[Cytoarchitecture and histochemical fluorescence studies of the cingulate cortex in the rat].

The cingular cortex of the rat was examined by means of Golgi's rapid impregnation technique, the glyoxylic acid technique for the detection of biogenic amines and a ninhydrin reaction in the octanoic milieu for the detection of GABA. Six interneuron types are described, differing with respects to their dendrite configurations and axon morphology. The noradrenaline innervation shows considerable differences as compared to the dopamine fibre system. GABA-containing neurons of various shapes and sizes are concentrated in the laminae II and III. The findings are discussed in connection with the conception according to the which the Gyrus cinguli as part of the limbic system might play a role in the genesis of psychopathological processes.

[Transmitter histochemistry of the olfactory tubercle in the rat].

The tuberculum olfactorium of the adult rat was investigated by means of neurohistological, fluorescence histochemical and enzyme histochemical methods. Using the rapid Golgi impregnation technique, seven neuron types could be demonstrated which can clearly be differentiated according to morphological criteria. Using the glyoxylic acid techniques, strongly fluorescent dopamine fibers are, in different densities, visible within the various laminae. Kainic acid application into the ventral tegmental area is followed by a strikingly reduced, no more structurally associated dopamine fiber fluorescence. After DFP intoxication, only a small proportion of the neuron population of the tuberculum olfactorium does exhibit an AChE activity; the AChE containing neurons are different as to their size and shape. The findings presented here are discussed with respect to the attributability of histochemical findings to cytoarchitectural features of this brain area.

[7 types of neurons in the substantia nigra of the rat. Golgi rapid-impregnation study].

The substantia nigra of the adult rat was investigated by means of the Golgi rapid impregnation technique. Seven neuron types could be distinguished which differ with respect to morphological features (size and shape of pericaryon and dendritic pattern), behaviour of axons and prevalent localization within the nuclear area: 1. Polygonal neurons with somatic spines 2. Polygonal neurons without somatic spines 3. Fusiform neurons 4. Trigonal neurons 5. Small spherical neurons 6. Small spindle-shaped neurons 7. Neuroglioform neurons. The neuron types 1-4 are considered to represent nigral projection neurons; the types 5-7 might be interneurons. The findings presented here are discussed in view of recent physiological and biochemical as well as behavioral-pharmacological results.

[Further studies on the cytoarchitecture of the nucleus accumbens septi of the rat].

The aim of the work presented here was to make some contributions on the elucidation of the cytoarchitecture of the nucleus accumbens septi of adult rat. After application of the rapid Golgi impregnation technique, five neuron types covered with spines and three aspiny neurons types could be distinguished. After DEP-pretreatment, two neuron types proved to be AChE-reactive. The findings elaborated by means of different methods are tempted to be correlated with on another. The cytoarchitectonic features described here make the nucleus accumbens septi of rat appear to represent a structure much alike the caudate-putman complex.

[Changes in Golgi rapid impregnation in the caudate-putamen complex of the rat after pharmacologic pretreatment].

The neostriatum of adult rats was investigated by means of the Golgi rapid impregnation procedure following surgical (unilateral stereotaxic MFB lesions or complete deafferentation) and pharmacological (i.p. application of pargyline, amphetamine, reserpine, haloperidol in submaximal doses) pretreatments, respectively. The neurons of this brain area--as a whole, ten types can be differentiated according to morphological criteria--do exhibit a characteristic impregnation behavior being in proportion to the mode of pretreatment in each case. The impregnation behavior of the neurons is postulated to depend on the polarization state of their membranes which may be explained in pharmacological and physiological terms.

[GABA containing neurons in the nucleus raphes dorsalis of the rat (author's transl)].

The Nucleus raphes dorsalis (NRD) of adult rat was investigated by means of fluorescence histochemical and neurohistological methods. After application of a modified ninhydrine reaction GABA-containing fusiform neurons and small nerve cells exhibiting fluorescence around their somata were found. Using the EINARSON-reaction for demonstrating the ribonucleic proteins in the NRD 3 neuron types could be demonstrated.

