Kinetic determination of iodide in pharmaceutical and food samples.

A kinetic method for the determination of iodide based on its inhibitory effect on the Pd(III) catalysed reaction between ethylenediaminetetraacetic acid (EDTA)-Co(III) and the hypophosphite ion is described. The reaction was followed spectrophotometrically by measuring the decrease in the absorbance at 540 nm. Under the optimum experimental conditions of 2.6 x 10(-3) mol dm(-3)Co(III)-EDTA, 0.4 mol dm(-3)H2PO2-, pH 3.2 (adjusted with Britton-Robinson buffer), 0.57 micrograms ml(-1) Pd(III) and 20 +/- 0.2 degrees C, iodide was determined in the range 2-28 ng ml(-1). The method was applied to the determination of iodide in pharmaceutical products, iodinated salts, cow's milk and infants' powdered milk.

A double-blind comparative clinical trial of citalopram vs maprotiline in hospitalized depressed patients.

In a double-blind clinical trial comprising 29 depressed patients citalopram, a highly selective 5-HT re-uptake inhibitor and maprotiline, a specific NA re-uptake inhibitor, were compared. Allowing for the small sample and taking into consideration that both groups consisted of severely ill, hospitalized patients, it is notable that half of them appeared to respond to treatment. Comparison of the clinical efficacy of the two drugs showed no significant difference, but the profiles of the side-effects appeared to be different. The patients treated with citalopram showed increased sweating, drowsiness, restlessness and headache. These side-effects were almost entirely reported by the non-responders. The maprotiline patients had anticholinergic symptoms, such as dryness of mouth and constipation, side-effects which were also reported by the responders. No correlation was found between plasma steady-state levels of either drug and clinical outcome. The Dexamethasone Suppression Test (DST) appeared to show some predictive value as regards treatment response. There was a tendency towards better overall treatment results in the non-suppressor group. Determination of post-probenecid 5-HIAA, HVA and MHPG concentrations in lumbar-CSF was made in 22 patients. There was a significant negative correlation between HVA and the severity of depression, as well as a significant negative correlation of MHPG with the Newcastle score. The 5-HIAA concentration was found to be correlated with HVA, but not with MHPG. Rather surprisingly significant negative correlation between 5-HIAA and treatment results with maprotiline was found, but no correlation with MHPG. The lumbar-CSF MHPG and HVA values did not appear to have any predictive value as regards treatment response to citalopram or maprotiline. As expected the serotonin (5-HT) concentration in blood and thrombocytes in patients treated with citalopram showed a highly significant reduction after 2 and 4 weeks of treatment.

Locus specificity in the mutability of L5178Y mouse lymphoma cells: the role of multilocus lesions.

Mouse L5178Y strain LY-S and its parental strain LY-R differ in their comparative sensitivities to the cytotoxic effects of various mutagenic agents--i.e., strain LY-S has been found to be more sensitive, less sensitive, or similarly sensitive to individual agents in comparison to strain LY-R. Nevertheless, strain LY-S has been found to be uniformly less mutable than strain LY-R at the hypoxanthine (guanine) phosphoribosyltransferase (Hprt) locus following treatment with x-radiation, UV radiation, or alkylating agents. In the present work we have isolated subclones of strains LY-R and LY-S that are heterozygous at the thymidine kinase (Tk) locus (chromosome 11). We have found that a heterozygous LY-S Tk+/Tk- strain shows as high or higher mutability at the Tk locus than do heterozygous LY-R strains following treatment with x-radiation, UV radiation, or ethyl methanesulfonate. Mutability of all heterozygous strains at the Tk locus is much higher than at the Hprt locus following treatment with these mutagenic agents, with the exception of one heterozygous LY-R strain that possesses only one chromosome 11 and that is poorly mutable at the Tk locus by x-radiation. On the basis of these results, we have suggested that because of a repair deficiency, multilocus lesions are formed in the DNA of LY-S strains following treatment with radiation or alkylating agents; multilocus lesions lead to poor recovery of viable mutants when the target locus is closely linked to essential genes and situated on a hemizygous chromosomal region (e.g., the Hprt locus on the X chromosome or the Tk locus in strains monosomic for chromosome 11); and x-radiation is a relatively poor mutagen at loci situated on hemizygous chromosomal regions, in repair-efficient and repair-deficient cells, because of its tendency to form multilocus lesions.

