant protein in E. coli did not changed significantly, however, its specific activity of the cytotoxicity on L929 cultural cells was decreased to 1/583 of that of the unmutant tumor necrosis factor. The results show that the 56th tyrosine residue or the region where tyrosine 56th located is important for the cytotoxic activity of tumor necrosis factor.