[Acetylcholinesterase containing neurons in the rat brain after DFP-pretreatment (author's transl)].

The AChE activity was demonstrated in some brain regions of adult rats after intoxication with DFP ( (di-isopropyl-fluorophosphate). Within these nuclear areas (Caudate-Putamen-Complex, Globus pallidus, Nucleus accumbens septi, Substantia nigra, Locus coeruleus) various proportions of the neuron population could be shown to be AChE positive. There is a characteristic morphology of the AChE containing neurons. Comparing them with staining and silver impregnation, cytological attributability appears feasible.

[GABA fluophore formation due to ninhydrin reaction in the octanolic milieu. Fluorescence histochemical investigation. 3. Information (author's transl)].

Various steps of the ninhydrin-reaction in the octanolic milieu (Wolman 1971; Pfister and Görne 1978) were tested on the cerebellum of adult rats. The results of the fluorescence histochemical procedure was found to be in comparison with the results ascertained to spectrofluorometric method. The findings were further indicative for a GABA-specificity of the reaction.

[Fluorescence histochemical investigations on the carbohydrate metabolism of the rat brain (author's transl)].

The Regio frontalis posterior of the motor cortex of adult rats was investigated by means of the fluorescence-PAS-reaction. Using various Schiff-type-reagents - substituted with acriflavine, rhodamine B, rhodamine 6G, safranine T - a fluorescent reaction product has been found in different amounts in the pyramidal neurons and in other cellular elements. After application of acriflavine different degrees of PAS-reactivity could be demonstrated in various cortices. Dexamethasone affected an increase of the fluorescent reaction product in individual pyramidal neurons. Ideas on the possible reaction mechanism after application of various Schiff-type-reagents have been proposed.

[4 spine-less neuron types in the neostriatum of the rat].

The caudate-putamen-complex of adult rat was investigated by means of the rapid Golgi impregnation technique. Untreated rats and rats with unilateral lesions of the MFB were analysed. Four aspiny neuron types and types extremely sparse in spines, resp., could be demonstrated: Aspiny giant neurons, aspiny spindle-shaped neurons, aspiny spidery neurons and neuroglioform neurons. The spine-less neuron types of the caudate putamen which can be differentiated according to morphological criteria are tempted to be classified in a functional manner. Especially based on the axon morphology, the giant neurons are considered to be projective neurons; the other cell types are, in all probability, neostriatal interneurons. The type 2 neurons (spindle-shaped neurons) which are regarded to be absolutely autonomous cell types do represent the vast majority of the population of the aspiny neurons of the caudate-putamen-complex. Comparing the findings laid down here with those of AChE-histochemistry after DFP treatment may permit conclusions with respect to the functional role of aspiny neurons within the neostriatum of the rat.

[Demonstration of spine-bearing dwarf neurons in the caudatus-putamen complex of the rat following pargyline application].

The neostriatum of adult rats pretreated wit pargyline was investigated by means of the rapid Golgi procedure. An hitherto unknown spine-bearing neuron typ with a soma size ranging from 6 to 8 microm could be demonstrated. Owing to its morphological features, this neuron type is regarded to be a striatal interneuron. The mode of transmission and the functional role of these neurons within the caudatus-putamen-complex is still to be examined.

[The cytoarchitecture of the rat globus pallidus].

The globus pallidus of the adult rat was investigated by means of the Golgi-rapid-impregnation technique. The cellular population of the globus pallidus is composed of 4 morphologically different neuron types: 1. Large-sized, spindle-shaped neurons; 2. Large-sized, polygonal neurons; 3. Small spindle-shaped neurons; 4. Small spine-bearing neurons. The large spindle-shaped neurons and the large polygonal neurons are considered to be efferent pallidal neurons; the small spindle-shaped neurons and the small spiny neurons are suggested to be pallidal interneurons. It is possible to adapt the neuron classification presented here to recent imaginations concerning the role of the globus pallidus within the extrapyramidal system.

[Spiny neurons in the rat caudate-putamen complex].