Ionic calcium levels during pregnancy, at delivery and in the first hours of life.

Free calcium ion concentration (mmol/l) and pH were determined in whole blood using a semiautomatic electrode system (ICA-1 Radiometer, Copenhagen, Denmark) in 37 normal women, 90 pregnant women (30 from each trimester of gestation), 28 mothers at delivery and their respective newborns. The blood samples from normal controls, pregnant women, umbilical cord and 40-50-hour-old infants were collected anaerobically in vacuum tubes. Duplicate samples drawn from newborns shortly after birth by heel puncture were collected in special heparinized capillary tubes. We observed that Ca2+ concentrations in the second (1.20 +/- 0.04) and third (1.20 +/- 0.05) trimesters of pregnancy, and at delivery (1.18 +/- 0.05), were lower than in the control group (1.23 +/- 0.04). The [Ca2+] in samples from the umbilical vein (1.44 +/- 0.11) and artery (1.45 +/- 0.08) and from newborns 2-5 min after birth (1.34 +/- 0.12) was greater than in control samples. The [Ca2+] in newborns 40-50 hours after birth was lower (1.16 +/- 0.14) than in the control group.

Bilateral and symmetrical osteomyelitis due to gram-negative bacilli. A report on two cases.

Gram-negative bacillary osteomyelitis secondary to sepsis is a rare condition. It most frequently attacks the vertebrae, occasionally in the long bones, and can on occasion be multiple. We present 2 patients with severe sepsis of urinary origin who, after a latent period, developed bilateral and symmetrical osteomyelitis due to the same microorganism that caused the sepsis. We discuss this peculiar presentation, its possible pathogenetic mechanism and the need to consider osteomyelitis in the presence of bilateral and symmetrical arthritis after an episode of severe sepsis.

Necrotizing fasciitis caused by Salmonella enteritidis.

A 76-yr-old man developed necrotizing fasciitis due to Salmonella enteritidis 1 month after an episode of gastroenteritis due to the same microorganism. The fact that S. enteritidis was the only organism isolated despite adequate anaerobic cultures confirm the ability of salmonellae to produce severe monomicrobial soft tissue infections.

Double outlet right ventricle: experimental morphogenesis in the chick embryo heart.

Double outlet right ventricle (DORV) was produced experimentally in the chick embryo by preventing the incorporation of the posteromedial conus into the left ventricle. This was accomplished by placing a ligature around the caudal end of the conus at a stage in which it is exclusively continuous with the right ventricle. The embryos were divided into two groups according to the stage of development of the conal ridge. In one group, the conal ridges had not appeared (stages 17-18); in the other they had begun to develop (stage 22). Both groups were divided into two subgroups; in one the ligature was left in place up to the mature heart stage and in the other it remained for only 24 hours. In all the embryos where the ligature remained until the mature heart stage a DORV was obtained, while in the embryos where the ligature remained in place for only 24 hours a DORV was obtained in only two-thirds of the cases. Two different types of DORV were produced: 1) with anterior and posterior infundibula in which the subpulmonary infundibulum was anterior to the subaortic infundibulum and there was a subaortic infracristal ventricular septal defect (VSD); 2) with side-by-side infundibula in which the subpulmonary infundibulum was on the left side and the subaortic infundibulum on the right side; the VSD was far from both infundibula. Our experiments show that DORV is an essentially infundibular malformation. In DORV there are two types of infundibular relationships: a) with anterior and posterior infundibula, and b) with side-by-side infundibula. The crista supraventricularis in the normal heart and the infundibular septum in DORV are two different anatomic and embryologic structures.