The neostriatum of adult normal rats and of rats after surgical lesion of MFB was investigated by means of the rapid Golgi impregnation procedure. Evidence was given for the spiny neuron population to be heterogenous. It was possible to demonstrate four neuron types clearly distinguishable with respect to size and shape of the soma, dendritic pattern, and axon morphology. The morphological heterogeneity of the spiny neurons of the caudatus-putamen-complex of the rat presented here is discussed in functional terms (interneuron and efferent neuron nature, resp.).

[Investigation on the cytoarchitecture of the nucleus accumbens septi of rat (author's transl)].

The nucleus accumbens septi of the normal adult rat was investigated by means of the rapid Golgi impregnation technique according to Valverde (1970). 5 neuron types could be differentiated: type 1: spiny neurons; type 2: few spiny neurons; type 3: aspiny giant neurons; type 4: spiny spindle shaped neurons; type 5: aspiny spidery neurons. The types 1, 4 and 5, resp., are considered, with respect to morphological criteria, to represent interneurons, whereas the types 2 and 3 are considered to be accumbens output neurons. The cytoarchitecture of the nc. accumbens septi thus displays striking similarities with the caudate-putamen-complex. An hypothetical correlation of these neurons in a functional -- biochemical sense as well as their position in the nc. accumbens which is suggested to be a transition area between limbic and extrapyramidal motor systems, is discussed.

[Cytoarchitecture of the olfactory bulb of the rat].

The olfactory tubercle of rat was investigated by means of the rapid Golgi impregnation procedure. Based on morphological criteria, seven neuron types can be distinguished in this brain region of adult rat. Two types (pyramidal neurons and large polygonal neurons) are considered to be efferent in nature whereas the other types (spine-bearing fusiform neurons; aspiny fusiform neurons; granule cells; dwarf neurons, and neuroglioform neurons) do, in all probability, represent tuberculum interneurons. It is obvious from impregnation studies done in the olfactory tubercle of very young rats (seven days postsnatally) that neurons are still poorly differentiated at this time.

[Topography and cytoarchitecture of the raphe nuclei in the rat].

Within the raphe complex of adult rat, ribonucleoproteids have been demonstrated by means of the Gallocyanin-Chromalaun-reaction according to EINARSON. The following subnuclei were characterized in a topographical and cytological view: nucleus raphe dorsalis, nucleus centralis superior, nucleus raphe pontis, nucleus raphe magnus, nucleus raphe obscurus, nucleus raphe pallidus. In all raphe nuclei 3 neuron types could be differentiated which differ both with respect to size and shape of their soma and nuclear morphology and intensity and distribution of the cellular ribonucleoproteids: 1. Large polygonal neurons, 2. medium-sized fusiform neurons, 3. small pyriform neurons. The results may serve as a basis for performing further investigations of these brain regions, employing morphological and pharmacological methods.

Electrical response of beef-heart submitochondrial particles bound to phospholipid-impregnated millipore filters during ATP hydrolysis.

1. Beef heart submitochondrial particles bound to asolectin impregnated Millipore filter, according to the method described earlier (Drachev et al. (1974) Nature 249, 321--324), are able to generate, upon addition of ATP, an electrical potential which can be directly measured. 2. The transmembrane electrical potential generated by ATP hydrolysis reaches values up to 80 mV. The half-time required to attain the plateau of potential is paradoxically long (5 to 10 min at room temperature) and is temperature-dependent. Among different phospholipid species which have been used to impregnate the Millipore filter, phosphatidylethanolamine was found to be the most effective for generation of electrical potential. 3. The potential generated by ATP hydrolysis is inhibited by inhibitors of mitochondrial ATPase, by the uncoupler FCCP and by reagents collapsing the membrane potential. 4. Addition of inhibitors of mitochondrial ATPase, when the plateau of potential is attained, results in a decay of potential. This decay of potential is as slow as the generation of potential induced by ATP hydrolysis. 5. The initial rise in electrical potential is proportional to the ATPase activity.
